In the present study, the three-dimensional fluorescence spectra of River A with great flow rate were investigated. The results showed that there existed three unambiguous peaks in the excitation-emission matrix of River A at lambda(ex)/lambda(em) of around 230/340, 280/320 and 250/450 nm respectively. The fluorescence intensity varied significantly and had sharp fluctuation sometimes. But the COD(Mn) of the samples remained quite stable. This study indicated that fluorescence technique could demonstrate the pollution in the water bodies with great flow rate and furthermore make up for the deficiency of the conventional parameters related to organic pollution, i. e. invalidation to exhibit the components of pollutants. It is a good tool for the early-warning of the water quality.

Classical swine fever virus (CSFV) has a tropism for vascular endothelial cells and immune system cells. The process and release of pro-inflammatory cytokines, including IL-1? and IL-18, is one of the fundamental reactions of the innate immune response to viral infection. In this study, we investigated the production of IL-1? from macrophages following CSFV infection. Our results showed that IL-1? was upregulated after CSFV infection through activating caspase-1. Subsequent studies demonstrated that reactive oxygen species may not be involved in CSFV-mediated IL-1? release. Recently, research has indicated a novel mechanism by which inflammasomes are triggered through detection of activity of viroporin. We further demonstrated that CSFV viroporin p7 protein induced IL-1? secretion which could be inhibited by the ion channel blocker amantadine and also discovered that p7 protein was a short-lived protein degraded by the proteasome. Together, our observations provided an insight into the mechanism of CSFV-induced inflammatory responses.

Reported herein is a series of Ru2(Xap)4 capped polyyn-diyl compounds, where Xap is either 2-anilinopyridinate (ap) or its aniline substituted derivatives. Symmetric [Ru2(Xap)4](?-C4k)[Ru2(Xap)4] (compounds 4ka (X = 3-isobutoxy) and 4kc (X = 3,5-dimethoxy) with k = 2, 3, 4, and 5) was obtained from the Glaser coupling reaction of Ru2(Xap)4(C2kH). Unsymmetric [Ru2(Xap)4](?-C(4k+2))[Ru2(ap)4] (compounds 4k+2b with k = 2, 3, and 4) were prepared from the Glaser coupling reaction between Ru2(Xap)4(C(2k+2)H) and Ru2(ap)4(C2kH). X-ray diffraction study of compound 12c revealed both the sigmoidal topology of the polyyn-diyl bridge and the fine structural detail about the Ru2 cores. Cyclic and differential pulse voltammetric (CV and DPV) measurements and spectroelectrochemical studies revealed that (i) the reduced monoanions [Ru2-C2m-Ru2](-1) (m = 4-8) belong to the Robin-Day class II mixed valent ions and (ii) the electronic coupling between Ru2 termini depends on the length of the polyyn-diyl bridge with an attenuation constant (?) between 0.12 and 0.15 Å(-1). In addition, spin-unrestricted DFT calculations provide insight about the nature of orbitals that mediate the long distance electronic coupling.

Sparse representation classification (SRC) is one of the most promising classification methods for supervised learning. This method can effectively exploit discriminating information by introducing a [Symbol: see text]1 regularization terms to the data. With the desirable property of sparisty, SRC is robust to both noise and outliers. In this study, we propose a weighted meta-sample based non-parametric sparse representation classification method for the accurate identification of tumor subtype. The proposed method includes three steps. First, we extract the weighted meta-samples for each sub class from raw data, and the rationality of the weighting strategy is proven mathematically. Second, sparse representation coefficients can be obtained by [Symnbol: see text]1 regularization of underdetermined linear equations. Thus, data dependent sparsity can be adaptively tuned. A simple characteristic function is eventually utilized to achieve classification. Asymptotic time complexity analysis is applied to our method. Compared with some state-of-the-art classifiers, the proposed method has lower time complexity and more flexibility. Experiments on eight samples of publicly available gene expression profile data show the effectiveness of the proposed method.

Fatal anaphylactic shock is common in forensic practice. However, it is difficult to diagnose for lacking specific pathological and morphologic changes in forensic autopsy. The application of some biochemical indicators is of great significance. This paper reviews the biological characteristics of some biochemical indicators and detection methods. The forensic application, problems and prospects of these indicators are also introduced in details. The stable biochemical indicators, IgE, tryptase and chymase, show great potential and advantages in the identification of fatal anaphylactic shock in forensic medicine.

The self-assembly of Ru2(CO3)4(3-) paddle-wheel precursors and Mn(2+) ions in aqueous solution yields various carbonate complexes. With appropriate selection of the synthetic conditions, we are able to intentionally tune the composition and structure of Mn-Ru2-carbonate assemblies to form infinite chain structural complexes, e.g., K[{Mn(H2O)4}2Ru2(CO3)4Br2]·H2O (1) and H[{Mn(H2O)4}2Ru2(CO3)4Br2]·6H2O (2). Complexes 1 and 2 are obtained at different temperatures (25 °C for 1 and 5 °C for 2, respectively), and their crystal structures consist of brick-wall stacked chains, in which neighboring Ru2(CO3)4Br2(5-) units are linked by two disubstituted octahedral Mn(H2O)4(2+) in a cis manner, resulting in two isomeric (twisted and zigzag) negative double-chain ?- and ?-{[Mn(H2O)4]2Ru2(CO3)4Br2}n(n-). The magnetic properties of complexes 1 and 2 were highly characterized. The alternating current (AC) susceptibility analysis of complex 1 reveals a two-step magnetism transition at T1 = 5.0 K and T2 = 2.6 K, respectively. Complex 2 exhibits metamagnetism behavior, with a transition field H(C) = ~1.2 kOe at 2.0 K.

