2. 使用注射用麻醉剂诱导麻醉
注射用麻醉剂主要由氯胺酮与镇静剂或肌肉松弛剂混合而成。
常见的组合有：1）啮齿类鸡尾酒，包含氯胺酮（100 mg/ml）、赛拉嗪（20 mg/ml）、乙酰丙嗪（10 mg/ml）和无菌生理盐水（0.9% NaCl）；2）氯胺酮/赛拉嗪 2:1 混合液，包含氯胺酮（100 mg/ml）、赛拉嗪（20 mg/ml）和无菌生理盐水（0.9% NaCl）；以及 3）氯胺酮/赛拉嗪小鼠混合液，包含氯胺酮（100 mg/ml）、赛拉嗪（20 mg/ml）和无菌生理盐水（0.9% NaCl）。&#160;使用氯胺酮/赛拉嗪组合时，仅可使用氯胺酮进行追加给药，不可使用赛拉嗪，原因在于这两种药物的半衰期不同。
氯胺酮需与镇静剂和/或肌肉松弛剂配伍，配制时应先制备成储备液，以便从中抽取单次剂量。配制过程中必须精确量取各成分，并用无菌生理盐水稀释，以确保向动物给予正确的剂量。由于氯胺酮属于管制药品，必须在&#34;管制药品记录表&#34;上记录瓶中药品的使用量，且每种混合制剂均须设有单独的&#34;管制物质记录表&#34;。配制混合液时，应缓慢将氯胺酮加入瓶中，因为若快速注入易产生泡沫。混合液应使用无菌带塞的20 ml瓶盛装。瓶子必须正确贴上标签，注明化合物名称、配制日期、有效期、氯胺酮批号（因其为管制物质）以及建议剂量。有效期可根据其中最早到期的成分确定（具体依机构或所在州的规定而定）。为准确记录氯胺酮的使用情况，空瓶和装满混合液的瓶子均需称重，并将重量记录在混合液标签及为每瓶单独准备的管制物质记录表上。氯胺酮混合液应储存于避光、温度可控的环境中，以保持其效价。

