Genetic-morphometric variation in Culex quinquefasciatus from Brazil and La Plata, Argentina.
Mem. Inst. Oswaldo Cruz
PUBLISHED: 04-29-2010
Show Abstract
Hide Abstract
Variation among natural populations of Culex (Culex) quinquefasciatus Say is associated with different vectorial capacities. The species Cx. quinquefasciatus is present in the equatorial, tropical and subtropical zones in the Brazilian territory, with intermediate forms between Cx. quinquefasciatus and Culex pipiens occurring in regions of latitudes around 33°-35° S. Herein, we studied geographically distinct populations of Cx. quinquefasciatus by genetic characterization and analysis of intra-specific wing morphometrics. After morphological analysis, molecular characterization of Cx. quinquefasciatus and intermediate forms was performed by polymerase chain reaction of the polymorphic nuclear region of the second intron of the acetylcholinesterase locus. Additionally, the morphology of adult female wings collected from six locations was analyzed. Wing centroid sizes were significantly different between some geographical pairs. Mean values of R2/R2+3 differed significantly after pairwise comparisons. The overall wing shape represented by morphometric characters could be divided into two main groupings. Our data suggest that Brazilian samples are morphologically and genetically distinct from the Argentinean samples and also indicated a morphological distinction between northern and southern populations of Brazilian Cx. quinquefasciatus. We suggest that wing morphology may be used for preliminary assessment of population structure of Cx. quinquefasciatusin Brazil.

[Assessment of the cervical cancer screening in the Family Health Strategy in Amparo, São Paulo State, Brazil].

Show Abstract
Hide Abstract
Uterine cervical cancer mortality has not been declining. The territorial distribution and registration of families in Brazils Family Health Strategy help identify women that have performed a Pap smear or that have not had one for more than three years. This study analyzed whether cervical cancer screening in Amparo, São Paulo State, Brazil, made progress in complying with the prevailing guidelines during the seven years of experience with the Strategy. The annual examination rate remained high, with a slight trend towards greater intervals between follow-up tests. Distribution of tests tended to increase in the 40-59-year age bracket and decrease in the over-60 age group, while annual coverage tended to decrease. The proportions of excess tests varied from 61.2% to 65.5%. Concluding, the upgrading of cervical cancer screening was slight, and did not change the opportunistic pattern of follow-up tests. Considering that community health agents can act to increase the coverage of these measures, it is essential to train them for this work.

Effect of dietary supplementation with collagen hydrolysates on bone metabolism of postmenopausal women with low mineral density.
Maturitas
PUBLISHED: 06-23-2009
Show Abstract
Hide Abstract
Collagen hydrolysates (CH), obtained by an enzymatic hydrolysis process of gelatins, have potential application as oral supplements. Objective: To evaluate the effects of diet supplementation with collagen hydrolysates on bone metabolism markers in postmenopausal women with low bone mineral density. Methods: A randomized double-blind clinical assay with postmenopausal women with osteopenia was planned. The volunteers ingested 10g/day of CH or a placebo for a period of 24 weeks. The bone resorption markers (carboxyl-terminal collagen crosslinks-CTX) and bone formation markers (osteocalcin-OSCAL and bone-specific alkaline phosphatase-BAP) were determined at baseline and at 12 and 24 weeks of treatment. Results: The sample consisted of 35 placebo and 36 treated subjects (aged 57.3+/-4.8 years and BMI of 27.4+/-4.5kg/m(2)). BAP levels showed no significant changes over the time. CTX levels showed a significant increase during the 24 weeks of treatment from 0.40 to 0.48ng/mL in CH group and 0.47 to 0.57ng/mL in placebo group (p<0.0001). OSCAL levels also showed a increase during the 24 weeks from 24.8 to 29.0ng/mL in CH group and 28.1 to 31.8ng/mL in placebo group (p<0.001). A comparison of levels of bone markers between CH and placebo group demonstrated no differences. Analysis of variance revealed no significant effect of dietary supplementation with collagen hydrolysates on biochemical bone markers. Conclusions: CH consumption did not produce any effects on bone metabolism as measured by biochemical indices of bone remodeling in postmenopausal women. The majority of patients exhibited inadequate calcium intake, as well as excess body weight.

Brazilian mosquito (Diptera: Culicidae) fauna: I. Anopheles species from Porto Velho, Rondônia state, western Amazon, Brazil.
Rev. Inst. Med. Trop. Sao Paulo
Show Abstract
Hide Abstract
This study contributes to knowledge of Anopheles species, including vectors of Plasmodium from the western Brazilian Amazon in Porto Velho, Rondônia State. The sampling area has undergone substantial environmental changes as a consequence of agricultural and hydroelectric projects, which have caused intensive deforestation and favored habitats for some mosquito species. The purpose of this study was to diagnose the occurrence of anopheline species from collections in three locations along an electric-power transmission line. Each locality was sampled three times from 2010 to 2011. The principal adult mosquitoes captured in Shannon trap were Anopheles darlingi, An. triannulatus, An. nuneztovari l.s., An.gilesi and An. costai. In addition, larvae were collected in ground breeding sites for Anopheles braziliensis, An. triannulatus, An. darlingi, An. deaneorum, An. marajoara, An. peryassui, An. nuneztovari l.s. and An. oswaldoi-konderi. Anopheles darlingi was the most common mosquito in the region. We discuss Culicidae systematics, fauna distribution, and aspects of malaria in altered habitats of the western Amazon.

Low genetic diversity in Wolbachia-Infected Culex quinquefasciatus (Diptera: Culicidae) from Brazil and Argentina.
Rev. Inst. Med. Trop. Sao Paulo
Show Abstract
Hide Abstract
Culex quinquefasciatus is a vector of human pathogens, including filarial nematodes and several viruses. Although its epidemiological relevance is known to vary across geographical regions, an understanding of its population genetic structure is still incipient. In light of this, we evaluated the genetic diversity of Cx. quinquefasciatus and Cx. pipiens x Cx. quinquefasciatus hybrids collected from nine localities in Brazil and one site in Argentina. We used mitochondrial genes cox1 and nd4, along with the coxA and wsp genes of the maternally-inherited Wolbachia endosymbiont. The nd4 fragment was invariant between samples, whilst cox1 exhibited four haplotypes that separated two types of Cx. quinquefasciatus, one clustered in southern Brazil. Low sequence diversity was generally observed, being discussed. Both Brazilian and Argentinian mosquitoes were infected with a single Wolbachia strain. As reported in previous studies with these populations, cox1 and nd4 diversity is not congruent with the population structure revealed by nuclear markers or alar morphology. Future Cx. quinquefasciatus research should, if possible, evaluate mtDNA diversity in light of other markers.

What is Visualize?

JoVE Visualize is a tool created to match the last 5 years of PubMed publications to methods in JoVE's video library.

How does it work?

We use abstracts found on PubMed and match them to JoVE videos to create a list of 10 to 30 related methods videos.

Video X seems to be unrelated to Abstract Y...

In developing our video relationships, we compare around 5 million PubMed articles to our library of over 4,500 methods videos. In some cases the language used in the PubMed abstracts makes matching that content to a JoVE video difficult. In other cases, there happens not to be any content in our video library that is relevant to the topic of a given abstract. In these cases, our algorithms are trying their best to display videos with relevant content, which can sometimes result in matched videos with only a slight relation.
