Abstract

Introduction

Protocol
该议定书得到赖特州立大学机构审查委员会的批准,在实验前,每个参与者都征得了知情同意。
1. 主题准备
- 在开始对主体进行任何测量之前,请为 FD-fNIRS 和 DCS 系统预热至少 10 分钟(有关详细信息,请参阅第 2 节和第 3 节)。图1显示了使用紧凑型DCS仪器进行主体测量的示例。
- 首先,使用磁带测量测量每个受试者头部的切口到内因之间的距离(图2A)。
- 以 nasion 作为起点,使用墨迹标记标记到 inion 距离 10% 的位置。这表示 EEG 10/20 蒙太奇的 Fp1 和 Fp2 之间的点(图 2A)。
- 使用 EEG 10/20 帽(参见材料表),调整盖,使标记点介于 Fp1 和 Fp2 之间。

- 调整探测器增益以获得最大信号后,断开探测器中的一个源光纤,并验证相应源光纤的直流电流 (DC) 是否小于每个测量周期的 20 个计数。如果大于此值,则探测器45中可能会泄漏过多的房间光线。如果是这种情况,系统应关闭电源,则应阻止/移除房间内的任何多余的光线,并重复步骤 2.4_2.5。
- 验证来自每个源和探测器的正确信号电平。系统将其定义为每个测量周期的 100 计数和低于 1,500 计数。

3. DCS 设置

- 运行基于 GUI 的 DCS 数据采集软件,该软件显示每个探测器的光子计数和半定量实时血流值。调整光纤位置、角度(光纤面应垂直于皮肤表面)和数据采集时间,以获得至少 5,000 计数/s的信号(对于足够的信噪比)和低于 1,000,000 计数/s(以避免损坏探测器)(图 3A)。
- 通过检查监视器上显示的光子计数级别和近实时自相关曲线,从每个探测器验证足够的光子计数级别(从步骤 3.3 起)。
- 通过检查监视器上显示的自相关曲线的 y 截取,验证足够的光纤接触,而不会产生任何环境光泄漏。最佳值为 ±1.5,不使用偏振器(图 3B)。
- 通过拧紧弹性带,使其足够紧,足以抵抗运动,但足够宽松,以防止物体有任何不适,验证探头和测量值是否容易产生运动伪影。用户还应同时检查监视器上的自相关曲线,以便自相关曲线衰减为 1,延长相关时间(> 10 ms)(图 3C)。
4. 数据收集
- 指示受试者在 8 分钟测量期间尽量减少任何运动。
- 关灯,确保受试者坐在一个舒适的位置,闭上眼睛。
- 使用将 FD-fNIRS 系统光学探头放置在 DCS 探头旁边的前额上,执行基准 FD-fNIRS 测量。然后,按 FD-fNIRS 获取 GUI 中的"获取"按钮。这些数据将提供静态光学特性、吸收参数和散射参数(μa ,μ's),s用于动态光学参数CBFi17、20,20的定量。
- 完成 FD-fNIRS 测量后,通过按下 DCS 数据采集 GUI 中的"运行"按钮,开始对光学 DCS 测量数据采集数据。收集数据共 8 分钟,最大集成时间为 2 秒(根据每个主体的信号噪声比,首选较少)。
- 如有必要,在初始实验的 1 小时内重复实验,或在一天的类似时间重复实验,以减少外部变化,如疲劳、兴奋剂或温度。
5. 数据分析

Representative Results
使用DCS测量功能连接的可行性被成功消除39。测量了九个受试者前额皮质的休息状态功能连接。结果显示,左侧区域区域(0.64 ± 0.25)和右侧 (0.62 = 0.23) 皮质的相关性较高, 与左侧区域间区域(0.32 = 0.32)、(0.34 ± 0.27)和右侧(0.34 ± 0.29)、(0.34 ± 0.26)皮质相比。(图5)。还进行了功率为0.8和显著性水平为0.05的功率分析,结果功率为0.82,样本大小为8(低于本研究分析的受试者数量)。

Discussion
为了确定由DCS测量的CBF是否准确检测到RSFC,检查了大脑中具有已知RSFC特性的两个区域。DLFC 区域之间以及 DLFC 和 IFC 之间的功能连接假定存在57、58,58、59。,选择左和右两个站点之间的连接,因为区域内连接通常较高。此外,IFC和DLFC之间的连接被选择,因为区域间连接已知较弱。