3. 麻醉评估

还应使用心率、呼吸频率、血压、黏膜颜色和毛细血管再充盈时间等生理指标。虽然一般性观察可用于检测动物呼吸频率的变化，但若要利用心率或血压评估麻醉深度，则需要专门的设备。如果有心电图仪，可以测量心跳的速率和强度。测量血压时，可使用多种适配于尾巴甚至整个身体的装置。如表1所述，物理刺激会导致这三项参数均升高。
观察黏膜、眼睛、耳朵、口腔、鼻部、肛门以及（在较小程度上）脚掌和尾巴的颜色变化。这些区域应呈粉红色，表明呼吸和心脏功能正常。当动物进入IV期麻醉时，呼吸停止，导致黏膜及周围皮肤出现发绀，表现为蓝色或灰色。
毛细血管再充盈时间是指对组织施加压力使其变白后，外部毛细血管床恢复颜色所需的时间。用压舌板或手指按压麻醉动物的牙龈、耳廓或甲床，观察变白区域恢复为粉红色所需的时间，正常不应超过1-2秒。再充盈时间延长提示心率减慢或心肌收缩力下降，表明动物可能麻醉过深，接近死亡状态。
评估麻醉深度时，应使用多种不同的参数。对同一脚趾或耳朵反复进行捏压会导致该区域感觉迟钝，从而抑制反应，无法准确评估麻醉深度。应交替选择不同的脚趾和耳朵部位进行捏压评估。在整个手术过程中，应每隔10至30分钟重新评估一次麻醉深度。
研究表明，麻醉动物会出现心肺功能变化。使用注射类药物麻醉时，动物的呼吸频率较为稳定，但在心输出量方面表现出变异性。据报道，不同品系的动物对注射麻醉剂的反应差异很大，因此很难标准化给药剂量。&#160;吸入性麻醉剂往往会降低呼吸频率，但对心血管系统的影响较小。由于在手术过程中可方便地调整吸入麻醉剂的剂量，该方法常为首选。
关键术语与定义 
为安全维持手术所需麻醉深度，必须对麻醉状态进行精确且持续的监测。需监测的参数包括心率、呼吸频率、体温和血液氧合水平。对于小鼠和大鼠而言，由于其体型较小，这些参数均难以直接监测。啮齿类动物心率极快，常规用于听诊的听诊器无法准确捕捉心率，仅可用于判断心跳是否存在。小鼠正常心率为每分钟328–780次，大鼠正常心率为每分钟250–600次。啮齿类动物的呼吸频率也较高，通过肉眼观察或听诊方式难以准确计数。小鼠正常呼吸频率为每分钟90–220次，大鼠为每分钟66–144次。为准确测定心率和呼吸频率，必须使用专用的电子监测设备。传感器可手术植入动物体内，也可体外放置，并与动物所处的监测平台相连接。 
啮齿类动物麻醉相关死亡最常见的原因是低体温。啮齿类动物体表面积与体重之比较高，且麻醉状态下动物失去通过颤抖维持体温的能力。因此，在可存活手术过程中，必须监测体温并提供辅助热源（如加热垫）。小鼠正常体温范围为96.6–99.7°F（35.8–37.4°C），大鼠为96.6–99.5°F（35.9–37.5°C）。大多数体温计是为较大动物设计的，其原型为人用体温计。水银体温计已基本被数字式和电子式体温计取代。尽管数字式和电子式体温计在直肠、口腔和耳部使用时已被证实准确，但其尺寸不适用于小型啮齿类动物。专为小鼠和大鼠设计的直肠探头已可商业获取，建议使用。 
血液氧合水平用于评估肺部摄取氧气是否充分，从而反映啮齿类动物动脉血中氧气浓度是否适当。监测氧摄取量也可间接评估呼吸与通气状况，因其可揭示氧气是否充分吸入以及代谢废气是否有效呼出。心率也影响血液氧合，心率下降将导致血氧水平降低，可能引起组织灌注不足。 
麻醉人员的目标是以最低有效剂量或浓度的麻醉剂，使动物充分制动并完全消除疼痛感觉。为实现此目标，必须准确评估麻醉深度。麻醉分为四个阶段，其中手术麻醉阶段又分为四个平面。第一阶段表现为痛阈逐渐升高，仅出现轻微行为改变。第二阶段动物可能出现定向障碍，并表现出不规则呼吸模式，包括呼吸频率变化及偶发屏气。 
第三阶段为手术麻醉阶段。I平面表现为翻正反射消失及呼吸模式改变。眼睑反射和吞咽反射在该平面减弱或消失，II平面则丧失喉部和角膜反射。&#160; I平面和II平面均无遗忘和镇痛作用，因此动物必须达到III平面后方可开始手术操作。III平面导致肋间肌麻痹，从而出现腹式呼吸。尽管在III平面初期仅有部分镇痛作用，但随着麻醉加深，将逐步发展为完全遗忘和镇痛。在此水平下，动物已充分麻醉，适合进行手术操作。IV平面表示麻醉过量，动物可迅速进入第四阶段。 
随着麻醉深度进一步加深，可能出现危及生命的并发症。第四阶段表现为肋间肌和膈肌完全麻痹，导致严重呼吸暂停，进而引发呼吸停止、延髓麻痹、血管运动功能衰竭，最终导致死亡。此时瞳孔散大并固定于散大状态，肌肉完全松弛。

当重量比初始重量增加五十克时，通常表明罐体已耗尽。&#160;

下一步是组装诱导室。确保输入端连接麻醉蒸发器，输出端连接废气清除系统。

首先，将动物放入诱导室并盖紧盖子。动物进入诱导室后，以每分钟1升的流速启动氧气。然后将精密挥发罐的设定调整至异氟烷3-5%的诱导浓度，或根据机构指南进行调整。

动物完全麻醉后，在轻轻取出动物之前，先关闭异氟烷并用氧气冲洗腔室。此步骤有助于防止人员接触麻醉气体。

另一种诱导方法是使用与精密蒸发器相连的鼻锥或面罩。然而，由于麻醉气体具有难闻的气味，动物可能会抗拒通过面罩进行诱导。 

此外，由于抓握过紧还存在导致窒息的风险。因此，首选方法是使用诱导箱进行麻醉诱导，随后通过鼻罩维持麻醉。

在大多数装置中，锥形管和诱导室都连接到同一个蒸发器，并通过切换开关在两者之间切换麻醉气体的输送。

在麻醉舱内对动物进行麻醉后，将其面部固定在鼻罩中，并切换开关以将气体流向重定向至鼻罩。&#160;

监测呼吸，确认动物已放松后，将麻醉剂浓度降低至1-2%的维持水平，或根据机构指南进行调整。同时，将眼膏涂抹于眼睛上，以防止角膜干燥。

在麻醉时间超过5分钟时，啮齿类动物容易发生低体温；因此，需要进行可控的保温支持，以维持体温并避免过热。

对于注射用麻醉剂，通常使用氯胺酮与其它镇静剂或肌肉松弛剂（如赛拉嗪和/或乙酰丙嗪）的混合物。可使用这些化合物配制不同的组合，常见配比参见下文。&#160;

请注意，氯胺酮属于管制药品，因此其使用量必须记录在管制药品登记簿中，配制的混合物也必须单独建立管制药品登记记录。
