Correlating multidimensional fetal heart rate variability analysis with acid-base balance at birth.

Fetal monitoring during labour currently fails to accurately detect acidemia. We developed a method to assess the multidimensional properties of fetal heart rate variability (fHRV) from trans-abdominal fetal electrocardiogram (fECG) during labour. We aimed to assess this novel bioinformatics approach for correlation between fHRV and neonatal pH or base excess (BE) at birth.We enrolled a prospective pilot cohort of uncomplicated singleton pregnancies at 38-42?weeks' gestation in Milan, Italy, and Liverpool, UK. Fetal monitoring was performed by standard cardiotocography. Simultaneously, with fECG (high sampling frequency) was recorded. To ensure clinician blinding, fECG information was not displayed. Data from the last 60?min preceding onset of second-stage labour were analyzed using clinically validated continuous individualized multiorgan variability analysis (CIMVA) software in 5?min overlapping windows. CIMVA allows simultaneous calculation of 101 fHRV measures across five fHRV signal analysis domains. We validated our mathematical prediction model internally with 80:20 cross-validation split, comparing results to cord pH and BE at birth.The cohort consisted of 60 women with neonatal pH values at birth ranging from 7.44 to 6.99 and BE from -0.3 to -18.7?mmol?L(-1). Our model predicted pH from 30 fHRV measures (R(2) = 0.90, P < 0.001) and BE from 21 fHRV measures (R(2) = 0.77, P < 0.001).Novel bioinformatics approach (CIMVA) applied to fHRV derived from trans-abdominal fECG during labor correlated well with acid-base balance at birth. Further refinement and validation in larger cohorts are needed. These new measurements of fHRV might offer a new opportunity to predict fetal acid-base balance at birth.

Helminth.net (http://www.helminth.net) is the new moniker for a collection of databases: Nematode.net and Trematode.net. Within this collection we provide services and resources for parasitic roundworms (nematodes) and flatworms (trematodes), collectively known as helminths. For over a decade we have provided resources for studying nematodes via our veteran site Nematode.net (http://nematode.net). In this article, (i) we provide an update on the expansions of Nematode.net that hosts omics data from 84 species and provides advanced search tools to the broad scientific community so that data can be mined in a useful and user-friendly manner and (ii) we introduce Trematode.net, a site dedicated to the dissemination of data from flukes, flatworm parasites of the class Trematoda, phylum Platyhelminthes. Trematode.net is an independent component of Helminth.net and currently hosts data from 16 species, with information ranging from genomic, functional genomic data, enzymatic pathway utilization to microbiome changes associated with helminth infections. The databases' interface, with a sophisticated query engine as a backbone, is intended to allow users to search for multi-factorial combinations of species' omics properties. This report describes updates to Nematode.net since its last description in NAR, 2012, and also introduces and presents its new sibling site, Trematode.net.

The increased prevalence of multi-drug resistant (MDR) pathogens heightens the need to design new antimicrobial agents. Antimicrobial peptides (AMPs) exhibit broad-spectrum potent activity against MDR pathogens and kills rapidly, thus giving rise to AMPs being recognized as a potential substitute for conventional antibiotics. Designing new AMPs using current in-silico approaches is, however, challenging due to the absence of suitable models, large number of design parameters, testing cycles, production time and cost. To date, AMPs have merely been categorized into families according to their primary sequences, structures and functions. The ability to computationally determine the properties that discriminate AMP families from each other could help in exploring the key characteristics of these families and facilitate the in-silico design of synthetic AMPs.

One-Year Outcomes of Emergency Department Patients With Atrial Fibrillation: A Prospective, Multicenter Registry in China.

There is lack of data about patient characteristics, practice patterns, and long-term adverse outcomes in patients with atrial fibrillation (AF) attending emergency departments (EDs) in China. A total of 2016 patients from 20 representative EDs were included. During 1 year, all-cause mortality was 291 (14.6%) cases, stroke/noncentral nervous system systemic embolism rate was 159 (8.0%) cases, and major bleeding was 26 (1.3%) cases. Heart failure, the major cause of mortality, accounted for 43.0% of deaths. Of 375 (18.6%) patients who used warfarin at baseline, only 217 (57.9%) patients were still on anticoagulation therapy during 1-year follow-up. Compared with the patients who continued on warfarin, the mortality rate was higher in those who did not continue (15.9% vs 5.5%, P < .001). Patients seen in ED with AF appear to have a high incidence rate of long-term all-cause mortality and inadequate anticoagulation rate.

The crosstalk severely affects the viewing experience for the auto-stereoscopic 3D displays based on frontal projection lenticular sheet. To suppress unclear stereo vision and ghosts are observed in marginal viewing zones(MVZs), aberration of the lenticular sheet combining with the frontal projector is analyzed and designed. Theoretical and experimental results show that increasing radius of curvature (ROC) or decreasing aperture of the lenticular sheet can suppress the aberration and reduce the crosstalk. A projector array with 20 micro-projectors is used to frontally project 20 parallax images one lenticular sheet with the ROC of 10 mm and the size of 1.9 m × 1.2 m. The 3D image with the high quality is experimentally demonstrated in both the mid-viewing zone and MVZs in the optimal viewing plane. The 3D clear depth of 1.2m can be perceived. To provide an excellent 3D image and enlarge the field of view at the same time, a novel structure of lenticular sheet is presented to reduce aberration, and the crosstalk is well suppressed.

A Gram-stain-negative, strictly aerobic, non-flagellated, non-gliding, oxidase-positive and rod shaped yellow-pigmented bacterium, designated strain SW169T, was isolated from surface seawater sample collected from the South Pacific Gyre (27°55' S 123°10' W) during the Integrated Ocean Drilling Program (IODP) Expedition 329. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain SW169T belonged to the genus Spongiibacterium and showed the highest 16S rRNA gene sequence similarity with Spongiibacterium flavum A11T (95.9%). It showed 93.2-94.8% 16S rRNA gene sequence similarity with respect to members of the genera Croceitalea and Flagellimonas in the family Flavobacteriaceae. Optimal growth occurred in the presence of 3-4% (w/v) NaCl, at pH 8.0 and at 28°C. The DNA G+C content of strain SW169T was 43.3 mol%. The major fatty acids were iso-C15:1 G, iso-C15:0, iso-C17:0 3-OH, iso-C15:0 3-OH and Summed Feature 3 (comprising C16:1 ?7c and/or C16:1 ?6c,). The major respiratory quinone was menaquinone-6. The major polar lipids were phosphatidylethanolamine, one unidentified aminolipid and one unidentified lipid. On the basis of the polyphasic analyses, strain SW169T is considered to represent a novel species of the genus Spongiibacterium, for which the name Spongiibacterium pacificum sp. nov. is proposed. The type strain is SW169T (=JCM 18379T =LMG 26997T).

