Detection of PrP(CWD) in Postmortem Rectal Lymphoid Tissues in Rocky Mountain Elk (Cervus Elaphus Nelsoni) Infected with Chronic Wasting Disease

Pubmed ID: 17121082 Preclinical diagnostic tests for transmissible spongiform encephalopathies have been described for mule deer (Odocoileus hemionus), using biopsy tissues of palatine tonsil, and for sheep, using lymphoid tissues from palatine tonsil, third eyelid, and rectal mucosa. The utility of examining the rectal mucosal lymphoid tissues to detect chronic wasting disease (CWD) was investigated in Rocky Mountain elk (Cervus elaphus nelsoni), a species for which there is not a live-animal diagnostic test. Postmortem rectal mucosal sections were examined from 308 elk from two privately owned herds that were depopulated. The results of the postmortem rectal mucosal sections were compared to immunohistochemical staining of the brainstem, retropharyngeal lymph nodes, and palatine tonsil. Seven elk were found positive using the brainstem (dorsal motor nucleus of the vagus nerve), retropharyngeal lymph nodes, and palatine tonsil. Six of these elk were also found positive using postmortem rectal mucosal sections. The remaining 301 elk in which CWD-associated abnormal isoform of the prion protein (PrP(CWD)) was not detected in the brainstem and cranial lymphoid tissues were also found to be free of PrP(CWD) when postmortem rectal mucosal sections were examined. The use of rectal mucosal lymphoid tissues may be suitable for a live-animal diagnostic test as part of an integrated management strategy to limit CWD in elk.

Oral Rabies Vaccine (ORV) Bait Uptake by Captive Striped Skunks

Pubmed ID: 17347398 Aerial delivery of oral rabies vaccine (ORV) baits has proven effective in large-scale efforts to immunize wildlife against rabies, and in North America this strategy currently is being used to immunize foxes (Urocyon cinereoargenteus and Vulpes vulpes), raccoons (Procyon lotor), and coyotes (Canis latrans). Skunks are also a major reservoir and vector of rabies, but at present oral vaccines for use in skunks are not licensed. Furthermore, given differences in morphology (smaller jaws) and behavior (food handling and consumption), it is unknown if baits currently used in ORV campaigns would be effective for skunks. Because oral vaccine delivery is contingent upon puncture of the vaccine container (VC), baits need to be sufficiently attractive to elicit selection and consumption. Manipulation of the bait to facilitate vaccine ingestion by the target species is a critical element for an effective ORV bait. The objectives of this study were to assess manipulation and consumption of current ORV baits by striped skunks (Mephitis mephitis). We conducted four independent trials with penned animals and various baits to assess bait selection frequency, VC puncture frequency, and consumption. Video recorded trials were used to assess attractiveness of baits and consumption behavior of skunks. Bait characteristics, such as texture, size, and flavor influenced selection and consumption. Fish and chicken flavors were preferred and vaccine containers within selected baits were likely to be punctured. Vaccine ingestion seemed more likely if VCs were directly coated with the bait matrix. To make baits attractive to skunks and to ensure puncture of the VC, modifications to current baits should consider a smaller size, a meat-flavored matrix, a slightly pressurized VC, and a direct coating of matrix on the VC.

Surveillance of Coyotes to Detect Bovine Tuberculosis, Michigan

Pubmed ID: 19046508 Bovine tuberculosis (TB) is endemic in white-tailed deer (Odocoileus virginianus) in the northeastern portion of Michigan's Lower Peninsula. Bovine TB in deer and cattle has created immense financial consequences for the livestock industry and hunting public. Surveillance identified coyotes (Canis latrans) as potential bio-accumulators of Mycobacterium bovis, a finding that generated interest in their potential to serve as sentinels for monitoring disease risk. We sampled 175 coyotes in the bovine TB-endemic area. Fifty-eight tested positive, and infection prevalence by county ranged from 19% to 52% (statistical mean 33%, SE 0.07). Digestione Enzimatica Dei Prioni Di Malattia Sprecare Cronico Associato Al Suolo

