  1. Hi,

    This is probably your most frequently asked questions. I have the correct Collagenase and I add 30 mg to the ²0 mL prewarmed HBSS with ² mM EDTA. Despite 1 hour at 37 at ²00 the intestine is very stubborn and refuses to digest. I mince it just as described. Any thoughts?? Should I work on increasing the collagenase concentration?

  2. Hello there,

    Thank you for your interest in the protocol. Your question seems very perplexing as we have never experienced difficulties with tissue digestion durations greater than 15 minutes, and you seem to be having difficulties after 1 hour. We do not use the ² mM EDTA during tissue digestion and the recommendations I can make are: do not digest the tissue for 1 hour, ensure that you are using an orbital shaker with the 50 ml conical tubes lying horizontally, mince the tissue more finely, and vortex for 50 ml conical tubes after digestion for about 15 to ²0 seconds so that any remaining intestinal tissue is dissociated and digested (you may get some strands of stromal tissue remaining at the end, which is normal).
