2. 腹腔注射
<ol>
	<li>小鼠注射采用单手手动固定法。调整手的位置，使其在颈部足够高的位置固定，以防止小鼠转头。</li>
	<li>用拇指和食指捏住颈背部的松弛皮肤。</li>
	<li>通过将背部的松弛皮肤夹在第二、第三、第四手指与拇指根部之间，或把尾巴夹在第三和第四手指之间，以稳定动物的后半身。</li>
	<li>大鼠腹腔注射需两人配合，一人固定大鼠，另一人实施注射。
	<ol>
		<li>用双手的食指和中指分别置于大鼠颈部两侧，手掌置于背部，拇指及第三、第四指环绕胸部，以防止大鼠前后移动。手指在颈部两侧的位置可防止大鼠转动头部。</li>
		<li>提起大鼠并将其翻转，使腹部朝上暴露。</li>
		<li>牢固抓住后肢和尾巴，并将其向身体外侧拉伸，以稳定后半身。</li>
	</ol>
	</li>

	<ol>
		<li>将针头垂直于脊柱方向，偏离中线，插入上述三角区域。将针头以一定角度刺入体壁平面，可利用腹部两侧进行安全注射。对于多次注射尤为重要，因可交替使用注射位点，从而减少组织损伤。</li>
		<li>当针头以角度刺入时，会“弹入”腹腔，便于判断进针深度。这也是针头正确就位的视觉和触觉提示。</li>
		<li>回抽注射器，确认针头位于腹膜腔内，而非膀胱、肠管或血管结构内。</li>
		<li>雄性小鼠应避免向尾侧注射，以防药液进入阴囊；雌性动物应避免注射至乳头部位，以防造成损伤。</li>
	</ol>
	</li>

3. 肌肉注射
<ol>
	<li>对小鼠和大鼠实施该技术的固定需要两人操作或使用固定管。此处描述的是一种利用固定装置的单人操作方法。
	<ol>
		<li>将动物放入固定装置中。</li>
		<li>动物进入固定管后，抓住其尾巴，使其后肢伸出管外，调整体位。</li>
		<li>抓住股骨近端部位的侧腹皮肤，以伸展腿部并防止膝关节弯曲。</li>
		<li>将固定装置放置于实验台面上，使动物头部朝下直立，或使动物朝向技术人员身体方向平放，以便在注射过程中稳定装置。</li>
		<li>注射定位标志如下：小鼠和大鼠的腿部均较小，臀肌（即后大腿肌肉）构成后肢中最大的肌群；后肢中第二大的肌群为腓肠肌（即小腿肌）。注射应从腿部后侧进行。
		<ol>
			<li>定位位于股骨后方的臀肌肌群。股骨可通过触诊确定，其周围的大块肌肉也易于感知。</li>
			<li>注意，从后方观察，腿部中线从跗关节（hock）延伸至尾部。毛发通常在中线处形成一条分界线，因为毛发生长方向分别来自外侧和内侧表面，呈相对分布。</li>
			<li>注射应避开中线，朝向大腿外侧进行。这可降低损伤位于腿部内侧表面的神经和血管的风险。</li>
			<li>对腓肠肌进行注射时，应从后侧进针，并需特别注意避免损伤外侧隐静脉。</li>
		</ol>
		</li>
		<li>在定位范围内进行注射
		<ol>
			<li>将针头垂直于股骨、偏离中线插入，如上所述。臀肌最大插入深度为 5 mm，腓肠肌最大插入深度为 3 mm。</li>
			<li>为避免肌肉损伤，必须防止针头在组织内不必要的移动。注射器应握持稳定，无需在注射过程中重新调整手部位置。</li>
			<li>回抽注射器以确认针头位于肌肉内而非血管内。</li>
			<li>以平稳、流畅的动作推注药液。切勿快速注射，应使肌肉缓慢扩张。快速注射将导致组织损伤。</li>

