[Exercise training in hypoxia prevents hypoxia induced mitochondrial DNA oxidative damage in skeletal muscle].

This study was undertaken to investigate the effect of exercise training on mitochondrial DNA (mtDNA) oxidative damage and 8-oxoguanine DNA glycosylase-1 (OGG1) expression in skeletal muscle of rats under continuous exposure to hypoxia. Male Sprague-Dawley rats were randomly divided into 4 groups (n = 8): normoxia control group (NC), normoxia training group (NT), hypoxia control group (HC), and hypoxia training group (HT). The hypoxia-treated animals were housed in normobaric hypoxic tent containing 11.3% oxygen for consecutive 4 weeks. The exercise-trained animals were exercised on a motor-driven rodent treadmill at a speed of 15 m/min, 5% grade for 60 min/day, 5 days per week for 4 weeks. The results showed that, compared with NC group, hypoxia attenuated complex I, II, IV and ATP synthase activities of the electron transport chain, and the level of mitochondrial membrane potential in HC group (P < 0.05 or P < 0.01). Moreover, hypoxia decreased mitochondrial OGG1, MnSOD, and GPx activities (P < 0.05 or P < 0.01), whereas elevated reactive oxygen species (ROS) generation and the level of 8-oxo-deoxyguanosine (8-oxodG) in mtDNA (P < 0.01). Furthermore, hypoxia attenuated muscle and mitochondrial [NAD(+)]/ [NADH] ratio, and SIRT3 protein expression (P < 0.05 or P < 0.01). Compared with HC group, exercise training in hypoxia elevated complex I, II, IV and ATP synthase activities, and the level of mitochondrial membrane potential in HT group (P < 0.05 or P < 0.01). Moreover, exercise training in hypoxia increased MnSOD and GPx activities and mitochondrial OGG1 level (P < 0.01), whereas decreased ROS generation and the level of 8-oxodG in mtDNA (P < 0.01). Furthermore, exercise training in hypoxia increased muscle and mitochondrial [NAD(+)]/[NADH] ratio, as well as SIRT3 protein expression (P < 0.05 or P < 0.01). These findings suggest that exercise training in hypoxia can decrease hypoxia-induced mtDNA oxidative damage in the skeletal muscle through up-regulating exercise-induced mitochondrial OGG1 and antioxidant enzymes. Exercise training in hypoxia may improve hypoxia tolerance in skeletal muscle mitochondria via elevating [NAD(+)]/[NADH] ratio and SIRT3 expression.

Midventricular obstructive hypertrophic cardiomyopathy (MVOHCM) is a rare form of hypertrophic cardiomyopathy. Knowledge regarding the diagnosis, morbidity and cardiovascular mortality is limited. In this study, we aimed to describe the long-term outcomes of patients with MVOHCM followed in a tertiary referral centre.Methods A retrospective study of 60 patients with MVOHCM diagnosed at FuWai Hospital was performed. Clinical features, mortality and cardiovascular morbidity were analysed.

Automatic blastomere recognition from a single embryo image.

The number of blastomeres of human day 3 embryos is one of the most important criteria for evaluating embryo viability. However, due to the transparency and overlap of blastomeres, it is a challenge to recognize blastomeres automatically using a single embryo image. This study proposes an approach based on least square curve fitting (LSCF) for automatic blastomere recognition from a single image. First, combining edge detection, deletion of multiple connected points, and dilation and erosion, an effective preprocessing method was designed to obtain part of blastomere edges that were singly connected. Next, an automatic recognition method for blastomeres was proposed using least square circle fitting. This algorithm was tested on 381 embryo microscopic images obtained from the eight-cell period, and the results were compared with those provided by experts. Embryos were recognized with a 0 error rate occupancy of 21.59%, and the ratio of embryos in which the false recognition number was less than or equal to 2 was 83.16%. This experiment demonstrated that our method could efficiently and rapidly recognize the number of blastomeres from a single embryo image without the need to reconstruct the three-dimensional model of the blastomeres first; this method is simple and efficient.

This study was aimed to investigate the genetic characteristics, identification method and transfusion strategy of rare blood type B(A). The rare blood group B(A) was typed by serological technique, PCR-SSP genotyping and sequencing of exon 6, 7 of ABO blood group. The genetic characteristics and molecular mechanism of B(A) blood group were also analyzed. Blood group compatibility test was conducted between blood donors of B(A) and recipients by clinical transfusion. The results showed that both forward and reverse grouping did not match the 3 cases of serological result in their family survey, while all of the 3 cases were grouped as AB blood group by forward grouping, B blood group by reverse grouping with serological result and B(A)04/001 group were genotyped by ABO genotyping. The patient of B blood group was transfused by 1 bag of washed red blood cells of donor of B(A) under closely monitroring, the patients condition changed, and a mild adverse transfusion reaction was appeared. Washed red blood cell of O blood group was transfused into B(A) patient without blood transfusion reaction. It is concluded that the forward ABO serological grouping and reverse ABO serological grouping are not compatible, that may be verified by family survey and molecular biological methods. If in some cases transfusion therapy was applied, and group B(A) can not be transfused to the patient with group B or AB. Thus, transfusion compatibility or autologous transfusion can be adopted to transfuse to the patient from group B(A).

