Hi, Regarding altering the number of functionally silenced synapses, for potassium treatment - after 4 hrs do you replace only half of the neurobasal media or all of it. At what day specifically do you perform these treatments? Thank you so much, Anamaria

Anamaria: After 4 hr treatment, we typically completely replace the culture medium with a defined HEPES buffered saline for the FM1-43 staining or for electrophysiology. We usually use DIV 10-14 cells because they have reached near synaptic maturity. Other details can be found in our primary publications. Steve Mennerick