Objectives: To conduct a pharmacoeconomic assessment of thrombolysis by intravenous recombinant tissue plasminogen activator (rt-PA) therapy for acute ischemic stroke (AIS) in 6 hours in a tertiary hospital in China. Materials and methods: A retrospective analysis was conducted using medical records data among patients hospitalized for AIS and receiving treatment with or without rt-PA (6 hours after AIS) from January 2006 to December 2008. The conservative therapy group was matched (1?1) on age, gender, risk factors, Glasgow Coma Scale, and National Institutes of Health Stroke Scale (NIHSS). Two groups were compared on fourteenth-day clinical outcomes. Utilities were estimated from modified Rankin Scale (mRS) scores and costs. All cost data reflected 2006-2008 values. Results: A total of 152 patients were enrolled in the study (76 patients in each group). No differences were found in mortality rate between these two groups at day 14. Among survivors on day 14, NIHSS and mRS were lower in rt-PA than conservative group. Barth Index (BI) was higher in the rt-PA group versus conservative group. The rt-PA group had a higher pharmacy cost (¥13?065±4197 versus ¥9622±5439; P ?=? 0·002) compared to the conservative group, but the total cost was not significantly different. Compared to conservative care, rt-PA therapy was associated with incremental cost of ¥4122 with 0·04 utilities gained, yielding an incremental cost-effectiveness ratio of ¥103?050 ($14?231) per utility gained. One-way sensitivity analysis showed that the results were most sensitivity to utility. Conclusions: Intravenous rt-PA was associated with lower patients' disabilities, fewer in-hospital days, and comparable total costs compared to conservative therapy for the management of AIS.

Abstract 1.? Endogenous compounds have been reported to be the regulators of UDP-glucuronosyltransferases (UGTs) isoforms. This study aims to investigate the regulatory effects of the activity of UGT isoforms by two important lipid components phosphatidylcholine (PC) and lysophosphatidylcholines (LPC) using in vitro incubation system. 2.? UGTs supersomes-catalyzed 4-methylumbelliferone (4-MU) glucuronidation was used as the probe reaction to evaluate the inhibition of compounds towards UGT isoforms except UGT1A4, and UGT1A4-catalyzed trifluoperazine (TFP) glucuronidation reaction was utilized to phenotype the activity of UGT1A4. 3.? About 50??M of LPC15:0, LPC16:0, LPC17:0, LPC18:0, LPC18:1 and PC16:0, 2:0 exhibited inhibition towards more than 90% activity of UGT isoforms, and other LPC and PC components showed negligible inhibitory potential towards all the UGT isoforms. UGT1A6 and UGT1A8 were identified to be the most sensitive UGT isoforms susceptible for the inhibition by LPC15:0, LPC16:0, LPC17:0, LPC18:0, LPC18:1 and PC16:0, 2:0, indicating the strong influence of these LPC and PC components towards UGT1A6 and UGT1A8-catalyzed metabolic reaction when the concentrations of these components increased.

Human antibodies are beginning to draw attention for use in immune gene therapy. The efficient generation of effective therapeutic monoclonal antibodies suitable for the treatment of cancers and infectious diseases would be enormously valuable. Antibody display methods are increasingly used to screen human monoclonal antibodies. Here we report the construction of a mammalian cell display method derived from a naive antibody repertoire, for which human single-chain variable fragments (scFv) have been transiently displayed on 293T cell surfaces based on a pDisplay vector. The sizes of the current pDisplay-scFv antibody repertoires have been estimated to be 0.74 × 10(7). An immunoblot assay confirmed the expression of the scFv antibody library. The subcellular distribution of ErbB3-scFv expression plasmid facilitated the display of ErbB3 scFv on the cell membrane surface and the efficiency of the display was evaluated by fluorescence-activated cell sorting. This method of mammalian cell display was verified by successfully screening ErbB3 scFv candidates. A published scFv control was used to confirm the feasibility of the ErbB3 scFv screening process. Three ErbB3 scFv candidates were produced and they were found to have affinity similar to the published scFv candidate. Thus, the present screening system provided an optimal alternative for rapid acquisition of a novel candidate scFv sequence to target genes with high affinity in vitro.

To build a patient specific respiratory motion model with a low dose, a novel method was proposed that uses a limited number of 3D lung CT volumes with an external respiratory signal. 4D lung CT volumes were acquired for patients with in vitro labeling on the upper abdominal surface. Meanwhile, 3D coordinates of in vitro labeling were measured as external respiratory signals. A sequential correspondence between the 4D lung CT and the external respiratory signal was built using the distance correlation method, and a 3D displacement for every registration control point in the CT volumes with respect to time can be obtained by the 4D lung CT deformable registration. A temporal fitting was performed for every registration control point displacements and an external respiratory signal in the anterior-posterior direction respectively to draw their fitting curves. Finally, a linear regression was used to fit the corresponding samples of the control point displacement fitting curves and the external respiratory signal fitting curve to finish the pulmonary respiration modeling. Compared to a B-spline-based method using the respiratory signal phase, the proposed method is highly advantageous as it offers comparable modeling accuracy and target modeling error (TME); while at the same time, the proposed method requires 70% less 3D lung CTs. When using a similar amount of 3D lung CT data, the mean of the proposed method's TME is smaller than the mean of the PCA (principle component analysis)-based methods' TMEs. The results indicate that the proposed method is successful in striking a balance between modeling accuracy and number of 3D lung CT volumes.