Jiedu Xiaozheng Yin (JXY) is a Chinese herbal decoction used to treat hepatocellular carcinoma (HCC). Previous studies have demonstrated that JXY can inhibit HCC cell proliferation via induction of G0/G1 phase arrest. In this study, we investigated whether the inhibitory effect of JXY on HCC cells is associated with the inhibition of the Wnt/??catenin pathway and the polycomb gene product Bmi1. Ethyl acetate extract from JXY (EE-JXY) was prepared. Methyl thiazolyl tetrazolium (MTT) and colony formation assays were used to measure cell proliferation. Immunofluorescence was used to analyze the expression and location of ?-catenin and Bmi1. Immunohistochemistry was used to examine the expression of proliferating cell nuclear antigen (PCNA), c-myc and cyclin D1. ?-catenin, Bmi1, c-myc, cyclin D1 and p16INK4A mRNA levels were detected by RT-PCR. The results demonstrated that EE-JXY inhibited the expression of PCNA, c-myc, cyclin D1 and Bmi1, and upregulated the expression of p16INK4A. We also found that EE-JXY could facilitate ?-catenin translocation from the cytoplasm and nuclei to the cytomembrane. Finally, suppression of cell proliferation and expression of Bmi1 and Wnt/?-catenin by EE-JXY was confirmed in a mouse xenograft model of HCC. Thus, EE-JXY can inhibit the proliferation of HCC partially via suppression of the Bmi1 and Wnt/?-catenin signaling pathways.

Association between APE1 T1349G polymorphism and prostate cancer risk: evidence from a meta-analysis.

APE1 T1349G polymorphism was considered to be associated with risk of cancer, but studies on the association between APE1 T1349G polymorphism and risk of prostate cancer remained inconclusive. A meta-analysis of published studies was performed to precisely assess the association between APE1 Asp148Glu polymorphism and prostate cancer risk. PubMed, Embase, and Wanfang databases were searched for published case-control studies investigating the association between APE1 T1349G polymorphism and prostate cancer risk. Odds ratios (ORs) and 95 % confidence intervals (95 % CIs) were used to assess the strength of the association. Overall, seven studies with a total of 3,063 individuals were finally included into the meta-analysis. The heterogeneity analysis did not find obvious heterogeneity among those included studies. Meta-analysis of total seven studies did not find an obvious association between APE1 T1349G polymorphism and prostate cancer risk (G vs T OR (95 % CI)?=?1.11 (0.99-1.24); GG vs TT OR (95 % CI)?=?1.25 (0.96-1.62); TG vs TT OR (95 % CI)?=?1.11 (0.95-1.30); GG/TG vs T OR (95 % CI)?=?1.13 (0.97-1.32); GG vs TT/TG OR (95 % CI)?=?1.16 (0.91-1.48)). Subgroup analyses by ethnicity showed that APE1 T1349G polymorphism was associated with increased risk of prostate cancer in Caucasians (G vs T OR (95 % CI)?=?1.26 (1.02-1.56), P?=?0.033; TG vs TT OR (95 % CI)?=?1.44 (1.06-1.94), P?=?0.019; GG/TG vs T OR (95 % CI)?=?1.45 (1.08-1.94), P?=?0.013). The meta-analysis suggests that APE1 T1349G polymorphism is associated with increased risk of prostate cancer, especially in Caucasians. More studies are needed to further identify the obvious association above.

In filamentous fungi, the pathogenic mitogen-activated protein kinase (PMK) pathway performs an important function in plant infection. STE12-like genes found in higher eukaryotes encode transcription factors and are regulated by the PMK pathway. However, the functions of STE12-like genes in foliar pathogens remain poorly understood. In this study, we cloned StSTE12 from Setosphaeria turcica and investigated its functions by RNA interference. Transformants ste12-3, ste12-2 and, ste12-1, in which the StSTE12 silencing efficiency increased in order, were confirmed by real time PCR. Compared with the wild-type (WT) strain, the transformants showed reduced growth rate, lighter colony color, and obviously decreased conidium production. More importantly, different to WT strain and ste12-3 with lower StSTE12silencing efficiency, ste12-1 and ste12-2 with higher StSTE12 silencing efficiency were nonpathogenic on intact leaves, but pathogenic on wounded leaves. However, the biological activity of HT-toxin from all transformants showed no difference on corn leaves. Furthermore, ste12-1 and ste12-2 did not penetrate artificial cellophane membrane and showed abnormal and delayed development appressoria. Although it could penetrate the cellophane membranes, ste12-3 formed appressoria after 48 h of inoculation more than WT. Therefore, StSTE12 was involved in vegetative growth, conidiation, appressorial development, penetration as well as the pathogenicity, but it was not related to HT-toxin biosynthesis. More interestingly, all the results suggested that StSTE12 was crucial for pathogenicity due to involvement in regulating appressoria development and penetration.