Pubmed ID: 20450190 Malattia del deperimento cronico (CWD) e della scrapie ovini possono essere trasmessi attraverso itinerari ambientali indiretti, ed è noto che il terreno può servire come un serbatoio di infettività prionica. Data la forte interazione tra la proteina prionica (PrP) e del suolo, abbiamo ipotizzato che associazione al suolo migliora la resistenza del prione per digestione enzimatica, facilitando la longevità da prioni nell'ambiente e fornire protezione da degradazione host. Abbiamo caratterizzato le prestazioni di un enzima subtilisina commercialmente disponibile, Prionzyme, a degradare CWD terreno associate e non associate e HY TME PrP in funzione del tempo di pH, temperatura e trattamento. L'enzima subtilisina degradata efficacemente PrP adsorbito in una vasta gamma di terreni e minerali del suolo di sotto dei limiti di rilevazione. Perdita di segnale si verificata rapidamente a pH elevato (12,5) ed entro 7 giorni sotto rappresentante le condizioni dell'ambiente naturale (pH 7.4, 22 gradi C). Abbiamo non osservato alcuna differenza apparente in efficacia enzima tra associati e non associati CWD PrP. I nostri risultati mostrano che nonostante prioni adsorbiti conservano relativa resistenza alla digestione enzimatica confrontato con altre proteine omogeneato di cervello, possono essere efficacemente degradati quando associato al suolo. Resource Selection by Elk in an Agro-forested Landscape of Northwestern Nebraska

Pubmed ID: 20872141 In recent years, elk have begun recolonizing areas east of the Rocky Mountains that are largely agro-forested ecosystems composed of privately owned land where management of elk is an increasing concern due to crop and forage depredation and interspecific disease transmission. We used a Geographic Information System, elk use locations (n = 5013), random locations (n = 25,065), discrete-choice models, and information-theoretic methods to test hypotheses about elk resource selection in an agro-forested landscape located in the Pine Ridge region of northwestern Nebraska, USA. Our objectives were to determine landscape characteristics selected by female elk and identify publicly owned land within the Pine Ridge for potential redistribution of elk. We found distance to edge of cover influenced selection of resources by female elk most and that in areas with light hunting pressure, such as ours, this selection was not driven by an avoidance of roads. Female elk selected resources positioned near ponderosa pine cover types during all seasons, exhibited a slight avoidance of roads during spring and fall, selected areas with increased slope during winter and spring, and selected north- and east-facing aspects over flat areas and areas with south-facing slopes during winter months. We used our models to identified a potential elk redistribution area that had a higher proportion of landcover with characteristics selected by elk in our study area than the current herd areas and more landcover that was publicly owned. With appropriate management plans, we believe elk within the Potential Elk Redistribution Area would predominantly occupy publicly owned land, which would help minimize crop and forage damage on privately owned lands.

Impact of Baiting on Feral Swine Behavior in the Presence of Culling Activities

Pubmed ID: 22305877 We studied the effects of baiting on feral swine (Sus scrofa) movements and corresponding likelihood of disease spread under real and simulated culling pressure. Our objectives were to determine the proportion of feral swine that used the bait station site, and if baiting of feral swine altered areas of utilization, distances from location centroids to treatment location (control or bait station), and movement rates by survivors during culling activities. We hypothesized that the bait station would increase the sedentary nature of feral swine, thus reducing the potential for dispersal and hence disease dispersal. Our experiment was conducted between February and May 2011 on the Rob and Bessie Welder Wildlife Foundation (WWF) in San Patricio County, Texas. We trapped 83 feral swine and placed GPS collars on 21 animals. We established and maintained a centralized bait station on one side of the WWF from 13 March to 27 April. We conducted population-wide culling activities, including trapping, controlled shooting, drive shooting, and aerial gunning, from 3 to 27 April and removed 143 feral swine (4.6feral swine/km(2)). Areas of utilization did not differ between treatments (control or bait station). However, we found location centroids of bait station site feral swine to be closer to the treatment location than those of control site animals and daily movement rates of bait station site feral swine to be 39% greater than movement rates of control site animals. Based on our observation that only 62% of feral swine trapped in proximity to the bait station used it, we cannot recommend baiting as an alternative to fences for containing animals during culling activities. However, there is value in using bait stations to describe patterns of feral swine movements, facilitate observation, and improve efficacy when conducting removals.