Tin (Sn) is widely used in daily life and distributed in many tissues and nutrients. Although over-ingestion of Sn can cause health problems, relatively little attention has been given to the toxic effects of Sn in livestock health and productivity. This study was performed to investigate the toxic effects of prolonged high intake of dietary Sn on broilers. 150 one-day-old Avian broilers were randomly divided into five treatment groups, with five replicates of six birds. For 6 weeks, each group was fed a corn-soybean basal diet (BD) or BD?+?Sn (as SnCl2) at 120, 240, 480, or 720 mg/kg, respectively. Compared with the control, hepatic glutathione peroxidase (GPX) and superoxide dismutase (SOD) activities were significantly decreased when supplemented with Sn up to 480 mg/kg, while malondialdehyde (MDA) was increased until Sn supplementation at 720 mg/kg. Moreover, dietary Sn supplementation at 720 mg/kg decreased BW gain, feed intake, and impaired feed conversion ratio. The 720 mg Sn/kg group also increased activities of alkaline phosphatase (AKP), while decreased hemoglobin (HGB), red blood cell (RBC), and hematocrit (HCT) in the blood. Furthermore, the accumulation of Sn in various tissues was dose dependent on Sn ingestion. It was found that the tibia and feather are the two main tissues for Sn accumulation, followed by the liver, kidney, and other tissues in broilers. In conclusion, the adverse effects on broilers were induced when diets supplemented with Sn up to 480 mg/kg. Sn levels also managed to accumulate in the tibia and feather of broilers.

Histone H3 lysine(27)-to-methionine (H3K27M) gain-of-function mutations occur in highly aggressive pediatric gliomas. We established a Drosophila animal model for the pathogenic histone H3K27M mutation and show that its overexpression resembles polycomb repressive complex 2 (PRC2) loss-of-function phenotypes, causing derepression of PRC2 target genes and developmental perturbations. Similarly, an H3K9M mutant depletes H3K9 methylation levels and suppresses position-effect variegation in various Drosophila tissues. The histone H3K9 demethylase KDM3B/JHDM2 associates with H3K9M-containing nucleosomes, and its misregulation in Drosophila results in changes of H3K9 methylation levels and heterochromatic silencing defects. We have established histone lysine-to-methionine mutants as robust in vivo tools for inhibiting methylation pathways that also function as biochemical reagents for capturing site-specific histone-modifying enzymes, thus providing molecular insight into chromatin signaling pathways.

Whey acidic proteins (WAP) belong to a large gene family of antibacterial peptides that perform critical immune system functions. The function of human epididymis protein 4 (HE4), a 124-amino acid long polypeptide that has two whey acidic protein four-disulfide core (WFDC) domains, is not well studied. Here, a fusion gene encoding the HE4 protein fused to an IgG1 Fc domain was constructed. The recombinant HE4 protein was expressed as a secretory protein in Pichia pastoris and mammalian HEK293-F cells and was subsequently purified. Our data suggested that the HE4 protein produced by these two expression systems bound to both gram-negative and gram-positive bacteria, but demonstrated slightly inhibitory activity towards the growth of Staphylococcus aureus. Moreover, HE4 exhibited proteinase inhibitory activity towards trypsin, elastase, matrix metallopeptidase 9, and the secretory proteinases from Bacillus subtilis. The effects of glycosylation on the biochemical characterization of HE4 were also investigated. LC-ESI-MS glycosylation analysis showed that the high-mannose glycosylated form of HE4 expressed by P. pastoris has lower biological activity when compared to its complex-glycosylated form produced from HEK293-F cells. The implications of this are discussed, which may be provide theoretical basis for its important role in the development of cancer and innate immune system.

Given the ever-increasing burden of end-stage renal disease (ESRD) in a global milieu of limited financial and health resources, interested parties continue to search for ways to optimize dialysis access. Government and payer initiatives to increase access to renal replacement therapies (RRTs), particularly peritoneal dialysis (PD) and hemodialysis (HD), may have meaningful impacts from clinical and health-economic perspectives; and despite similar clinical and humanistic outcomes between the two dialysis modalities, PD may be the more convenient and resource-conscious option. This review assessed country-specific PD-First/Favored policies and their associated background, implementation, and outcomes. It was found that barriers to policy-implementation are broadly associated with government policy, economics, provider or healthcare professional education, modality-related factors, and patient-related factors. Notably, the success of a given country's PD-Favored policy was inversely associated with the extent of HD infrastructure. It is hoped that this review will provide a foundation across countries to share lessons learned during the development and implementation of PD-First/Favoredpolicies.

To improve the photocatalytic activity of a transition metal coordination polymer modified polyoxometalate (TMCP/POM), polypyrrole (PPy) was loaded on its surface through a facile in situ chemical oxidation polymerization process. Under the irradiation of visible light, the PPy loaded TMCP/POM composite material exhibited better photocatalytic activity than TMCP/POM, PPy and their mechanically blended products with respect to the degradation of Rhodamine B (RhB). Optical and electrochemical tests illustrated that the enhancement of photocatalytic activity can be attributed to the high separation efficiency of the photogenerated electron-hole pair on the interface of PPy and TMCP/POM, which originates from a synergy effect between them. Furthermore, the influence of reaction temperature on the morphology, wettability, conductivity and photocatalytic performance of the resulting composite material was discussed and an optical temperature to fabricate the photocatalyst with high efficiency has been obtained. These results suggest that the loading of PPy on the surface of TMCP/POM would be a feasible strategy to enhance its photocatalytic activity.

Long-term performance of capacitive deionization (CDI) and membrane-assisted capacitive deionization (MCDI) single cells equipped with the same pristine carbon xerogel (CX) electrodes configured as the anode and cathode was investigated. Unlike CDI, which was subject to performance degradation in a short period of time, MCDI showed performance preservation during the 50 h of operation due to its ability to mitigate charge leakage from parasitic electrochemical reactions that result in carbon oxidation. Differential capacitance measurements of the used CDI and MCDI electrodes revealed shifting of the potential of zero charge (EPZC) of the CDI anode from -0.1 to 0.4 V but only to 0.1 V for the MCDI anode. CDI and MCDI cells tested with electrodes having EPZCs at -0.1 and 0.5 V showed strongly contrasting results depending on the anode-cathode EPZC configuration. The MCDI cell configured with a 0.5 V EPZC cathode and -0.1 V EPZC anode displayed the best performance of all the tested cells, benefiting from increased counterion excess within the potential window, and the membrane was in-place to reject expelled co-ions from accessing the bulk.

