Genomic DNA in Eukaryotes

In Eukaryoten ist doppelsträngige DNA speziell in einem membrangebundenen Kern angeordnet, um den begrenzten Raum der Zelle zu nutzen.
 Auf der ersten Stufe der Verdichtung wird die DNA eng um bestimmte Proteine, sogenannte Histone, gewickelt.
 Ein Histonkern und die um ihn gewickelte DNA werden zusammen als Nukleosom bezeichnet, die Grundeinheit der DNA-Verpackung.
 Die Nukleosomen sind durch Linker-DNA miteinander verbunden, was Perlen an einer Schnur zu ähnelt.
 In der nächsten Verpackungsebene wickeln sich Nukleosomen und Linker-DNA zu Chromatinfasern.
 Schließlich verdichten zusätzliche faserige Proteine das Chromatin noch weiter und lassen so lange DNA-Längen so dicht verbundenen Einheiten werden, die je nach Phase der Zellteilung als Chromosomen bezeichnet werden.

Eukaryoten besitzen im Vergleich zu Prokaryoten große Genome. Damit ihr Genom in eine Zelle passt, ist die eukaryotische DNA außergewöhnlich dicht im Zellkern verpackt. Dafür wird die DNA eng um Proteine, die Histone, gewickelt. Sie sind in Nukleosomen verpackt, welche durch Linker-DNA verbunden sind und sich zu Chromatinfasern winden. Weitere Proteine verdichten das Chromatin noch mehr, so dass es in bestimmten Phasen der Zellteilung als Chromosomen erkennbar ist.
 
 <h4>Das menschliche Genom gemessen in Metern</h4>
 
 Die meisten Zellen im menschlichen Körper enthalten etwa 6 Milliarden Basenpaare an DNA, die in 23 Chromosomenpaare verpackt sind. Es ist daher schwer vorstellbar, wie viel DNA diese Zahl genau darstellt. Man kann sich daher ebenfalls nur schwierig vorstellen, wie dicht gepackt die DNA sein muss, um in eine Zelle zu passen. Wir können einen gewissen Einblick gewinnen, wenn wir das Genom in seiner Länge darstellen. Würden wir die DNA einer einzelnen diploiden Zelle zu einer geraden Linie anordnen, wäre sie etwa zwei Meter lang!
 
 Der Mensch hat jedoch kein ungewöhnlich großes Genom. Viele Fische, Amphibien und blühende Pflanzen haben viel größere Genome als der Mensch. Zum Beispiel enthält das haploide Genom der japanischen Blütenpflanze Paris japonica etwa 50-mal mehr DNA als das menschliche haploide Genom. Diese Zahlen unterstreichen die erstaunliche Arbeit, die Histone und andere chromatinumformende Proteine leisten müssen, um die DNA zu verpacken.

Eukaryotic DNA fits inside the nucleus by packing into a highly compact form. Eukaryotes have large genomes compared with prokaryotes, so their DNA must be tightly organized to fit inside one cell.

The process starts when DNA wraps around proteins called histones. These DNA-histone units form nucleosomes, which are connected by linker DNA. The nucleosomes then coil into chromatin fibers, and additional fibrous proteins compact the chromatin even more.

This packed material is called chromatin, and it becomes recognizable as chromosomes during certain stages of cell division. In human cells, there are about 6 billion base pairs of DNA arranged into 23 pairs of chromosomes. If the DNA from one diploid cell were stretched out in a straight line, it would measure about two meters long.

Humans are not unusual in genome size. Many fish, amphibians, and flowering plants have much larger genomes than humans. The Japanese flowering plant Paris japonica has a haploid genome with about 50 times more DNA than the human haploid genome. These examples show how strongly histones and other chromatin remodeling proteins must work to package DNA.