The meteorological satellite spectral image is an effective tool for researches on meteorological science and environmental remote sensing science. Image registration is the basis for the application of the meteorological satellite spectral image data. In order to realize the registration of the satellite image and the template image, a new registration method based on the Fourier-Mellin transform is presented in this paper. Firstly, we use the global coastline vector map data to build a landmark template, which is a reference for the meteorological satellite spectral image registration. Secondly, we choose infrared sub-image of no cloud according to the cloud channel data, and extract the edges of the infrared image by Sobel operator. Finally, the affine transform model parameters between the landmark template and the satellite image are determined by the Fourier-Mellin transform, and thus the registration is realized. The proposed method is based on the curve matching in essence. It needs no feature point extraction, and can greatly simplify the process of registration. The experimental results using the infrared spectral data of the FY-2D meteorological satellite show that the method is robust and can reach a high speed and high accuracy.

This study was aimed to investigate one case with rare type B(A) in ABO blood group by using serological and molecular biological methods, and analyze the cause of inconsistency resulting from multiple detections. The serological method was used to identify the serum type of ABO blood group, at the same time the PCR sequencing method was used to detect the genotypes. The results indicated that the group typing and reverse typing for the blood donor were inconsistent, the group typing was AB, the reverse typing was B. The ABO genotype was B(A) 04 /001. This genotype was involved in nt640A > G point mutation which caused valine replacing methionine at 214. It is concluded that the sample inconsistent between the group typing and reverse typing could be typed by molecular biological method, and the molecular basis of weak expression of ABO blood group is elucidated too.

The degradation behavior of polypropylene and polypropylene/stabilizer composites, caused by gamma radiation, was studied in the present work The stabilizers used were hindered phenol antioxidant and hindered amine light stabilizer. FTIR spectroscopy and DSC analysis were used to determine the structural variation induced by gamma radiation. It can be seen that the evolution of PP and composites PP/stabilizers on gamma irradiation is an increase in absorbance in the hydroxyl and carbonyl absorption regions. Carbonyl index calculated from FTIR spectra was used to characterize the rate of degradation. When the irradiation dose was small (<50 kGy), the degradation of pure polypropylene and polypropylene/stabilizers composites was not obvious; while the irradiation dose increased (> or =50 kGy), the carbonyl indexes of all the samples increased obviously, and the degradation degree of polypropylene/stabilizer composites was bigger than pure polypropylene. This result might be partially attributed to the molecular chain scission of hindered phenol and hindered amine under larger irradiation dose. The chain scission of stabilizers forms hydroperoxides and peroxide radicals, catalyzing the degradation of polypropylene. As the irradiation dose was small (<50 kGy), the crystallization temperatures of pure polypropylene and polypropylene/stabilizer composites all showed no remarkable changes; as the irradiation dose exceeded 50 kGy, the crystallization temperatures of pure polypropylene and polypropylene/stabilizer composites all decreased obviously. Correspondingly, the melting peaks of both pure polypropylene and polypropylene/stabilizer composites moved to lower temperature and split into two peaks with increasing the irradiation dose. The decrease of crystallization and melting temperatures might be attributed to the destruction of chemical structure and stereo-regularity of the molecular chain, due to the increasing of carbonyl and hydroxyl groups brought by the oxidation of polypropylene molecular chain. At the same irradiation dose (> or = 50 kGy), the crystallization and melting temperatures of polypropylene/stabilizer composites were lower than that of pure polypropylene. It indicates that the stabilizers accelerate the degradation of polypropylene. The results were accordant with the IR results.

This study was aimed to investigate the serologic characteristics, genetic background and population distribution of B2 and AB2 subtype in Chinese ABO blood group. The classic blood group serological technology was used to detect ABO blood group of the propositus and their family members, the anti-B1 serum prepared by yourself was used to investigate the distribution of B1/B2 and AB1/AB2 subtype of the blood donor. The results indicated that the antigen of propositus was AB2 subtype and that of his child was B2 subtype. The anti-B1 antibody was detected in blood serum of propositus; the antigen of 3 from 2318 blood donors with B blood group were found to be B2 subtype, the antigen of 2 from 826 blood donors with AB blood group were found to be AB2 subtype. The investigation on propositus and the 3 B2 blood donor families showed that B2 antigen displays genetic characteristics of blood group. It is concluded that B2/AB2 subtype is from family inheritance, while B2 subtype is amounted to 0.129% in B blood group, and AB2 subtype is amounted to 0.224% in AB blood group.