Protein-protein interactions are the basis of biological functions, and studying these interactions on a molecular level is of crucial importance for understanding the functionality of a living cell. During the past decade, biosensors have emerged as an important tool for the high-throughput identification of proteins and their interactions. However, the high-throughput experimental methods for identifying PPIs are both time-consuming and expensive. On the other hand, high-throughput PPI data are often associated with high false-positive and high false-negative rates. Targeting at these problems, we propose a method for PPI detection by integrating biosensor-based PPI data with a novel computational model. This method was developed based on the algorithm of extreme learning machine combined with a novel representation of protein sequence descriptor. When performed on the large-scale human protein interaction dataset, the proposed method achieved 84.8% prediction accuracy with 84.08% sensitivity at the specificity of 85.53%. We conducted more extensive experiments to compare the proposed method with the state-of-the-art techniques, support vector machine. The achieved results demonstrate that our approach is very promising for detecting new PPIs, and it can be a helpful supplement for biosensor-based PPI data detection.

LD motifs (leucine-aspartic acid motifs) are short helical protein-protein interaction motifs that have emerged as key players in connecting cell adhesion with cell motility and survival. LD motifs are required for embryogenesis, wound healing and the evolution of multicellularity. LD motifs also play roles in disease, such as in cancer metastasis or viral infection. First described in the paxillin family of scaffolding proteins, LD motifs and similar acidic LXXLL interaction motifs have been discovered in several other proteins, whereas 16 proteins have been reported to contain LDBDs (LD motif-binding domains). Collectively, structural and functional analyses have revealed a surprising multivalency in LD motif interactions and a wide diversity in LDBD architectures. In the present review, we summarize the molecular basis for function, regulation and selectivity of LD motif interactions that has emerged from more than a decade of research. This overview highlights the intricate multi-level regulation and the inherently noisy and heterogeneous nature of signalling through short protein-protein interaction motifs.

Results from studies comparing health care resource use (HCRU), costs of treatment, and cost-effectiveness of linezolid compared with vancomycin therapy in the treatment of hospitalized patients with methicillin-resistant Staphylococcus aureus (MRSA) nosocomial pneumonia are limited in the published literature. We therefore conducted an analysis to compare the HCRU, costs of treatment, and cost-effectiveness of linezolid compared with vancomycin in the treatment of hospitalized patients with MRSA nosocomial pneumonia using data from a Phase IV clinical trial. The economic effect of moderate to severe adverse events (MSAEs) and the development of renal failure were also evaluated.

Chinese sturgeon, Acipenser sinensis, is a critically endangered anadromous fish species spawning in the Yangtze River of China during October and November. In this study, we analyzed the effects of hydrological factors, such as water temperature and discharge, on the natural reproduction time of the Chinese sturgeon and evaluated the impact of the impoundment of the Three Gorges Reservoir (TGR) based on our survey data from 1998 to 2011. The results showed that the first spawning dates were significantly (P < 0.05) correlated with the date of water temperature reaching 20°C (20°C WT dates), October mean discharge (Oct. discharge), and the discharge change from October to November (Oct-Nov ?discharge). Regression analysis suggested that one day delay of 20°C WT dates could postpone the first spawning date by 0.673 days. A discharge increase of 1000 m(3)s(-1) in October could bring forward the first spawning date by two days. Our results also indicated that the impoundment of the TGR had delayed the first spawning time due to water temperature lag and flow regulation. We suggest the following ecological regulations in order to facilitate conservation of the Chinese sturgeon: to eliminate water temperature lag by regulating water temperature downstream of the Three Gorges Dam (TGD), to enhance water discharge downstream in October, and to complete impoundment before October.

Usefulness of the Admission Shock Index for Predicting Short-Term Outcomes in Patients With ST-Segment Elevation Myocardial Infarction.

Current risk scores of ST-segment elevation myocardial infarction (STEMI) need sophisticated algorithm and were limited for bedside use. Our study aimed to evaluate the usefulness of admission shock index (SI) for predicting the short-term outcomes in patients with STEMI. Included were 7,187 consecutive patients with STEMI. The admission SI was defined as the ratio of admission heart rate and systolic blood pressure. Patients were divided into 2 groups with SI <0.7 and ?0.7, respectively, based on the receiver operating characteristic curve analysis. The major end points were 7- and 30-day all-cause mortality. Of 7,187 patients, 5,026 had admission SI <0.7 and 2,161 had admission SI ?0.7. Those who presented with SI ?0.7 had greater 7- and 30-day all-cause mortality and major adverse cardiovascular events than patients with SI <0.7. After multivariate adjustment, patients with SI ?0.7 had a 2.2-fold increased risk of 7-day all-cause mortality (hazard ratio 2.21, 95% confidence interval [CI] 1.71 to 2.86) and 1.9-fold increased risk of 30-day all-cause mortality (hazard ratio 1.94, 95% CI 1.54 to 2.44). Moreover, admission SI ?0.7 was also associated with 1.6- and 1.5-fold increased risk of 7- and 30-day major adverse cardiovascular events (hazard ratio 1.63, 95% CI 1.36 to 1.95 and hazard ratio 1.47, 95% CI 1.24 to 1.74, respectively). The C statistic of admission SI for predicting 7- and 30-day all-cause mortality was 0.701 and 0.686, respectively, compared with 0.744 and 0.738 from the Thrombolysis In Myocardial Infarction risk score. In conclusion, admission SI, an easily calculated index at first contact, may be a useful predictor for short-term outcomes especially for acute phase outcomes in patients with STEMI.

In this study, a three-dimensional finite element model was used to investigate the changes in tissue composition and mechanical signals within human lumbar intervertebral disc during the degenerative progression. This model was developed based on the cell-activity coupled mechano-electrochemical mixture theory. The disc degeneration was simulated by lowering nutrition levels at disc boundaries, and the temporal and spatial distributions of the fixed charge density, water content, fluid pressure, Von Mises stress, and disc deformation were analyzed. Results showed that fixed charge density, fluid pressure, and water content decreased significantly in the nucleus pulposus (NP) and the inner to middle annulus fibrosus (AF) regions of the degenerative disc. It was found that, with degenerative progression, the Von Mises stress (relative to that at healthy state) increased within the disc, with a larger increase in the outer AF region. Both the disc volume and height decreased with the degenerative progression. The predicted results of fluid pressure change in the NP were consistent with experimental findings in the literature. The knowledge of the variations of temporal and spatial distributions of composition and mechanical signals within the human IVDs provide a better understanding of the progression of disc degeneration.

