Proteomics has emerged as the next great scientific challenge in the post-genome era. But even the most basic form of proteomics, proteome profiling, i.e., identifying all of the proteins expressed in a given sample, has proven to be a demanding task. The proteome presents unique analytical challenges, including significant molecular diversity, an extremely wide concentration range, and a tendency to adsorb to solid surfaces. Microfluidics has been touted as being a useful tool for developing new methods to solve complex analytical challenges, and, as such, seems a natural fit for application to proteome profiling. In this review, we summarize the recent progress in the field of microfluidics in four key areas related to this application: chemical processing, sample preconcentration and cleanup, chemical separations, and interfaces with mass spectrometry. We identify the bright spots and challenges for the marriage of microfluidics and proteomics, and speculate on the outlook for progress.

Mais m&#xEA;me la forme la plus fondamentale de la prot&#xE9;omique, profilage prot&#xE9;ome, c&#x26;#39;est &#xE0; dire, d&#x26;#39;identifier toutes les prot&#xE9;ines exprim&#xE9;es dans un &#xE9;chantillon donn&#xE9;, s&#x26;#39;est av&#xE9;r&#xE9; &#xEA;tre une t&#xE2;che exigeante. Le prot&#xE9;ome pr&#xE9;sente des d&#xE9;fis uniques d&#x26;#39;analyse, y compris la diversit&#xE9; mol&#xE9;culaire importante, une plage de concentrations extr&#xEA;mement large, et une tendance &#xE0; adsorber sur des surfaces solides. Proteomics hat als die n&auml;chste gro&szlig;e wissenschaftliche Herausforderung in der POSTGENOM-&Auml;ra entstanden. Aber auch die Grundform der Proteomik, Proteom, Profilerstellung, d. h. Ermittlung aller der Proteine, die in einer bestimmten Probe erweist sich eine anspruchsvolle Aufgabe. Das Proteome pr&auml;sentiert einzigartige analytische Herausforderungen, darunter auch bedeutende Molekulare Vielfalt, einer extrem breiten Konzentrationsbereich und eine Tendenz, auf festen Oberfl&auml;chen adsorbieren. Mikrofluidik wurde angepriesen als ein n&uuml;tzliches Werkzeug f&uuml;r die Entwicklung neuer Methoden zur L&ouml;sung komplexer analytischer Herausforderungen und, als solcher, scheint eine nat&uuml;rliche Erg&auml;nzung f&uuml;r Proteome Profilerstellung der Anwendung. In diesem Bericht, fassen wir die j&uuml;ngste Fortschritte auf dem Gebiet der Mikrofluidik in vier zentralen Bereichen mit Bezug zu dieser Anwendung: chemische Verfahrenstechnik, Beispiel Preconcentration und Bereinigung, chemischen Trennungen und Schnittstellen mit Massenspektrometrie. Proteomica &egrave; emerso come la prossima grande sfida scientifica in epoca post-genome. Ma anche le pi&ugrave; elementare forma di proteomics, proteome profilatura, vale a dire, identificando tutte le proteine espresse in un dato campione, ha dimostrato di essere un compito impegnativo. Il proteoma presenta sfide uniche analitiche, tra cui significativa diversit&agrave; molecolare, una gamma estremamente ampia di concentrazione e la tendenza ad assorbire a superfici solide. Microfluidica &egrave; stato propagandato come un utile strumento per lo sviluppo di nuovi metodi per risolvere complessi problemi analitici e, come tale, sembra una naturale adatta per l&#39;applicazione all&#39;analisi del proteoma. We report a new method for fabricating nanospray ionization tips for MS, formed from glass substrates and the inert polymer, parylene-C. Using a single photolithography step, the emitters are formed contiguously with microchannels, such that no dead volumes are observed. In addition, because the devices are very thin (approximately 0.3 mm) and the tips are formed at rectangular corners, the Taylor cone volumes are small, which makes the method attractive for future integration with microfluidic separations. Device performance was demonstrated by evaluating diverse analytes, ranging from synthetic polymers, to peptides, to nucleic acids. For all analytes, performance was similar to that of conventional emitters (pulled-glass capillaries and the Agilent HPLC Chip) with the advantage of rapid, batch fabrication of identical devices.

Digital microfluidics has become a popular tool for biochemical and biomedical applications. However, its current format is restricted to actuation of droplets on a single plane. Here, we introduce a new method for fluid handling on flexible devices, which we have termed all-terrain droplet actuation (ATDA). We show that ATDA can be used to manipulate droplets across a wide range of geometries, including inclined, declined, vertical, twisted, and upside-down architectures. These new geometries enable flexible, straightforward integration of distinct physicochemical environments on monolithic devices. To illustrate this capacity, we developed temperature- and oxygen-sensitive colorimetric sensors, as well as an automated method for selective enrichment of DNA from a heterogeneous mixture. We anticipate that ATDA will be a useful new tool in the growing trend toward laboratory miniaturization.

Hier stellen wir eine neue Methode f&uuml;r Fluid handling auf flexible Ger&auml;te, die wir haben All-Terrain-Tr&ouml;pfchen Bet&auml;tigung (ATDA) bezeichnet. Wir zeigen, dass ATDA verwendet werden kann, um Tr&ouml;pfchen in einem breiten Spektrum von Geometrien, einschlie&szlig;lich geneigte, abgelehnte, vertikale, verdrehte und Kopf-Architekturen zu manipulieren. Diese neuen Geometrien erm&ouml;glichen flexible, unkomplizierte Integration von unterschiedlichen physikalisch-chemischen Umgebungen auf monolithischen Ger&auml;ten. Um diese F&auml;higkeit zu veranschaulichen, haben wir die Temperatur und sauerstoffempfindliche farbmetrisch Sensoren sowie eine automatisierte Methode zur selektiven Anreicherung von DNA aus einer heterogenen Mischung entwickelt. Aqu&#xED;, presentamos un nuevo m&#xE9;todo para la manipulaci&#xF3;n de fluidos en los dispositivos flexibles, que hemos denominado todo terreno gota de actuaci&#xF3;n (ADTA). Se demuestra que ADTA puede ser usado para manipular las gotas a trav&#xE9;s de una amplia gama de geometr&#xED;as, como inclinado, declinado, las arquitecturas verticales y retorcidas, y al rev&#xE9;s. Todo terreno gotas de Actuaci&#xF3;n

Digital microfluidics, a technique for manipulation of droplets, is becoming increasingly important for the development of miniaturized platforms for laboratory processes. Despite the enthusiasm, droplet motion is frequently hindered by the desorption of proteins or other analytes to surfaces. Current approaches to minimize this unwanted surface fouling involve the addition of extra species to the droplet or its surroundings, which might be problematic depending on the droplet content. Here, a new strategy is introduced to move droplets containing cells and other analytes on solid substrates, without extra moieties; in particular, droplets with bovine serum albumin could be moved at a concentration 2000 times higher than previously reported (without additives). This capability is achieved by using a soot-based superamphiphobic surface combined with a new device geometry, which favors droplet rolling. Contrasting with electrowetting, wetting forces are not required for droplet motion.
