May I transfect SCG neurons via either electroporation or Lipofectamine protocols? Can you kindly siggest me a possible/suitable protocol to be ised to transfect SCG neurons? Thank you very much!

SCG neurons are quite difficult to transfect. I have tried Lipofectamine 2000 and a few other transfection reagents with no luck. I have never done electroporation and I don't think you can once the neurons or any type of cells are grown as a monolayer culture on a plate. You may be able to use a gene gun to transfect your neurons but once again, I have no experience in using it. One method that I have tried is lentiviral transduction. It works beautifully and I get nearly 100% efficiency using lentivirus method.

Am having problems transducing rat sympathetic neurons, my shRNA is in TRIPZ plasmid am am making the virus in 293T cell with titer of ~1million. I infect the neurons with MOI 10, 20 and 40 but am only getting about 1/2 neurons infected and a few glial cells. What could i be doing wrong? Thanks