A Gram-stain-negative, strictly aerobic, non-motile, cream, long rod-shaped bacterium, designated strain ZL136(T), was isolated from deep water of the South China Sea. Phylogenetic analysis based on 16S rRNA gene sequences indicated that this strain belonged to the genus Roseivivax with highest sequence similarity to Roseivivax halodurans OCh 239(T) (97.0%), followed by Roseivivax isoporae sw-2(T) (96.9%). Growth occurred at 4-37 °C (optimum 32 °C), pH 6.0-10.0 (optimum 8.0) and in the presence of 0-12% (w/v) NaCl (optimum 3-4%) in marine broth 2216. Strain ZL136(T) did not produce bacteriochlorophyll a. The predominant fatty acids were C(18?:?1)?7c and/or C(18?:?1)?6c, C(18?:?0), C(16?:?0) and 11-methyl C(18?:?1)?7c. The major polar lipids of ZL136(T) were phosphatidylcholine, phosphatidylethanolamine, phosphatidylglycerol, diphosphatidylglycerol and an unidentified lipid. The major respiratory quinone was ubiquinone Q-10. The genomic DNA G+C content of strain ZL136(T) was 67.0 mol%. On the basis of phenotypic, chemotaxonomic and phylogenetic analysis, strain ZL136(T) was classified as a representative of a novel species in the genus of Roseivivax, for which the name Roseivivax marinus sp. nov. is proposed. The type strain is ZL136(T) (?=?JCM 19386(T)?=?KCTC 32470(T)).

A strategy for scaffold-free self-assembly of multiple oligomeric enzymes was developed by exploiting enzyme oligomerization and protein-protein interaction properties, and was tested both in vitro and in vivo. Octameric leucine dehydrogenase and dimeric formate dehydrogenase were fused to a PDZ (PSD95/Dlg1/zo-1) domain and its ligand, respectively. The fusion proteins self-assembled into extended supramolecular interaction networks. Scanning-electron and atomic-force microscopy showed that the assemblies assumed two-dimensional layer-like structures. A fluorescence complementation assay indicated that the assemblies were localized to the poles of cells. Moreover, both in vitro and in vivo assemblies showed higher NAD(H) recycling efficiency and structural stability than did unassembled structures when applied to a coenzyme recycling system. This work provides a novel method for developing artificial multienzyme supramolecular devices and for compartmentalizing metabolic enzyme cascades in living cells.

To improve photocatalytic activity of a coordination polymer (CP) in the visible light region, five different transition metal ions (Fe(3+), Cr(3+), Ru(3+), Co(2+) and Ni(2+)) were introduced into its framework through an ion-exchange process. Among all the resulting transition metal ion doped coordination polymers (TMI/CPs), the one doped with Fe(3+) took on the most excellent photocatalytic activity and the highest quantum yields in the visible light region, decomposing 94% Rhodamine B (RhB) in 8 hours. It can be attributed to the doping of Fe(3+), which reduced the band gap (Eg) of the original CP, facilitating photocatalysis of the obtained polymer. Compared with the coordination polymer with Fe(3+) as a dopant, products doped with other metal ions presented weaker photocatalytic activities in the visible light region, while under the irradiation of ultraviolet light, they showed favorable photocatalytic properties. The results suggest that to dope transition metal ions into the framework of CPs would be an ideal option for enhancing the photocatalytic activity of coordination polymers.

The present study reports a rare case of large capillary hemangioma of the temporal bone with a dural tail sign. A 57-year-old female presented with pulsatile tinnitus and episodic vertigo associated with a ten-year history of an intermittent faint headache. Magnetic resonance imaging revealed a mass in the right petrous bone, which was hypointense on T1-weighted images and heterogeneously hyperintense on T2-weighted images, and showed a dural tail sign following gadolinium administration. Pre-operatively, this tumor was believed to be a meningioma. During surgery, the vascular tumor was removed by a modified pterional approach. A histopathological examination indicated that the tumor was a capillary hemangioma. Although intraosseous capillary hemangiomas are rare, they most frequently affect the temporal bone. Hemangiomas of the temporal bone may mimic other more common basal tumors. The diagnosis is most often made during surgical resection. The dural tail sign is not specific for meningioma, as it also occurs in other intracranial or extracranial tumors. The treatment of intratemporal hemangiomas is complete surgical excision, with radiotherapy used for unresectable lesions. To the best of our knowledge, the present study is the fourth case of intraosseous intracranial capillary hemangioma, but the largest intratemporal hemangioma to be reported in the literature to date.

Although several cross-sectional surveys have shown that certain traits such as extraversion and neuroticism are related to emotion regulation, few studies have explored the nature of this relationship. The present study tried to explore the longitudinal relation between traits and emotion regulation strategies. The Interpersonal Self-Support Scale for Middle School Students (ISSS-MSS) and the Emotion Regulation Questionnaire (ERQ) were administrated to 374 middle school students two times across a 6-month interval. A path analysis via structural equation modeling of the five interpersonal self-support traits and the two emotion regulation strategies was tested. The results showed that interpersonal independence predicted expressive suppression and cognitive reappraisal, and that interpersonal initiative also predicted reappraisal, while reappraisal predicted interpersonal flexibility and interpersonal openness 6 month later. These results support the hypotheses that some personality traits influence certain emotion regulation strategies, while other traits may be influenced by specific emotion regulation strategies.

Von Hippel-Lindau (VHL) disease is an autosomal dominant syndrome caused by germline mutations in the synonymous VHL gene encoding a tumor suppressor. Affected individuals are susceptible to various benign and malignant tumors. Based on the phenotypes, VHL disease is classified as type 1 and type 2. Here, we describe a Chinese family diagnosed as VHL disease type 2, with different metabolic status of tumors on FDG PET-CT. Genetic analysis revealed a germline c.264G > A point mutation, resulting in premature termination at codon 88 (p.W88X). This pedigree represents a rare link between p.W88X nonsense mutation (genotype) and VHL disease type 2 (phenotype), which has not been previously described. This is also the first nonsense mutation to manifest as VHL disease type 2 in ethnic Chinese. We also reviewed the literature and provided an outline of mutations associated with VHL disease in China.