4. 利用尾静脉进行静脉注射
<ol>
	<li>动物的固定方式取决于动物是否处于麻醉状态。
	<ol>
		<li>对于清醒的小鼠或大鼠，使用圆柱形固定管，但无毛或裸鼠除外。由于无毛动物皮肤油脂较多，其皮肤容易粘附在刚性塑料上，因此很难重新定位或将其从丙烯酸固定装置中取出。此时应使用柔性塑料锥形固定器。</li>
		<li>其他注射辅助装置包括带照明平台、加热平台，以及刚性塑料锥形固定器，其中小鼠仅通过尾部张力固定在锥形器内。</li>
		<li>麻醉状态下的小鼠可能无需额外固定。</li>
		<li>加热整个动物身体或仅加热尾部，以确保尾部血管充分扩张。
		<ol>
			<li>使用设定为中等温度的电热垫或循环水毯对动物全身进行加热。</li>
			<li>将动物放入固定装置中，并用热源包裹。</li>
			<li>密切观察动物，一旦血管扩张即刻移除热源。也可使用加热灯照射笼具实现全身加热。使用加热灯时必须密切观察动物，因其容易过热。</li>
			<li>当无法进行全身加热时，可通过温热敷布（如加热毛巾）、热水袋或将尾部浸入温水中单独加热尾部。需特别注意避免烫伤尾部，因为热水袋、加热毛巾和温水通常缺乏温度控制，初始温度可能过高，且这些热源冷却速度较快。</li>
		</ol>
		</li>
		<li>使用组织油是增强血管可视化的另一种方法。
		<ol>
			<li>将棉签浸入组织油中，并将油涂抹于尾部被握持固定区域以外的部位。组织油可使血管轮廓更加清晰。</li>
			<li>市面上有售无毒油剂，其配方中含有可在蒸发时引起皮肤温热感并促进血管扩张的化学成分。涂抹此类油剂后，数分钟后待血管显现，用棉球擦去多余油分。</li>
			<li>所有类型的油均可能使尾部更难抓握。</li>
			<li>使用油剂时，一旦血管清晰可见，应立即进行注射。</li>
		</ol>
		</li>
		<li>尾静脉注射的解剖定位如下：
		<ol>
			<li>尾侧静脉位于尾部两侧。尾部背侧和腹侧中线也存在血管，但不适合用于注射。</li>
			<li>可旋转固定装置中的动物，使侧方血管朝上，便于注射；或直接旋转尾部，将血管调整至适合注射的位置。</li>
			<li>这些血管位置非常表浅，越靠近尾根部越为表浅。</li>
		</ol>
		</li>
		<li>将物质注入尾静脉需要一定技巧。为避免损伤血管，必须防止针头不必要的移动。持注射器的方式应确保在注射过程中无需重新调整手部位置。
		<ol>
			<li>调整尾部位置，使血管清晰可见并处于适度张力下。避免对尾部施加过大张力，否则可能导致血管被拉伸，血流减少。</li>
			<li>将针头尽可能远端地直接置于血管上方。</li>
			<li>施加轻微压力，将针头平行于脊椎方向滑入尾部。避免针头向下倾斜，否则会切断血管。推进针头时应能观察到其进入血管腔内至少2 mm。</li>
			<li>缓慢、平稳地推注药物，避免血管破裂。</li>
			<li>注射过程中需特别注意尾部反应。若针头正确进入血管，推注时应无阻力感，且可见血管因注射物推动血液而变白（褪色）。</li>
			<li>若针头未进入血管，推注时会感到明显阻力。若强行注射，药物将进入皮下间隙，导致尾部肿胀隆起。此时应立即停止注射，因为设计用于静脉注射的药物可能对周围组织具有腐蚀性。退出针头，并尝试在尾部更靠近头部的位置再次注射。</li>
			<li>成功注射后，拔出针头并对注射部位施加压力，确保充分止血后再将动物放回笼中。</li>
		</ol>

由于生物医学研究中使用的许多测试化合物是尚未商业化的新物质，因此正确的物质制备至关重要。必须充分考虑测试化合物及其溶解/悬浮所用介质或载体在无菌性、黏度以及生理相容性方面的基本问题。无论是经肠给药还是肠外给药，给药溶液都必须调节至适合该化合物吸收的生理pH值，以确保有效吸收并防止组织损伤。溶液的黏度可能决定给药途径的选择：若某物质过于黏稠，无法通过小鼠常用注射部位所需的小口径针头，则可能需要重新配制以便口服给药。所有肠外注射用溶液均须无菌，以避免将病原体引入动物体内。
注射时针头的选择取决于给药途径、溶液的黏度以及动物的大小。通常应选择能够注射溶液的最小规格针头；小鼠一般使用22-30G，大鼠则使用20-25G。注射器的选择同样应为能满足精确给药剂量所需最小规格且具备正确刻度的类型。
胃肠外注射有多种途径。本视频主要介绍最常用的几种途径（皮下注射 [SQ]、腹腔注射 [IP]、静脉注射 [IV] 和肌肉注射 [IM]）。其他注射技术，如皮内注射（ID）、足垫注射、鼻内注射以及尾静脉静脉注射，在另一段视频中介绍。
吸收速率取决于化合物的溶解度和给药途径。通常，静脉给药的吸收速度最快，其次是腹腔和肌肉注射。尽管腹腔注射属于非口服给药途径，其吸收机制实际上更接近口服给药。皮下给药是一种方便给予较大体积液体的方法，其吸收速率较其他途径慢，可产生持续作用。给药途径的选择是实验方案中的关键组成部分。

静脉注射是物质给药最有效的途径，因为药物可直接进入循环系统。然而，由于小鼠可用于静脉给药的血管较细，其应用受到限制。如果需要重复进行静脉给药，应考虑使用血管通路装置或其他专用给药设备，以保障动物福利。所选用的针头应尽可能细，但需能允许被注射物质的黏度通过，通常为27至30号针头。注射体积根据小鼠体型，每次可在0.05 ml至最多0.5 ml之间。