Estimating Prion Adsorption Capacity of Soil by BioAssay of Subtracted Infectivity from Complex Solutions (BASICS)

Pubmed ID: 23484043 Prions, the infectious agent of scrapie, chronic wasting disease and other transmissible spongiform encephalopathies, are misfolded proteins that are highly stable and resistant to degradation. Prions are known to associate with clay and other soil components, enhancing their persistence and surprisingly, transmissibility. Currently, few detection and quantification methods exist for prions in soil, hindering an understanding of prion persistence and infectivity in the environment. Variability in apparent infectious titers of prions when bound to soil has complicated attempts to quantify the binding capacity of soil for prion infectivity. Here, we quantify the prion adsorption capacity of whole, sandy loam soil (SLS) typically found in CWD endemic areas in Colorado; and purified montmorillonite clay (Mte), previously shown to bind prions, by BioAssay of Subtracted Infectivity in Complex Solutions (BASICS). We incubated prion positive 10% brain homogenate from terminally sick mice infected with the Rocky Mountain Lab strain of mouse-adapted prions (RML) with 10% SLS or Mte. After 24 hours samples were centrifuged five minutes at 200 × g and soil-free supernatant was intracerebrally inoculated into prion susceptible indicator mice. We used the number of days post inoculation to clinical disease to calculate the infectious titer remaining in the supernatant, which we subtracted from the starting titer to determine the infectious prion binding capacity of SLS and Mte. BASICS indicated SLS bound and removed ≥ 95% of infectivity. Mte bound and removed lethal doses (99.98%) of prions from inocula, effectively preventing disease in the mice. Our data reveal significant prion-binding capacity of soil and the utility of BASICS to estimate prion loads and investigate persistence and decomposition in the environment. Additionally, since Mte successfully rescued the mice from prion disease, Mte might be used for remediation and decontamination protocols.

Experimental Infection of White-tailed Deer (Odocoileus Virginianus) with Northern European Bluetongue Virus Serotype 8

Pubmed ID: 23876932 Bluetongue (BT) is an insect-transmitted, economically important disease of domestic and wild ruminants. Although only five of the 26 reported bluetongue virus (BTV) serotypes are considered endemic to the USA, 10 exotic serotypes have been isolated primarily in the southeastern region of the country since 1999. For an exotic BTV serotype to become endemic there must be susceptible animal species and competent vectors. In the USA, sheep and white-tailed deer (WTD) are the primary sentinel livestock and wildlife species, respectively. In 2006, BTV-8 was introduced into Northern Europe and subsequently overwintered, causing unprecedented livestock disease and mortality during the 2006-2007 vector seasons. To assess the risk of the European strain of BTV-8 to North American WTD, and understand the role they could play after a similar introduction, eight bluetongue-seronegative WTD were inoculated with BTV-8. Body temperatures and clinical signs were recorded daily. Blood samples were analyzed for BTV RNA with quantitative real time reverse transcriptase polymerase chain reaction (qRT-PCR), serum analyzed for BTV antibodies by cELISA, and tissues taken for histopathology and qRT-PCR. All eight deer became infected and developed moderate to severe clinical disease from days 8 to 15. Peak viremia was from day 7 to 10 with detectable titers through the end of the study (28 days) in most deer. Serum antibody was detected by day 6, peaked by day 10 and continued through day 28. We conclude that North American WTD are highly susceptible to BTV-8 and would act as clinical disease sentinels and amplifying hosts during an outbreak.