It is challenging to noninvasively determine the mechanical properties of biological soft tissues in vivo. In this study, based on the biphasic theory and the transport models, a new constitutive model for hydration-dependent mechanical properties in hydrated soft materials was derived: HA = An(1-f?)(f?)2-n/2(2-f?), where HA(=?+2 ?) is the aggregate modulus, ?(f) is the volume fraction of fluid (i.e., hydration), A and n (>2) are two parameters related to the transport properties of the biphasic materials. A linear model for hydration-dependent shear modulus in the literature was verified for hydrogels. The effects of tissue hydration on mechanical properties (aggregate modulus and Poisson's ratio) were investigated. It was found that the value of Poisson's ratio was very sensitive to the tissue hydration in soft materials with high water content. The predictions of the aggregate modulus and shear modulus for hydrogels by the model compared well with those from experimental results. This study is important for developing new techniques for noninvasively assessing the mechanical properties of biological soft tissues using quantitative MRI methods as well as for designing scaffolds with proper mechanical properties for tissue engineering applications.

Immobilization of enzymes onto nanoparticles and retention of their structure and activity, which may be related to the orientation of enzymes on nanoparticles, remain a challenge. Here, we developed a novel enzyme-orientation strategy to enhance the activity of formate dehydrogenase immobilized on polydopamine-coated iron oxide nanoparticles via site-directed mutation. Seven mutants were constructed based on homology modeling of formate dehydrogenase and immobilized on polydopamine-coated iron oxide nanoparticles to investigate the influence of these mutations on immobilization. The immobilized mutant C242A/C275V/C363V/K389C demonstrated the highest immobilization yield and retained 90% of its initial activity, which was about 3-fold higher than that of wild-type formate dehydrogenase. Moreover, co-immobilization of formate dehydrogenase and leucine dehydrogenase was performed for the synthesis of l-tert-leucine. The catalytic efficiency of the co-immobilized mutant C242A/C275V/C363V/K389C and leucine dehydrogenase increased by more than 4-fold compared to that of co-immobilized wild-type formate dehydrogenase and leucine dehydrogenase.

Despite significant advances in automated nuclear magnetic resonance-based protein structure determination, the high numbers of false positives and false negatives among the peaks selected by fully automated methods remain a problem. These false positives and negatives impair the performance of resonance assignment methods. One of the main reasons for this problem is that the computational research community often considers peak picking and resonance assignment to be two separate problems, whereas spectroscopists use expert knowledge to pick peaks and assign their resonances at the same time. We propose a novel framework that simultaneously conducts slice picking and spin system forming, an essential step in resonance assignment. Our framework then employs a genetic algorithm, directed by both connectivity information and amino acid typing information from the spin systems, to assign the spin systems to residues. The inputs to our framework can be as few as two commonly used spectra, i.e., CBCA(CO)NH and HNCACB. Different from the existing peak picking and resonance assignment methods that treat peaks as the units, our method is based on 'slices', which are one-dimensional vectors in three-dimensional spectra that correspond to certain ([Formula: see text]) values. Experimental results on both benchmark simulated data sets and four real protein data sets demonstrate that our method significantly outperforms the state-of-the-art methods while using a less number of spectra than those methods. Ovarian cancer (OC) is the third most common type of gynecological cancer. Measurements of human epididymis protein 4 (HE4) levels have been suggested for improving the specificity of the laboratory identification of OC. For this meta-analysis, the Medline, Embase and Cochrane databases were searched to identify relevant studies. All the included studies for diagnostic performance were combined with sensitivity, specificity, positive likelihood ratio, negative likelihood ratio, diagnostic odds ratios (DORs) with 95% confidence intervals (CIs), summary receiver operating characteristic (SROC) curves and areas under the SROC curves (AUC). A total of 25 studies including 4,729 patients were identified as eligible for inclusion in the final analysis. The pooled sensitivities and respective 95% CIs for HE4 and carbohydrate antigen 125 (CA125) were 0.74 (0.72-0.76) and 0.74 (0.72-0.76), respectively. The pooled specificities and respective 95% CIs for HE4 and CA125 were 0.90 (0.89-0.91) and 0.83 (0.81-0.84), respectively. The summary DORs and 95% CIs for HE4 and CA125 were 43.35 (29.13-64.51) and 17.06 (10.97-26.51), respectively and the AUCs for HE4 and CA125 were 0.8915 and 0.8538, respectively. In total, 9 studies investigated the diagnostic accuracy of HE4 combined with CA125 for the diagnosis of OC. The pooled sensitivity and 95% CIs of HE4, CA125 and HE4+CA125 in this subgroup were 0.71 (0.67-0.75), 0.74 (0.69-0.78) and 0.90 (0.87-0.92), respectively; the pooled specificity and 95% CIs of HE4, CA125 and HE4+CA125 were 0.92 (0.90-0.94), 0.73 (0.69-0.76) and 0.85 (0.82-0.87), respectively. The diagnostic accuracy of HE4 in distinguishing OC from other benign gynecological diseases was found to be to be superior to that of CA125 and the combination of HE4 and CA125 may enhance the diagnostic sensitivity.

Morbidity and mortality of prostate cancer (PCa) have increased in recent years worldwide. Currently existing methods for diagnosis and treatment do not make the situation improve, especially for hormone refractory prostate cancer (HRPC). The lack of molecular probes for PCa hindered the early diagnosis of metastasis and accurate staging for PCa. In this work, we have developed a new aptamer probe Wy-5a against PCa cell line PC-3 by cell-SELEX technique. Wy-5a shows high specificity to the target cells with dissociation constants in the nanomolar range, and does not recognize other tested PCa cell lines and other tested tumor cell lines. The staining of clinical tissue sections with fluorescent dye labeled Wy-5a shows that sections from high risk group with metastasis exhibited stronger fluorescence and sections from Benign Prostatic Hyperplasia (BPH) did not exhibit notable fluorescence, which suggests that aptamer Wy-5a may bind to protein related to the progression of PCa. The high affinity and specificity of Wy-5a makes this aptamer hold potential for application in diagnosis and target therapy of PCa.