Fuzheng Qingjie (FZQJ) recipe is a polyherbal Chinese medicine capable of suppressing tumor growth and is used as an adjuvant therapy for various types of cancer. However, its anticancer mechanisms are yet to be fully elucidated. In the present study, we explored whether p38 mitogen-activated protein kinase (MAPK) was involved in FZQJ-mediated mitochondria-dependent apoptosis in human hepatocellular carcinoma cells. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assays were used to measure the viability of HepG2 cells. 4,6-Diamidino-2-phenylindole (DAPI) and Annexin-V fluorescein isothiocyanate (FITC) were used to analyze the apoptosis of HepG2 cells. The mitochondrial membrane potential (??) and phosphorylated P38 MAPK protein were examined by a flow cytometer following 5,5',6,6'-tetrachloro?1,1',3,3'-tetraethylbenzimidazolcarbocyanine iodide (JC-1) and Alexa Fluor® 647 mouse anti-phosphorylated P38 MAPK antibody staining, respectively. The activation of caspase-9 and caspase-3 were measured using colorimetric assays. Additionally, Bcl-2 and Bax expression were examined using reverse transcription polymerase chain reaction (RT-PCR) and western blot analysis. The results demonstrated that water extract of FZQJ was able to induce apoptosis of HepG2 cells in vitro. FZQJ-induced apoptosis was accompanied by the loss of ??, downregulation of Bcl-2 and upregulation of Bax expression, and the activation of caspase-3, -9 and P38 MAPK. These results indicated that FZQJ induced apoptosis in HepG2 cells at least via P38 MAPK activation and the mitochondria-dependent apoptotic pathway.

Glucose-regulated protein (GRP) 78 and C/-EBP homologous protein (CHOP) are commonly used as markers of endoplasmic reticulum (ER) stress. As an ER chaperone, GRP78 functions as a potent anti-apoptotic factor and confers drug resistance, whereas CHOP is a key initiating factor of ER stress-related cell death. We aimed at investigating the predictive values of GRP78 and CHOP in breast cancer patients who underwent adjuvant chemotherapy. An immunohistochemistry screen for GRP78 and CHOP was performed using a tissue microarray containing 250 tumors from female patients diagnosed with invasive ductal breast carcinoma at the Fudan University Shanghai Cancer Center. The staining results were scored semi-quantitatively, and a prediction model was constructed to verify the hypothesis. In this retrospective cohort study, CHOP correlated with prolonged disease-free survival (HR = 0.385, 95 % CI 0.215-0.688; P = 0.001), whereas GRP78 showed an opposite association (HR = 4.573; 95 % CI 2.291-9.128; P < 0.001). Moreover, in a GRP78-positive subset, CHOP overexpression correlated with a lower risk of recurrence. In the receiver operating characteristic analysis, the prediction capability of the predictive model combining the above two markers surpassed that of the traditional model (P = 0.0085 for the area under the curve comparison). Within the anthracycline-treatment subgroup, the combined GRP78 and CHOP exhibited similar predictive significance. Cumulatively, our findings suggest a tight association between ER stress markers and clinical outcomes for patients with breast cancer.

A new water-soluble glycocluster based on perylene bisimides PBI-12-Man has been designed and synthesized, and its specific and selective binding property with Concanavalin A (Con A) has been investigated by fluorescence spectroscopy and circular dichroism (CD) spectroscopy, which showed strong binding affinity for Con A with the binding constant of 8.2×10(5)M(-1) for monomeric mannose unit, two orders of magnitude higher than the corresponding monosaccharide ligand. Most interestingly, a fluorescence enhancement of PBI-12-Man was observed upon binding with Con A because of deaggregation of the self-assembly of PBI-12-Man induced by carbohydrate-protein interaction, and the further study of the fluorescence enhancement with macrophage cells showed that PBI-12-Man as a biocompatible agent had fluorescence imaging of the surface mannose receptor of the cells. Such fluorescence turn-on sensing of protein based on carbohydrate-protein interactions would facilitate the development of new protein-specific fluorescent probe for diagnosis and molecular imaging under live cell conditions.

This review describes the history of taxonomic research on scorpions and provides an updated checklist and key of the scorpions currently known in China. This checklist is based on a thorough review of the extant literatures on scorpion species whose presence has been confirmed in China through field expeditions and examination of scorpion collections, excepting a few members that have no clear distribution or are currently in doubt. Totally, the scorpion fauna of China consists of 53 species and subspecies belonging to 12 genera crossing five families, with 33 species (62.3%) and one genus being recorded as endemic. Additionally, identification key and the distribution of scorpions from China are provided.

Schistosomiasis japonica, caused by Schistosoma japonicum infection, remains a major public health concern in China, and the geographical distribution of this neglected tropical disease is limited to regions where Oncomelania hupensis, the intermediate host of the causative parasite, is detected. The purpose of this study was to monitor the transmission of S. japonicum in potential risk regions of China during the period from 2008 through 2012. To monitor the transmission, 10 fixed surveillance sites and 30 mobile sentinel sites were selected in 10 counties of four provinces, namely Anhui, Jiangsu, Chongqing and Hubei. There were 8, 9, 6, 2 and 3 cases infected with S. japonicum detected in the 30 mobile sentinel sites during the 5-year study period, while 27 subjects were positive for the antibody-based serum test in the 10 fixed sentinel sites; however, no infection was found. In addition, neither local nor imported livestock were found to be infected. No O. hupensis snails were detected in either the fixed surveillance or the mobile sentinel sites; however, the snail host was found to survive and reproduce at Chaohu Lake, inferring the potential of transmission of the disease. It is suggested that the continuous surveillance of schistosomiasis japonica should be carried out in both the endemic foci and potential risk regions of China, and an active, sensitive system to respond the potential risk of transmission seems justified.

