Researchers use antibiotic resistance genes to identify bacteria that possess a plasmid containing their gene of interest. Antibiotic resistance naturally occurs when a spontaneous DNA mutation creates changes in bacterial genes that eliminate antibiotic activity. Bacteria can share these new resistance genes with their offspring and other bacteria. The overuse and misuse of antibiotics have created a public health crisis, as resistant and multi-resistant bacteria continue to develop. Antibiotics, such as penicillin, are drugs that kill or stop bacterial growth. Bacteria that naturally or artificially acquired antibiotic resistance genes do not respond to antibiotics. Scientists exploit this by designing plasmids—small, self-replicating pieces of DNA—that carry both an antibiotic resistance gene and a gene of interest. Antibiotic resistance is an integral part of DNA cloning that allows a researcher to identify cells that absorbed a DNA of interest. The researcher’s DNA of interest is introduced into bacterial cells using a process called transformation. Bacterial transformation involves temporarily creating small holes in the bacterial cell wall to permit the uptake of external DNA such as a plasmid. Only some bacterial cells absorb new DNA. The Periodic Table and Organismal Elements

Elements are the smallest units of matter that cannot be broken down further by chemical processes. There are 118 known elements, but not all of these are naturally-occurring, and fewer still are essential for life. Living matter is composed primarily of carbon, nitrogen, hydrogen, and oxygen, with smaller amounts of other elements like calcium, phosphorus, potassium, and sulfur. Other elements are also necessary for life but only in trace amounts. The periodic table organizes elements based on their physical and chemical properties. The atomic number of an element corresponds to the number of protons found in its nucleus, and each square in the periodic table also provides the full name, chemical symbol, and atomic weight of an element. The number of protons provides information about the size of an element, but it is not the only organizational principle underlying the structure of the periodic table. Elements are organized into columns (groups) and rows (periods) based on other physical and chemical properties, such as reactivity, the location of their outermost electrons, and the ability to make certain types of bonds. Elements in the same group (i.e., column) vary in size but have many chemical properties in common with one another. Deductive Reasoning

When scientists set out to explore and explain natural phenomena, they often start with specific observations that highlight a particular question or problem. They then induce a possible answer or solution, known as a hypothesis. This type of logical thinking, which uses observations to reach general rational conclusions, is called inductive reasoning.

After a hypothesis has been established, scientists deduce that certain events should occur if the hypothesis is true. Researchers use these predictions, the result of deductive reasoning, to test the hypothesis. Compared to inductive reasoning, deductive reasoning works in the opposite direction, starting with general principles or laws (i.e., conclusions) and using them to predict specific results (i.e., future observations). Deductive tests are often formulated as “If...then” statements: if the hypothesis is true, then the prediction should be observed. Although deductive reasoning is at the heart of hypothesis-driven science, whereas inductive reasoning is mostly associated with descriptive science, both forms of logic are integral to research and often tie in together within the same experiments.

Osmoregulation in Insects

Malpighian tubules are specialized structures found in the digestive systems of many arthropods, including most insects, which handle excretion and osmoregulation. The tubules are typically arranged in pairs and have a convoluted structure that increases their surface area.

Malpighian tubules extend from the digestive tract, typically the area between the midgut and hindgut, into the hemolymph—a mixture of blood and interstitial fluid found in insects and other arthropods, as well as most mollusks. Unlike other excretory systems, the excretory processes of Malpighian tubules lack a filtration step. Metabolic wastes, like uric acid, diffuse into the tubules from the hemolymph. The tubules are lined with a layer of transport epithelia. These specialized epithelial cells contain pumps that actively transport ions, like sodium (Na+) and potassium (K+), from the hemolymph into the interior of the tubule, called the lumen. Osmosis allows water to follow ions into the tubules passively. From the tubule lumen, water, ions, and waste travel from the intestine to the rectum. Tiny, protruding microvilli lining the inside of the tubules help maximize solute-water coupling and the propulsion of uric acid crystals through the tubules. In the rectum, specialized glands pump many of the ions back into the hemolymph. Osmosis