Summary: Objective: To compare associations of lipid accumulation product (LAP) and body mass index (BMI) with risk of hypertension.Methods: Demographic characteristics, blood pressure and body measurements were obtained, blood lipids and fast plasma glucose were examined, BMI and LAP were calculated for 2589 Mongolians. Multiple linear model and covariance analysis was used to analyze the relationship between LAP and blood pressure levels, multivariate logistic analysis was used to analyze the association between BMI and LAP and hypertension.Results: LAP and BMI were all significantly higher in hypertensives than in normotensives in both genders. Multivariate adjusted mean levels of SBP and DBP increased with increased LAP and there was a linear association between LAP and blood pressure. LAP had a stronger association with risk of hypertension compared with BMI, linear trend between LAP and risks of hypertension was better than that between BMI and risks of hypertension in males. Compared to the first quartile, the odds ratios [95% confidence intervals (95% CIs)] of hypertension associated with the second, third and fourth quartiles of LAP were 1.85(1.23,2.79), 2.20(1.47,3.28), 4.21(2.78,6.38) in males; compared to the first quartile, the odds ratios (95% CIs) of hypertension associated with the second, third and fourth quartiles of BMI were 0.93(0.62,1.38), 1.23(0.83,1.84), 2.80(1.86,4.21) in males.Conclusion: LAP was more closely associated with risk of hypertension than BMI in Mongolian males, this study suggested that LAP might be a preferred anthropometry measurement to predict risk of hypertension in males. Future prospective cohort studies should be conducted to test the causal relationship between LAP and the risk of hypertension in Mongolian population.

The generation of hematopoietic stem cells (HSCs) from hemogenic endothelium within the aorta, gonad, mesonephros (AGM) region of the mammalian embryo is crucial for development of the adult hematopoietic system. We described a deletion of a Gata2 cis-element (+9.5) that depletes fetal liver HSCs, is lethal at E13-14 of embryogenesis, and is mutated in an immunodeficiency that progresses to myelodysplasia/leukemia. Here, we demonstrate that the +9.5 element enhances Gata2 expression and is required to generate long-term repopulating HSCs in the AGM. Deletion of the +9.5 element abrogated the capacity of hemogenic endothelium to generate HSC-containing clusters in the aorta. Genomic analyses indicated that the +9.5 element regulated a rich ensemble of genes that control hemogenic endothelium and HSCs, as well as genes not implicated in hematopoiesis. These results reveal a mechanism that controls stem cell emergence from hemogenic endothelium to establish the adult hematopoietic system.

The neuronal sortilin-related receptor (SORL1) has been reported to modulate the risk of Alzheimers disease (AD) in a variety of populations, but replication studies have been inconsistent. Amnestic mild cognitive impairment (aMCI) is characterized by episodic memory impairment and represents the prodromal stage of AD. However, the relationship between SORL1 and aMCI remains unclear. This study aimed to investigate the relationship between SORL1 genetic variation and aMCI in the Han Chinese population. We conducted a case-control study using a single-nucleotide polymorphism (SNP), rs668387 (SNP8), in the 5 region of SORL1, and three SNPs [rs2070045 (SNP19), rs3824968 (SNP23), rs2282649 (SNP24)] in the 3 region of SORL1, along with a haplotype analysis, in 139 aMCI patients and 213 cognitively-healthy controls from a northern Han Chinese population. We observed that SNP19 had a significantly different allele frequency between aMCI patients and controls (p = 0.006). Moreover, the GAT haplotype at SNPs 19-23-24 was associated with an increased risk of aMCI [odds ratio (OR) 1.377], while the TTC haplotype at SNPs 19-23-24 was associated with a decreased risk (OR 0.708). These results indicated that the SNPs in the 3 region of SORL1 are associated with aMCI in northern Han Chinese.

A further understanding of the structure of lignin from herbaceous crops is needed for advancing technologies of lignocellulosic biomass processing and utilization. A method was established in this study for analyzing structural motifs found in milled straw lignin (MSL) and cellulase-digested lignin (CEL) isolated from wheat straw by combining quantitative (13)C and HSQC NMR spectral analyses. The results showed that guaiacyl (G) was the predominant unit in wheat straw cell wall lignin over syringyl (S) and hydroxyphenyl (H) units. Up to 8.0 units of tricin were also detected in wheat straw lignin per 100 aromatic rings. Various interunit linkages, including ?-O-4, ?-5, ?-?, ?-1, ?, ?-diaryl ether, and 5-5/4-O-? as well as potential lignin-carbohydrate complex (LCC) bonds, were identified and quantified. These findings provide useful information for the development of biofuels and lignin-based materials.

Histone H3 lysine 4 (H3K4) can be mono-, di-, and trimethylated by members of the COMPASS (complex of proteins associated with Set1) family from Saccharomyces cerevisiae to humans, and these modifications can be found at distinct regions of the genome. Monomethylation of histone H3K4 (H3K4me1) is relatively more enriched at metazoan enhancer regions compared to trimethylated histone H3K4 (H3K4me3), which is enriched at transcription start sites in all eukaryotes. Our recent studies of Drosophila melanogaster demonstrated that the Trithorax-related (Trr) branch of the COMPASS family regulates enhancer activity and is responsible for the implementation of H3K4me1 at these regions. There are six COMPASS family members in mammals, two of which, MLL3 (GeneID 58508) and MLL4 (GeneID 8085), are most closely related to Drosophila Trr. Here, we use chromatin immunoprecipitation-sequencing (ChIP-seq) of this class of COMPASS family members in both human HCT116 cells and mouse embryonic stem cells and find that MLL4 is preferentially found at enhancer regions. MLL3 and MLL4 are frequently mutated in cancer, and indeed, the widely used HCT116 cancer cell line contains inactivating mutations in the MLL3 gene. Using HCT116 cells in which MLL4 has also been knocked out, we demonstrate that MLL3 and MLL4 are major regulators of H3K4me1 in these cells, with the greatest loss of monomethylation at enhancer regions. Moreover, we find a redundant role between Mll3 (GeneID 231051) and Mll4 (GeneID 381022) in enhancer H3K4 monomethylation in mouse embryonic fibroblast (MEF) cells. These findings suggest that mammalian MLL3 and MLL4 function in the regulation of enhancer activity and that mutations of MLL3 and MLL4 that are found in cancers could exert their properties through malfunction of these Trr/MLL3/MLL4-specific (Trrific) enhancers.