A highly efficient demulsifying strain, LH-6, was isolated from petroleum-contaminated soil and identified as Bacillus cereus by 16S rDNA gene analysis. It achieved 95.61 and 95.40 % demulsifying ratios within 12 h for water-in-oil (W/O) and oil-in-water (O/W) model emulsions, respectively. Fourier transform infrared spectroscopy (FT-IR) and thin-layer chromatography (TLC) detections indicated that the LH-6's extracellular biodemulsifiers were different types of lipopeptides for the W/O and O/W emulsions. Optimization of the culture medium composition was conducted to improve the biosynthesis and demulsifying efficiency of the biodemulsifier. The optimal carbon source was liquid paraffin, while waste frying oil could also be an alternative carbon source. The optimal nitrogen sources were ammonium sulfate and yeast extract. To further enhance the biodemulsifier efficiency, the optimal cultivation conditions were determined using response surface methodology (RSM) based on central composite rotation design (CCRD). Using the optimized cultivation conditions, the demulsifying ratios increased to 98.23 and 97.65 % for the W/O and O/W model emulsions, respectively.

A novel water-soluble fluorescent glycodendrimer based on perylene bisimides is synthesized, which exhibits high fluorescence quantum yield of 54%. While the binding interactions of PBI-Man with Concanavalin A (Con A) are studied by fluorescence spectra and CD spectra, which show strong binding affinity for Con A with the binding constant of 3.8 × 10(7) m(-1) for monomeric mannose, nearly four orders of magnitude higher affinity than the monovalent mannose ligand. Furthermore, the fluorescence imaging of macrophage cell with PBI-Man is investigated, and shows selectively binding interaction with the mannose receptor-medicated cell entry. Moreover, the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) activities of PBI-Man show that PBI-Man as a biocompatible agent is noncytotoxic to living cells.

Clostridium acetobutylicum can propagate on fibrous matrices and form biofilms that have improved butanol tolerance and a high fermentation rate and can be repeatedly used. Previously, a novel macroporous resin, KA-I, was synthesized in our laboratory and was demonstrated to be a good adsorbent with high selectivity and capacity for butanol recovery from a model solution. Based on these results, we aimed to develop a process integrating a biofilm reactor with simultaneous product recovery using the KA-I resin to maximize the production efficiency of biobutanol.

Background: The gastroenteropancreatic neuroendocrine neoplasm (GEP-NEN) is the most common type of neuroendocrine neoplasm. We summarized data in our centre to investigate the clinicopathological features, diagnostic methods, therapeutic approaches and prognosis for this neoplasm to increase knowledge of this disease in Asian populations. Method: A total of 122 patients treated at Sun Yet-san Memorial Hospital of Sun Yat-sen University between January 2000 and December 2011 were analyzed retrospectively. Results: Pancreas was the most common site of involvement (65/122, 53.3%); this disease has no special symptoms; positive rates of chromogranin A (CgA) and synaptophysin (Syn) were 81.1% and 87.7%, respectively. The positive rate of Syn had statistical difference among the three grades, but not CgA. Some 68 patients had G1 tumors, 32 G2 tumors and 22 G3 tumors, and Chi-square test showed that higher grading was correlated with worse prognosis (?2=32.825, P=0.0001). A total of 32 patients presented with distant metastasis, and 8 cases emerged during following up. Cox proportional hazards regression modeling showed that the tumor grade (P=0.01), lymphatic metastasis (P=0.025) and distant metastasis (P=0.031) were predictors of unfavorable prognosis. The overall 5-year survival rate was 39.6%, the 5-year survival rate of G1 was 55.7%, and the G2 and G3 were 34.2% and 0%, respectively.Conclusions: The incidence of gastroenteropancreatic neuroendocrine tumors has risen over the last 12 years. All grades of these diseases metastasize readily, and further research regarding the treatment of patients after radical surgery is needed to prolong disease-free survival.

Accurate identification of cancer from inflammation and normal tissue in a rapid, sensitive, and quantitative fashion is important for cancer diagnosis and resection during surgery. Here we report the use of cyclooxygenase-2 as a marker for identification of cancer from inflammation and the design of a novel smart COX-2-specific fluorogenic probe (NANQ-IMC6). The probes fluorescence is "turned on" in both inflammations and cancers where COX-2 is overexpressed. Intriguingly, the fluorescent emission is quite different at these two sites with different expression level of COX-2. Hence, NANQ-IMC6 can not only distinguish normal cells/tissues from cancer cells/tissues but also distinguish the latter from sites of inflammation lesions by the different fluorescence recognition of NANQ-IMC6 for COX-2 enzymes. Following spraying with the NANQ-IMC6 solution, cancerous tissue, inflamed tissues, and normal tissues can be accurately discriminated in vivo by the unaided eye using a hand-held ultraviolet lamp emitting at 365 nm. So the probe may have potential application varying from cancer inflammation diagnosis to guiding tumor resection during surgery.

Diruthenium(II,III) carboxylates Ru2(esp)2Cl (1a), [Ru2(esp)2(H2O)2]BF4 (1b), and Ru2(OAc)4Cl (2) efficiently catalyze the oxygenation of organic sulfides. As noted in a previous work, 1a is active in oxygenation of organic sulfides with tert-butyl hydroperoxide (TBHP) in CH3CN. Reported herein in detail is the oxygenation activity of 1a, 1b, and 2, with the latter being highly selective in oxo-transfer to organic sulfides using TBHP under ambient conditions. Solvent-free oxidation reactions were achieved through dissolving 1a or 1b directly into the substrate with 2 equiv of TBHP, yielding TOF up to 2056 h(-1) with 1b. Also examined are the rate dependence on both catalyst and oxidant concentration for reactions with catalysts 1a and 2. Ru2(OAc)4Cl may be kinetically saturated with TBHP; however, Ru2(esp)2Cl does not display saturation kinetics. By use of a series of para-substituted thioanisoles, linear free-energy relationships were established for both 1a and 2, where the reactivity constants (?) are negative and that of 1a is about half that of 2. Given these reactivity data, two plausible reaction pathways were suggested. Density functional theory (DFT) calculation for the model compound Ru2(OAc)4Cl·TBHP, with TBHP on the open axial site, revealed elongation of the O-O bond of TBHP upon coordination.

