CD4 + T-Lymphozyten Erfassen mit einem Einweg-Mikrofluidik-Chip für HIV

Summary

Protocol

1. Bereiten mikrofluidischen Kanal mit anti-CD4 ⁺ biotinlayted Oberfläche.
2. Injektion von 10 ml Probe Blut bei 5 ml / min in den mikrofluidischen Kanal mit Spritzenpumpe.
3. Spülen Sie das Blut bei 20 ml / min für 20 min mit PBS.
4. Inject verdünnt FACS Lyse-Lösung (verdünnt auf 10% von Stammlösung mit PBS) bei 5ul/min für 15 min.
5. Inkubation: 15 min.
6. Inject primären CD3 ^{+-Antikörper-Lösung} (verdünnt auf 10% von Stammlösung mit PBS) bei 5 ml / min für 20 min.
7. Inkubieren für 2 Stunden.
8. Spülen Sie Kanal mit PBS auf 5 ml / min für 15 min.
9. Inject 20% AF488 CD4 ^{+-Antikörper,} 0,2% DAPI-Stammlösung und 10% sekundäre CD3 ⁺ Antikörper bei 5 ml / min für 20 min zur gleichen Zeit.
10. Inkubieren für 20 min.
11. Spülen Sie den Kanal mit PBS auf 5 ml / min für 15 min.
12. Zählen von Zellen unter Fluoreszenzmikroskop:
    1. White (optisch) - Schwarz und Weiß: Zell-und Staub
    2. UV (DAPI) - Blue: Zelle nur
    3. Blue (CD4 ⁺⁾ - Grün: T-Lymphozyten und Monozyten
    4. Green (CD3 ⁺⁾ - Rot: T-Lymphozyten nur
