DNA-Extraktion ist die Entfernung und Reinigung von DNA aus Zellen.
 Zun&#xE4;chst werden Zellen lysiert, aufgebrochen, normalerweise durch eine Kombination aus physikalischer St&#xF6;rung und Behandlung mit Chemikalien wie Detergenzien, die die Zelle und die Kernmembranen aufl&#xF6;sen.
 Der Inhalt kann dann frei in die umgebende L&#xF6;sung flie&#xDF;en.
 Als n&#xE4;chstes muss die DNA von den anderen vorhandenen Molek&#xFC;len getrennt werden.
 Organische L&#xF6;sungsmittel wie Phenol und Chloroform werden &#xFC;blicherweise verwendet, um Nukleins&#xE4;uren aufgrund ihrer unterschiedlichen L&#xF6;slichkeit von Proteinen zu trennen.
 Zus&#xE4;tzlich k&#xF6;nnen Proteinase- und RNase-Enzyme verwendet werden, um Proteine und RNA abzubauen, wobei die DNA intakt bleibt.
 &#xDC;blicherweise wird der Probe Salz zugesetzt, das die negativ geladenen Phosphatgruppen im R&#xFC;ckgrat der DNA stabilisiert und nach Zugabe von eiskaltem Alkohol die DNA aus der L&#xF6;sung ausf&#xE4;llt.
 Der wei&#xDF;e Niederschlag wird durch Drehen in einer Zentrifuge gesammelt, wo er sich am Boden des R&#xF6;hrchens absetzt.
 Nach dem Waschen und Resuspendieren in einer Pufferl&#xF6;sung kann die extrahierte DNA schlie&#xDF;lich f&#xFC;r Forschungs- oder Biotechnologieanwendungen verwendet werden.

DNA Isolation

DNA aus Zellen wird f&#xFC;r viele Anwendungen in der Biotechnologie und Forschung ben&#xF6;tigt. Dazu geh&#xF6;rt beispielsweise die molekulare Klonierung. Um DNA aus Zellen zu extrahieren und zu reinigen, verwenden die Forscher verschiedene Methoden der DNA-Extraktion. W&#xE4;hrend die Besonderheiten der verschiedenen Protokolle variieren k&#xF6;nnen, liegen dem Prozess der DNA-Extraktion einige allgemeine Konzepte zugrunde.
 
 <h4>Der Prozess</h4>
 
 Zuerst m&#xFC;ssen die Zellen lysiert bzw. aufgebrochen werden, um die DNA in eine L&#xF6;sung zu bringen. Die Zellen k&#xF6;nnen physisch mit Ger&#xE4;ten wie einem Homogenisator, einem Sonikator oder einem Bead-Beater lysiert werden. Hierbei werden die Zellen zermahlen oder auf andere Weise durch Kraft aufgebrochen. H&#xE4;ufig werden w&#xE4;hrend der Zelllysis Substanzen wie Detergenzien hinzugef&#xFC;gt, um die lipidbasierten Zellmembranen chemisch aufzubrechen. So wird die DNA aus dem Zellkern und der Zelle freigesetzt. Durch das Zentrifugieren werden die Zelltr&#xFC;mmer auf dem Boden sedimentiert. Das Lysat, das zellul&#xE4;re Materialien enth&#xE4;lt, wird f&#xFC;r die weitere Verarbeitung gesammelt.
 
 Die DNA muss dann von anderen zellul&#xE4;ren Molek&#xFC;len, wie RNA und Proteinen, getrennt werden. Deshalb werden oft Enzyme wie die RNase und Proteinase K w&#xE4;hrend oder nach der Zelllysis hinzugef&#xFC;gt, um RNA bzw. Proteine abzubauen. Zus&#xE4;tzlich werden h&#xE4;ufig organische L&#xF6;sungsmittel wie Phenol und Chloroform verwendet, um die DNA von den Proteinen zu trennen. Typischerweise wird die Probe mit Phenol-Chloroform gevortext und dann zentrifugiert, um die w&#xE4;ssrigen und organischen Phasen im Eppendorfgef&#xE4;&#xDF; zu trennen. Die DNA-haltige w&#xE4;ssrige Phase am oberen Ende kann dann abpipettiert werden, wobei die Proteine in der organischen Phase zur&#xFC;ckbleiben.
 
 Salze werden oft w&#xE4;hrend der DNA-Extraktion hinzugef&#xFC;gt, um die DNA zu stabilisieren und die Ausf&#xE4;llung aus der L&#xF6;sung zu unterst&#xFC;tzen. Eine Standardmethode der DNA-Pr&#xE4;zipitation ist die Zugabe von eiskaltem Alkohol, wie z.B. Ethanol oder Isopropanol. Dadurch bildet die DNA ein wei&#xDF;es, tr&#xFC;bes Pr&#xE4;zipitat innerhalb der Fl&#xFC;ssigkeit des Eppendorfgef&#xE4;&#xDF;es.
 
 Die Probe wird dann zentrifugiert, wodurch sich das DNA-Pr&#xE4;zipitat am Boden des Eppendorfgef&#xE4;&#xDF;es als Pellet sammelt. Das Pellet wird dann gewaschen, indem die Fl&#xFC;ssigkeit entfernt, Alkohol zugegeben und erneut zentrifugiert wird. Nach dem letzten Waschschritt wird das Pellet wieder in einem Puffer suspendiert. Dadurch entsteht eine w&#xE4;ssrige L&#xF6;sung der gereinigten DNA, die f&#xFC;r Untersuchungen oder zur Verwendung in der Biotechnologie genutzt werden kann.

DNA extraction is the process of removing and purifying DNA from cells. It is used in many biotechnology and research applications, including molecular cloning. The main goal is to free DNA from the cell and prepare it for later study or use.

The first step is cell lysis, which means breaking cells open. Researchers may use a homogenizer, sonicator, or bead beater to physically lyse the cells. They also often add detergent to chemically disrupt the lipid-based cell membranes. This helps release DNA from the nucleus and the rest of the cell.

After lysis, the sample is spun in a centrifuge. This causes cell debris to settle at the bottom, while the liquid lysate is kept for the next steps. The DNA then has to be separated from other cellular molecules, such as RNA and proteins. Enzymes like RNase and Proteinase K are often added to break down RNA and proteins, and phenol-chloroform can be used to separate DNA from protein.

During the phenol-chloroform step, the sample is vortexed and centrifuged again. The tube separates into an aqueous phase and an organic phase. The DNA stays in the aqueous phase at the top, and this layer can be pipetted off. Salts are often added to help stabilize the DNA and support precipitation.

To collect the DNA, ice-cold alcohol such as ethanol or isopropanol is added. The DNA then forms a white, cloudy precipitate in the tube. Another centrifuge spin pulls this DNA into a pellet at the bottom. The pellet is washed with alcohol, spun again, and then re-suspended in buffer to make a purified DNA solution ready for biotechnology work.