Currently, there are lots of methods to select informative SNPs for haplotype reconstruction. However, there are still some challenges that render them ineffective for large data sets. First, some traditional methods belong to wrappers which are of high computational complexity. Second, some methods ignore linkage disequilibrium that it is hard to interpret selection results. In this study, we innovatively derive optimization criteria by combining two-locus and multilocus LD measure to obtain the criteria of Max-Correlation and Min-Redundancy (MCMR). Then, we use a greedy algorithm to select the candidate set of informative SNPs constrained by the criteria. Finally, we use backward scheme to refine the candidate subset. We separately use small and middle (>1,000 SNPs) data sets to evaluate MCMR in terms of the reconstuction accuracy, the time complexity, and the compactness. Additionally, to demonstrate that MCMR is practical for large data sets, we design a parameter w to adapt to various platforms and introduce another replacement scheme for larger data sets, which sharply narrow down the computational complexity of evaluating the reconstruct ratio. Then, we first apply our method based on haplotype reconstruction for large size (>5,000 SNPs) data sets. The results confirm that MCMR leads to promising improvement in informative SNPs selection and prediction accuracy.

A field experiment with successive planting of tomato was conducted to study the effects of adding different amounts of winter wheat straw (2.08 g x kg(-1), 1N; 4.16 g x kg(-1), 2N; and 8.32 g x kg(-1), 4N) to the soil seriously suffered from root knot nematode disease on the soil microbial biomass and protozoa abundance. Adding straw carbon source had significant effects on the contents of soil microbial biomass carbon (MBC) and microbial biomass nitrogen (MBN) and the abundance of soil protozoa, which all decreased in the order of 4N > 2N > 1N > CK. The community structure of soil protozoa also changed significantly under straw addition. In the treatments with straw addition, the average proportion of fagellate, amoeba, and ciliates accounted for 36.0%, 59.5%, and 4.5% of the total protozoa, respectively. Under the same adding amounts of wheat straw, there was an increase in the soil MBC and MBN contents, MBC/MBN ratio, and protozoa abundance with increasing cultivation period.

Carotid endarterectomy(CEA)has been proved to be an effective surgery to treat the cerebral ischemia caused by carotid atherosclerotic stenosis. However,there is still no effective mean for the early diagnosis of the CEA-related severe complications such as stroke and death. Many studies have explored the potential biomarkers for stroke alert,although there is still a long way to go for their actual application in clinical settings. The carotid atherosclerotic stenosis,the perioperative complications of CEA,and the stroke share similar pathogenic mechanisms,and some biomarkers such as S100B,matrix metalloproteinase 9,asymmetric dimethylarginine,and neuron-specific enolase have been studied in the clinical trails of CEA. This article summarizes recent advances in this field.

Estrogen reportedly facilitates visceral nociception at the spinal or supraspinal level. The present study was aimed to investigate whether estrogen modulates visceral pain through vagal pathway. Ovariectomized rats received estradiol, which was subcutaneously (to act through both vagal and spinal pathways) or intraduodenally (to preferentially act through vagal pathway) administered. Luminally applied estradiol induced a rapid and significant decrease in the visceromotor response (VMR) to colorectal distension (CRD), with increased c-Fos expression in nodose ganglion neurons. Systemically injected estradiol increased VMR and c-Fos expression in both nodose and dorsal root ganglion (T6-12) neurons. The antinociceptive effect of estrogen was abolished by surgically vagotomy or chemical denervation of vagal afferents. Both luminally and systemically administered estradiol elicited selective 5-HT secretion from the duodenum. Granisetron, a 5-hydroxytryptamine 3 receptor (5-HT3R) antagonist, reversed the antinociceptive effect of estrogen. Intestinal mucosal mast cell (IMMC) stabilizers prevented estradiol-induced antinociception and 5-HT secretion. Ultrastructural analysis revealed that estradiol caused piecemeal degranulation of IMMCs. The actions of estradiol were inhibited by an estrogen receptor (ER) ? antagonist and mimicked by an ER? agonist. These results suggest that estrogen can trigger vagus-mediated antinociception, which is masked by its spinally mediated pronociception.

The present paper studied fluorescence fingerprint properties of the municipal wastewater with industrial wastewater as major components. There existed three typical fluorescence peaks in the excitation-emission matrix of the municipal wastewater, locating at about lambda(ex)/lambda(em) of 275/310, 230/340 and 220/310 nm respectively. The wastewater didnt display typical protein-like fluorescence as the municipal wastewater with domestic sewage as major component. The fluorescence intensity of the wastewater was quite high with remarkable difference between workday and weekend. These might relate to the high content of industrial wastewater. The advantages of the fluorescence fingerprint such as easy and fast measurement and rich information about the components of wastewater make it a novel tool in water quality monitoring and early-warning.

