接下来，通过 40 微米细胞过滤器过滤浆液，并向单细胞悬液中加入 25 毫升解剖培养基。在细胞培养箱中孵育 4 小时，以允许成纤维细胞粘附。在孵育的最后一小时开始时，在每只小鼠的两个 35 毫米培养皿中，在低温 IMDM 培养基中涂上 500 μL 1% 基底膜基质，在室温下包被 1 小时。
在预铺板结束时，收集上清液并离心漂浮细胞。重悬成肌细胞和生长培养基，计数细胞后，将体积调整至适当的细胞浓度以进行铺板。然后用 DPBS 洗涤基底膜基质包被的板，并立即将细胞接种在细胞培养箱中孵育。
三天后，根据制造商的说明用适当的转染试剂处理细胞，并将细胞与目标脂质复合物在 37 摄氏度、5% 二氧化碳中孵育 5 小时。然后，在分化培养基中进行两次洗涤，并将细胞放回培养箱中过夜。第二天，用 200 μL 50% 基底膜基质和冰冷分化培养基替换每个培养皿中的培养基。
在细胞培养箱中再放置 30 分钟后，用聚集蛋白和新鲜分化培养基补充培养物，每天监测细胞的分化和活力，直到它们完全成熟。对于免疫染色，在感兴趣的实验时间点，在 DPBS 中洗涤细胞，然后在室温下固定在 4% 多聚甲醛中。10 分钟后，在 DPBS 中再洗涤细胞 2 次，然后在室温下用 0.5% Triton X 100 透化 5 分钟。
再洗涤两次 DPBS 后，在室温下用封闭溶液封闭非特异性结合 30 分钟。然后，将细胞与在封闭溶液中稀释的目标一抗在 4 摄氏度下孵育过夜。第二天早上，在室温下用 DPBS 洗涤 3 次 5 分钟冲洗细胞，并将培养物与适当的二抗在 0.2 μg/mL DAPI中孵育 1 小时。
如刚才所示，再洗涤 3 次 DPBS 后，将样品封片在 200 微升封固剂中并对细胞进行成像。在增殖第 2 天，成肌细胞应该已经粘附在板上，并且应该显示出典型的梭形形状，广泛的增殖导致自发性肌管形成。肌管迅速伸长并显示多个中心对齐的细胞核，成熟肌纤维的数量随着时间的推移而增加，显示出更高的细胞厚度、条纹和外周细胞核。
