Abstract
De hersenen bestaat uit vier primaire celtypen inclusief neuronen, astrocyten, microglia en oligodendrocyten. Hoewel zij niet het meest voorkomende celtype in de hersenen, neuronen de meest bestudeerde van deze celtypen gezien hun directe rol in gedrag beïnvloeden. Andere celtypen in de hersenen beïnvloeden ook neuronale functie en gedrag via de signaalmoleculen ze produceren. Neuroscientists moet de interactie tussen celsoorten inzicht in de hersenen beter begrijpen hoe deze interacties beïnvloeden neurale functie en ziekte. Tot op heden de meest voorkomende werkwijze voor het analyseren eiwit of genexpressie gebruikt de homogenisering van hele weefselmonsters, meestal bloed, en zonder acht te slaan celtype. Deze aanpak is een informatieve aanpak voor de behandeling van algemene veranderingen in gen of eiwit expressie die neurale functie en het gedrag kunnen beïnvloeden; echter, deze analysemethode leent zich niet tot een beter begrip van de cel-type-specifieke genexpressie en het effect van cel-tot-cel communicatie op neurale functie. Analyse van behavioral epigenetica heeft een oppervlakte van groeiende aandacht die wordt onderzocht is hoe wijzigingen van het desoxyribonucleïnezuur (DNA) structuur langetermijneffect genexpressie en gedrag; echter, deze informatie alleen relevant indien geanalyseerd in een celtype-specifieke wijze gezien de differentiële lijn en dus epigenetische markers die aanwezig zijn op bepaalde genen van afzonderlijke neurale celtypes kunnen zijn. De Fluorescentie Activated Cell Sorting (FACS) techniek hierna beschreven is een eenvoudige en effectieve manier om afzonderlijke neurale cellen te isoleren voor daaropvolgende analyse van genexpressie, eiwitexpressie of epigenetische modificaties van DNA. Deze techniek kan ook worden gemodificeerd om specifiekere neurale celtypes te isoleren in de hersenen voor daaropvolgende celtype-specifieke analyse.