Product quality control is crucial for industrial production, but there is lack of simple and effective detect methods. In this study, the chromatographically pure N-hexane from different manufacturers and same manufacturers with different batches was detected with fluorescence fingerprint technology. The results showed that the fluorescence fingerprints of all samples were different from each other. The numbers of fluorescence peaks of the fingerprints of the famous international manufacturer was stable and the intensity was low. The chromatographically pure N-hexane made in China generally had more peaks, higher intensities and greater changes as compared to the imported product. This indicated that the domestic products had more impurities with high concentration and the product quality was unstable. The study showed that the fluorescence fingerprint can be used as a novel method for quality control of chemical reagents.

p16(INK4A) is strongly expressed in tissues diagnosed as cervical intraepithelial neoplasia (CIN) and cancer in women infected with human papillomavirus (HPV), but few prospective studies have evaluated p16(INK4A) as a marker for the risk of low-grade CIN (CIN1) progression. We investigated the prevalence of p16(INK4A) immunostaining by CIN grade and whether overexpression of p16(INK4A) in CIN1 predicts future risk for high-grade CIN in Chinese women. 6,557 Chinese women aged 30-49 years were screened from 2003 to 2005 using cytology and carcinogenic HPV test. Colposcopy was performed on women with any abnormal result. p16(INK4A) Immunostaining was performed on biopsies from all women with CIN1, as well as randomly selected women with normal or CIN grade 2 and worse (CIN2+) biopsies. Women with CIN1 were followed up without treatment. Colposcopy was performed on all untreated women at a 2-year interval. The prevalence of p16(INK4A) staining was 2.7%, 42.7%, 75.5%, 79.6% and 100% among women with normal, CIN1, 2, 3 and cancer biopsies respectively (P<0.001). HPV positivity was strongly associated with p16(INK4A) staining (OR=12.8; 95% CI: 5.2-31.6). p16(INK4A) staining of CIN1 biopsies at baseline was associated with an increased risk of finding high-grade CIN over two years of follow up (OR=1.43; 95% CI: 0.52, 3.91). The two-year cumulative incidence of CIN2+ for p16(INK4A) positive women was higher at 10.71% than for p16(INK4A) negative women at 1.30% (crude RR=8.25, 95%CI: 1.02, 66.62). p16(INK4A) overexpression is strongly associated with grade of CIN and risk of progression to high-grade CIN in women with low-grade lesions. © 2013 Wiley Periodicals, Inc.

A new 8,4-oxyneolignane glucoside 1 has been isolated from the stems of Dendrobium aurantiacum var. denneanum together with six known phenolic glucosides 2?7. The structure of the new compound, including its absolute configuration, was determined by spectroscopic and chemical methods as (?)-(7S,8R,7E)-4-hydroxy-3,3,5,5-tetramethoxy-8,4-oxyneolign-7-ene-7,9,9-triol 7,9-bis-O-?-D-glucopyranoside (1). In the in vitro assays, compound 1 and (?)-syringaresinol-4,4-bis-O-?-D-glucopyranoside (2) showed evident activity against glutamate-induced neurotoxicity in PC12 cells. Shashenoside I (4) showed a selective cytotoxic activity with the IC?? value of 4.17 ?M against the acute myeloid leukemia cell line MV4-11, while it was inactive against 10 other human tumor cell lines.

Schistosomiasis japonica was endemic in 12 provinces (including municipalities and autonomous regions) in the Peoples Republic of China (PR China). Despite the tremendous decrease of schistosomiasis incidence after almost 60 years of control, the distribution of snail-breeding sites has not been reduced significantly. In order to verify current transmission risks and identify the potential establishment of new foci of schistosomiasis driven by environmental changes, we conducted surveillance in selected risk areas of three provinces: Jiangsu, Anhui and Shandong from 2008 to 2010 in addition to routine snail surveillance. We investigated populations and possible reservoirs in sentinel sites and report that the total number of new acute cases did not diminish further in spite of ongoing control activities. In Anhui Province the local count compared to the national count was 43% (19/44) in 2008, 33% (25/75) in 2009 and 40% (17/42) in 2010. In all, 31.58km(2) areas of snail breeding sites were newly detected nationwide through the year 2008-2010, of which the proportion of Anhui was 42% (5.03/11.98) in 2008, 95% (8.39/8.79) in 2009 and 79% (8.52/10.81) in 2010. Sentinel surveillance showed eight, nine and five confirmed cases of acute schistosomiasis in mobile populations (fishermen, migrant workers) in 2008, 2009 and 2010, respectively. All these cases were detected in Chaohu County, which must therefore be deemed an area at risk. We conclude that continuous surveillance with an emphasis on snails must be enhanced in potential risk areas in PR China.

Understanding associations between genotypes and complex traits is a fundamental problem in human genetics. A major open problem in mapping phenotypes is that of identifying a set of interacting genetic variants, which might contribute to complex traits. Logic regression (LR) is a powerful multivariant association tool. Several LR-based approaches have been successfully applied to different datasets. However, these approaches are not adequate with regard to accuracy and efficiency. In this paper, we propose a new LR-based approach, called fish-swarm logic regression (FSLR), which improves the logic regression process by incorporating swarm optimization. In our approach, a school of fish agents are conducted in parallel. Each fish agent holds a regression model, while the school searches for better models through various preset behaviors. A swarm algorithm improves the accuracy and the efficiency by speeding up the convergence and preventing it from dropping into local optimums. We apply our approach on a real screening dataset and a series of simulation scenarios. Compared to three existing LR-based approaches, our approach outperforms them by having lower type I and type II error rates, being able to identify more preset causal sites, and performing at faster speeds.