The precise three-dimensional (3-D) segmentation of airway from CT image is essential for the image-guided therapy, which helps avoid a serious airway injury. We proposed a new segmentation algorithm for the calculation. Firstly, region growing method was employed to segment the main bronchioles (rough segmentation). And then region growing driven by morphology dilation was used to expand the airway region, where centerline of airway tree was extracted. Terminal bronchia (fine segmentation) were segmented along the centerline. Ultimately, rough and fine segmentation results are combined by a logical OR as final airway tree. Quantitative comparisons with 6 sets of manual segmentation results showed that the algorithm could be used to segment up to 9 bronchia, and the average branch sensitivity of 6th was 63.5%, meeting the requirement of airway tree in the image-guided therapy.

We conducted a genome-wide association study meta-analysis of mean arterial pressure and pulse pressure among 26,600 East Asian participants (stage 1) followed by replication study of up to 28,783 participants (stage 2). For novel loci, statistical significance was determined by a P<5.0×10(-8) in joint analysis of stage 1 and stage 2 data. For loci reported by the previous mean arterial and pulse pressure genome-wide association study meta-analysis in Europeans, evidence of transethnic replication was determined by consistency in effect direction and a Bonferroni-corrected P<1.4×10(-3). No novel loci were identified by the current study. Five independent mean arterial pressure variants demonstrated robust evidence for transethnic replication including rs17249754 at ATP2B1 (P=7.5×10(-15)), rs2681492 at ATP2B1 (P=3.4×10(-7)), rs11191593 at NT5C2 (1.1×10(-6)), rs3824755 at CYP17A1 (P=1.2×10(-6)), and rs13149993 at FGF5 (P=2.4×10(-4)). Two additional variants showed suggestive evidence of transethnic replication (consistency in effect direction and P<0.05), including rs319690 at MAP4 (P=0.014) and rs1173771 at NPR3 (P=0.018). For pulse pressure, robust evidence of replication was identified for 2 independent variants, including rs17249754 at ATP2B1 (P=1.2×10(-5)) and rs11191593 at NT5C2 (P=1.1×10(-3)), with suggestive evidence of replication among an additional 2 variants including rs3824755 at CYP17A1 (P=6.1×10(-3)) and rs2681492 at ATP2B1 (P=9.0×10(-3)). Replicated variants demonstrated consistency in effect sizes between East Asian and European samples, with effect size differences ranging from 0.03 to 0.24 mm?Hg for mean arterial pressure and from 0.03 to 0.21 mm?Hg for pulse pressure. In conclusion, we present the first evidence of transethnic replication of several mean arterial and pulse pressure loci in an East Asian population.

In this work, ultra-performance LC with ESI quadrupole TOF-MS (UPLC-ESI-Q-TOF-MS) and automated MetaboLynx analysis was used to rapidly separate and identify the chemical constituents of Danggui San, a traditional Chinese medical formula. The analysis was performed on a Waters UPLC BEH C18 column using a gradient elution system. A hyphenated ESI and Q-TOF analyzer was used for the determination of the accurate mass of the protonated or deprotonated molecule and fragment ions in both positive and negative modes. Based on retention times, accurate mass, and the mass spectrometric fragmentation characteristics, a total of 47 compounds distributed over the chemical groups of phthalides, flavonoids, monoterpene glycosides, sesquiterpenoids, phenolics, and alkaloids, were simultaneously separated within 18 min and identified or tentatively elucidated in Danggui San for the first time. UPLC-ESI-Q-TOF-MS analysis revealed the complexity of the chemical composition of this formula. The method developed is rapid, accurate, reliable, and highly sensitive to characterize the chemical constituents of Danggui San.

The aim of this study is to identify rat nasal septum respiratory mucosa-derived mesenchyme stem cells (RM-MSCs) and to compare its neural lineage differentiation capacity with bone marrow-derived mesenchyme stem cells (BM-MSCs) after a short period of neural induction culture in vitro. The cell morphology was observed with light microscopy; cell proliferation was assessed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT). The characteristics of the cells were evaluated with flow cytometry, immunofluorescence, real-time quantitative PCR (RT-PCR), and Western blotting. The results showed that rat nasal respiratory mucosa contains RM-MSCs that exhibited similar proliferation rate as BM-MSCs in vitro. Both RT-PCR and Western blotting analyses demonstrated that RM-MSCs showed higher expression of neural lineage markers than BM-MSCs after a short period of neural induction culture, and secreted higher level of brain-derived neurotrophic factor. RM-MSCs were more amenable to differentiate into neural or glial cell after a short period of neural induction culture than BM-MSCs in vitro; and it could be considered as another optimal source of stem cells for cell-based therapy to neurological diseases.

Hypoxia, a critical regulator of tumor growth and metastasis, induces the transcriptional activation of several pathways involved in proliferation, migration and invasion. Gankyrin was found to be overexpressed, and also promoted the metastasis in breast cancer cells, which is also involved in the regulation of hypoxia inducible factor?1 and hypoxia?inducible factor?1?. The present study showed that gankyrin mRNA and protein expression were increased under hypoxic conditions in the BT474 breast cancer cell line, accompanied with increased ability of cell migration and invasion. Lentivirus?mediated siRNA targeting gankyrin was transfected into BT474 cells. Wound?healing and transwell experiments showed that gankyrin deletion abrogated the increased migration and invasion of BT474 cells due to hypoxia. In addition, E?cadherin was found to be involved in the gankyrin induced invasion of breast cancer cells due to hypoxia. The present study indicated that gankyrin deletion abrogated the increased metastatic potential of breast cancer cells under hypoxic conditions partly through regulating E?cadherin, suggesting that an improved understanding of gankyrin may offer a potential therapeutic target for the treatment of human breast cancer metastasis.

In developing our video relationships, we compare around 5 million PubMed articles to our library of over 4,500 methods videos. In some cases the language used in the PubMed abstracts makes matching that content to a JoVE video difficult. In other cases, there happens not to be any content in our video library that is relevant to the topic of a given abstract. In these cases, our algorithms are trying their best to display videos with relevant content, which can sometimes result in matched videos with only a slight relation.
