Identification of Circular RNAs using RNA Sequencing Shobana Sekar1,2, Philipp Geiger1,2, Lori Cuyugan1,2, Annalee Boyle1,2, Geidy Serrano2,3, Thomas G. Beach2,3, Winnie S. Liang1,2 1Translational Genomic Research Institute, 2 Circular RNAs (circRNAs) are non-coding RNAs that may have roles in transcriptional regulation and mediating interactions between proteins. Following assessment of different parameters for construction of circRNA sequencing libraries, a protocol was compiled utilizing stranded total RNA library preparation with RNase R pre-treatment and is presented here.
Preparation and Metabolic Assay of 3-dimensional Spheroid Co-cultures of Pancreatic Cancer Cells and Fibroblasts Pawan Noel1, Ruben Muñoz1, George W. Rogers2, Andrew Neilson2, Daniel D. Von Hoff1, Haiyong Han1 1Molecular Medicine Division, Translational Genomics Research Institute, 2Agilent Technologies Here, a method is described for the preparation of 3-dimensional (3D) spheroid co-culture of pancreatic cancer cells and fibroblasts, followed by measurement of metabolic functions using an extracellular flux analyzer.
A Modified Precipitation Method to Isolate Urinary Exosomes Rupesh Kanchi Ravi1, Mahdieh Khosroheidari1, Johanna K. DiStefano1 1Diabetes, Cardiovascular and Metabolic Diseases Division, Translational Genomics Research Institute (TGen) This manuscript describes a protocol for isolating exosomes from human urine using a modified precipitation technique.