At present, approximately 20% of Hodgkin lymphomas (HL) are relapsed and refractory, and therapeutic methods including chemotherapy, radiotherapy, and even stem cell transplantation are unsatisfactory. Brentuximab vedotin, composed of CD30 antibody and a chemotherapeutic agent, is a new targeted drug that eradicates tumor cells by binding to the CD30 antigen on their surface. In clinical trials, the response rate and complete remission rate of this drug were 73% and 40%, respectively, for relapsed and refractory HL. Here we report a case of CD30-positive relapsed and refractory HL that was treated with brentuximab. Before the treatment with brentuximab, the patient underwent chemotherapy, radiotherapy, and autologous stem cell transplantation. However, the disease continued to progress, affecting multiple organs and prompting symptoms such as persistent fever. After the treatment with brentuximab, the patients condition improved. Body temperature returned to normal after 4 days. Lung nodules were reduced in size and number after a single course of treatment, and PET/CT showed partial remission and complete remission after 3 and 6 courses of treatment, respectively. The entire treatment process progressed smoothly, though the patient experienced some symptoms due to chemotherapy, including peripheral neuritis of the limbs, irritating dry cough, and mild increase in aminotransferase. No serious adverse effects were observed. The current general condition of the patient is good; the continuous complete remission has amounted to 6 months.

Transglutaminase 4 has been shown to enhance various biological properties of prostate cancer cells, e.g., cell-matrix adhesion, invasiveness and the epithelial-mesenchymal transition. The objectives of this study were to investigate the associations between transglutaminase 4 expression and the established features and biochemical recurrence of prostate cancer. Transglutaminase 4 immunostaining was performed on a tissue microarray. The expression of transglutaminase 4 was evaluated by a scoring method based on the intensity and extent of staining. The clinical and pathological information was obtained through a review of medical records. Follow-up data were obtained by consulting the hospital medical records and the prostate cancer database of our department and by contacting patients or family members. We then compared the transglutaminase 4 expression levels between the prostate cancer tissues and the paracarcinoma tissues and evaluated the correlation of transglutaminase 4 expression with the clinical parameters and biochemical recurrence of prostate cancer. Our results indicated that the transglutaminase 4 staining was significantly higher in tumour tissue than in paracarcinoma tissue (P<0.001) and was positively associated with higher Gleason score (P<0.001) and higher prostate-specific antigen level (P=0.005). Patients with transglutaminase 4 overexpression experienced shorter biochemical recurrence-free survival after surgery (P=0.042) in the univariate analysis but not in the multivariate analysis (P=0.139), which indicated that transglutaminase 4 may serve as a potential predictor of biochemical recurrence of prostate cancer.

Previous data have shown that the type II cGMP?dependent protein kinase (PKG II) inhibits the EGF?induced MAPK signaling pathway. In order to thoroughly investigate PKG, it is necessary to elucidate the function of another type of PKG, PKG I. The aim of this study was to investigate the possible inhibitory effect of PKG II and PKG I activity on the basic fibroblast growth factor (bFGF)?induced proliferation and migration of U251 human glioma cells and the possible underlying mechanisms. U251 cells were infected with adenoviral constructs encoding cDNA of PKG I (Ad?PKG I) or PKG II (Ad?PKG II) to increase the expression levels of PKG I or PKG II and then treated with 8?Br?cGMP and 8?pCPT?cGMP, respectively, to activate the enzyme. An MTT assay was used to detect the proliferation of the U251 cells. The migration of the U251 cells was analyzed using a Transwell migration assay. Western blot analysis was used to detect the phosphorylation/activation of the fibroblast growth factor receptor (FGFR), MEK and ERK and the nuclear distribution of p-ERK. The results showed that bFGF treatment increased the proliferation and migration of U251 cells, accompanied by increased phosphorylation of FGFR, MEK and ERK. Furthermore, the nuclear distribution of p-ERK increased following bFGF treatment. Increasing the activity of PKG II through infection with Ad-PKG II and stimulation with 8-pCPT-cGMP significantly attenuated the aforementioned effects of the bFGF treatment, while increased PKG I activity did not inhibit the effects of bFGF treatment. These data suggest that increased PKG II activity attenuates bFGF?induced proliferation and migration by inhibiting the MAPK/ERK signaling pathway, whereas PKG I does not.

It has been demonstrated that the spectrum of minor allelic frequencies (MAFs) of risk genetic variants ranges from common to rare. Although association studies are shifting to incorporate rare variants (RVs) affecting complex traits, existing approaches do not show a high degree of success, and more efforts should be considered.

This study demonstrates that ethinylestradiol (EE2), a priority estrogenic contaminant in water, can be rapidly and selectively removed from aqueous solutions using industrial-grade polyamide 612 (PA612) particles as adsorbents. Isothermal studies showed that nonporous low surface area (20 m(2) g(-1)) PA612 particles had a maximum adsorption capacity of 25.4 mg g(-1) for EE2 in water, which is higher or comparable to the results obtained with two benchmark activated carbon (AC) adsorbents (10.4-27.6 mg g(-1)). The adsorption of EE2 on PA612 followed pseudo-second order kinetics with a high adsorption rate exceeding those of the ACs by 5.3- to 22.4-fold. Computational chemistry calculations and molecular modeling showed that the strong binding affinity between EE2 and PA612 originates from the hydrophobic partitioning of EE2 solutes and hydrogen bonding interactions on PA612 amide groups. PA612 showed high adsorption selectivity for EE2 in water with highly consistent adsorption capacities for EE2 under the influence of a range of water chemistry parameters, including water salinity (NaCl, 1 mM-1 M), metal ions (K(I), Ca(II), and Zn(II); 0.1 M), natural organic matter (humic acids, 0.2-5 mg L(-1)), and pH level (4.8-9.1).