The J-aggregates and the photosensitive properties of two kinds of mixed cyanine dye and the corresponding independent cyanine dyes absorbed on AgBr cubic microcrystal were investigated. The reflection spectra of the J-aggregates of the mixed cyanine dyes and the corresponding independent cyanine dyes were measured by the use of UV-Vis spectroscopy. The results showed that the absorption band of the mixed J-aggregates absorbed on AgBr cubic microcrystal had two-peak wavelengths and accorded with the independent cyanine dyes, respectively. The sensitive performance of the mixed cyanine dyes was much enhanced compared to the corresponding cyanine dyes.

The rapid development of astronomical observation has led to many large sky surveys such as SDSS (Sloan digital sky survey) and LAMOST (large sky area multi-object spectroscopic telescope). Since these surveys have produced very large numbers of spectra, automated spectral analysis becomes desirable and necessary. The present paper studies the spectral classification method based on Bayes decision theory, which divides spectra into three types: star, galaxy and quasar. Firstly, principal component analysis (PCA) is used in feature extraction, and spectra are projected into the 3D PCA feature space; secondly, the class conditional probability density functions are estimated using the non-parametric density estimation technique, Parzen window approach; finally, the minimum error Bayes decision rule is used for classification. In Parzen window approach, the kernel width affects the density estimation, and then affects the classification effect. Extensive experiments have been performed to analyze the relationship between the kernel widths and the correct classification rates. The authors found that the correct rate increases with the kernel width being close to some threshold, while it decreases with the kernel width being less than this threshold.

Photooxidation degradation of polypropylene (PP)/organomontmorillonite (OMMT) nanocomposites was studied under UV irradiation at wavelength 365 nm. The OMMT was dispersed homogeneously in the PP matrix. The structural variation induced by photooxidation was detected by FTIR spectroscopy. It can be seen that the evolution of PP and nanocomposites PP/OMMT on irradiation is an increase in absorbance in the hydroxyl and carbonyl absorption regions. Carbonyl index was used to characterize the rate of photooxidative degradation. At the early stage of irradiation (shorter than 11 hours), the rate of photooxidative degradation of PP/OMMT nanocomposites was much faster than that of pure PP. With the increase in the irradiation time, the rate of photooxidative degradation of PP/OMMT nanocomposites became slower than that of pure PP. On the other hand, the OMMT content can also influence the photooxidative degradation rate. The variation in the photooxidative degradation rate is proposed to originate from the competition between the light shielding effect and accelerating photo-oxidative degradation effect of organomontmorillonite. These two effects play roles in the photooxidation process of PP simultaneously. At the early stage of irradiation, the accelerating photooxidative degradation effect dominates. With the increase in the irradiation time, the active sites such as SiOH, --OH etc and transition metal ions (Fe2+, Fe3+ etc) in the OMMT which can accelerate photooxidation are gradually consumed, and then the light shielding effect may play the main role. The method of curve-fitting was used to characterize the influence of organomontmorillonite on the photooxidative degradation products. Compared with pure PP, the content of products of carboxylic acid and anhydride were found to increase, whereas that of ester decreases in PP/OMMT nanocomposites.

[Investigation of N-hexadecylated polycaprolactam by infrared spectroscopy].

N-hexadecylated polycaprolactam (PA6) comb-like polymers (PA6C16) with different alkylation degree, were synthesized using the method of N-alkylation reaction. The thermal transition behavior of these polymers were characterized by a combination of differential scanning calorimetry (DSC) and Fourier transform infrared spectroscopy (FTIR). DSC results indicated that the hexadecyl side chain can crystallize. Furthermore, the melting point of the hexadecyl side chain crystallization of the sample with high alkylation degree(PA6C16-H) is lower than that of sample with lower alkylation degree(PA6C16-L). FT-IR spectroscopic results showed that the methylene groups of the hexadecyl side chain of PA6C16-L tend to crystallize in orthorhombic form, while the methylene groups of the hexadecyl side chain of PA6C16-H tend to be packed into hexagonal form. Variable temperature IR spectroscopic results revealed that accompanying the melting of the side chain crystallization, the backbones of PA6C16-H undergo a conformation transition, while the conformation of backbones of PA6C16-L shows no obvious transition.

N-hexadecylated poly(p-phenylene terephthamide) (PPTA) comb-like polymers (PPTAC16), which are composed of PPTA backbone and hexadecyl side chains, were prepared by method of N-alkylation reaction. The molecular structure and thermal transition behavior of PPTAC16 were characterized by a combination of differential scanning calorimetry (DSC) and Fourier transform infrared spectroscopy (FT-IR) technique. DSC results indicated that the crystallization of hexadecyl side chains, and the side chain crystallization is less perfect than that of n-hexadecane crystal. IR results showed that the ordered degree of the packing of CH2 group in the side chain crystallization is lower than that of n-hexadecane crystal. Variable temperature IR spectroscopic results revealed that the stretching vibration and bending vibration bands of methylene group experience an abrupt change with increasing of temperature which indicate the melting of the side chain crystallization, furthermore the main chain of PPTAC16 was found to undergo an irreversible change accompanying the melting of the alkyl side chain crystallization. And the molten state of CH2 group in the hexadecyl of PPTAC16 is different from that of liquid n-hexadecane.