The survival rate of patients with metastatic colorectal cancer (mCRC) has significantly improved with applications of molecularly targeted drugs, such as bevacizumab, and led to a substantial improvement in the overall survival rate. These drugs are capable of specifically targeting the inherent abnormal pathways in cancer cells, which are potentially less toxic than traditional nonselective chemotherapeutics. In this review, the recent clinical information about molecularly targeted therapy for mCRC is summarized, with specific focus on several of the US Food and Drug Administration-approved molecularly targeted drugs for the treatment of mCRC in the clinic. Progression-free and overall survival in patients with mCRC was improved greatly by the addition of bevacizumab and/or cetuximab to standard chemotherapy, in either first- or second-line treatment. Aflibercept has been used in combination with folinic acid (leucovorin)-fluorouracil-irinotecan (FOLFIRI) chemotherapy in mCRC patients and among patients with mCRC with wild-type KRAS, the outcomes were significantly improved by panitumumab in combination with folinic acid (leucovorin)-fluorouracil-oxaliplatin (FOLFOX) or FOLFIRI. Because of the new preliminary studies, it has been recommended that regorafenib be used with FOLFOX or FOLFIRI as first- or second-line treatment of mCRC chemotherapy. In summary, an era of new opportunities has been opened for treatment of mCRC and/or other malignancies, resulting from the discovery of new selective targeting drugs.

To evaluate the screening performance of individual and combined use of clinical breast examination, ultrasonography and mammography in Chinese women, we conducted a biennial breast cancer screening program among 14,464 women aged 35 to 74 years old who lived in Qibao County, Minhang district of Shanghai, China, between May 2008 and Sept 2012. All participants were submitted to clinical breast examination, and then women with positive results and all women at age of 45-69 years old were preformed breast ultrasonography and mammography. The examination results were compared against pathological findings as the gold standard of reference. A total of 66 women were diagnosed with breast cancer in the two rounds of the screening, yielding an incident rate of 194 per 100,000 person-years. The sensitivity of clinical breast examination, ultrasonography and mammography alone were 61.4%, 53.7% and 67.3%, respectively. While mammography performed better in elder age groups and hormone receptor positive disease groups, ultrasonography had a higher sensitivity in younger age group and did not differ in sensitivity by estrogen receptor or progesterone receptor status. Combined use of the two imaging examinations increased the sensitivity in almost all age groups, but had a higher sensitivity in hormone receptor positive cancers than in those negative. Our results suggest that the Qibao modality is an effective strategy for breast cancer screening among Chinese women, especially for early detection of elder and hormone receptor positive breast cancer.

Petrochemical wastewater is one of major types of industrial wastewater in China. It is of huge quantity and causes serious pollution. Wastewater contains lots of fluorescence matters. Its fluorescence spectrum could exhibit organic components and unique for each sample like fingerprint. Thus it is referred to as fluorescence fingerprint of water quality, in brief aqueous fingerprint. This paper presented that there were almost 10 peaks in the aqueous fingerprint of petrochemical wastewater from a large-scale petrochemical plant, including the peak at the excitation/emission wavelengths (lambda(ex)/lambda(em)) of around 230/340 nm. That peak exists in the fluorescence spectra of various petroleum materials. The aqueous fingerprint was divided into 3 zones according to the relationship of fluorescence intensity of peaks: the linear relation between the peaks from different zones was not significant while that between the peaks from same zone was significant with coefficients of above 0.85. The zone around lambda(ex)/lambda(em) = 230/305 nm might relate to benzene compounds and the zone around lambda(ex)/lambda(em) = 220-320/230-440 nm (excluding the zone around lambda(ex)/lambda(em) = 230/305 nm) showed close relation with the raw materials (petroleum materials). The intensity of each peak had a fixed range. That range and relationships between peaks could be the evidences for diagnosing if the performance of the production processes is proper.

Two new diruthenium(II,III) tetramidate compounds, Ru(2)(NHOCC(CH(3))(2))(4)Cl (1) and Ru(2)(NHOCCH(2)CH(3))(4)Cl (2) have been prepared and structurally characterized by X-ray crystallography. The activity of promoting sulfide oxygenation using simple oxidants such as hydrogen peroxide (H(2)O(2)) and tert-butyl hydroperoxide (TBHP) was studied. A UV-kinetics study indicated that the initial rates of 1 and 2 are comparable to the previously studied diruthenium tetracarboxylates in promoting TBHP oxygenation of methyl phenyl sulfide (MPS). Using excess oxidant and CH(3)CN as the solvent, organic sulfides MPS and diphenyl sulfide (PPS) were oxidized using 1 mol% of the catalytic species. Compound 1 is more effective than 2 in converting sulfides to sulfoxide under the same conditions. Fast conversion was achieved when the reactions were carried out in the solvent-free conditions, and the major oxidation product was the sulfoxide. The electronic structure of the title compounds was studied with DFT calculations to gain an understanding of the activation of peroxy reagents.

The conventional parameters such as COD and BOD only could represent information about total organic content. Fluorescence spectrum can display organic composition and it is unique for each sample, so it is referred as "fluorescence fingerprint". In the present study transformation of excitation-emission matrix of municipal wastewater with sewage as major components after aerobic treatment was investigated and then the zones of biodegradable and non-biodegradable organic matters were figured out: the fluorescence at excitation wavelength/emission wavelength of about 280/340 nm and 225/240 nm derived from biodegradable organics and those of the zone of excitation wavelength above 300 nm and the zone of excitation wavelength below 300 nm and emission wavelength above 400 nm were mainly related with non-biodegradable organics. The above-mentioned results indicated that fluorescence fingerprint could be used to evaluate the performance and instruct design and operation of aerobic systems.

In developing our video relationships, we compare around 5 million PubMed articles to our library of over 4,500 methods videos. In some cases the language used in the PubMed abstracts makes matching that content to a JoVE video difficult. In other cases, there happens not to be any content in our video library that is relevant to the topic of a given abstract. In these cases, our algorithms are trying their best to display videos with relevant content, which can sometimes result in matched videos with only a slight relation.
