Das Ruhemembranpotential ist die Differenz des elektrischen Potentials zwischen der Innenseite und der Außenseite der Zellmembran von Neuronen in Ruhe, wenn sie nicht stimuliert werden.
 Normalerweise liegt der Wert bei minus 70 Millivolt, was bedeutet, dass er im Inneren negativer ist.
 Zellmembranen sind selektiv durchlässig, da die meisten Ionen und Moleküle nicht passiv über sie diffundieren können.
 Sie benötigen häufig Transmembranproteine, wie Ionenkanäle, um sie passieren zu lassen.
 Wenn sich ein Neuron in Ruhe befindet, sind Kaliumkanäle die Hauptart von offenen Ionenkanälen.
 Ein weiteres Transmembranprotein, die Natrium-Kalium-Pumpe, nutzt Energie, um Natrium kontinuierlich aus der Zelle und Kalium in die Zelle zu befördern.

 Die relative Differenz der elektrischen Ladung oder Spannung zwischen der inneren und äußeren Seite der Zellmembran wird als Membranpotential bezeichnet. Es wird durch Unterschiede in der Durchlässigkeit der Membran für verschiedene Ionen und deren unterschiedlichen Konzentrationen entlang der Membran erzeugt.
 
 <h4>Das Innere eines Neurons ist stärker negativ</h4>
 
 Das Membranpotential einer Zelle kann gemessen werden, indem man eine Mikroelektrode in die Zelle einführt und die Ladung mit einer Referenzelektrode in der extrazellulären Flüssigkeit vergleicht. Das Membranpotential eines Neurons im Ruhezustand ist negativ. Ein Neuron befindet sich im Ruhezustand, wenn es in dem Moment keine Nachrichten empfängt oder sendet und die Spannung liegt typischerweise dann bei -70 Millivolt (mV). Dies wird das Ruhepotential der Membran bezeichnet. Der negative Wert gibt an, dass die Innenseite der Membran relativ betrachtet negativer ist als die Außenseite—sie ist polarisiert. Das Ruhepotential resultiert aus zwei Hauptfaktoren: der selektiven Permeabilität der Membran und den Unterschieden in der Ionenkonzentration zwischen dem Zellinneren und dem Zelläußeren.
 
 <h4>Membrandurchlässigkeit</h4>
 
 Zellmembranen sind selektiv durchlässig, da die meisten Ionen und Moleküle die Lipiddoppelschicht nicht ohne Hilfe durchqueren können, jedoch können Ionenkanalproteinen, die die Membran durchspannen den Transport ermöglichen. Das liegt daran, da die geladenen Ionen nicht durch das ungeladene, hydrophobe Innere der Membranen diffundieren können. Die häufigsten intra -und extrazellulären Ionen, die im Nervengewebe gefunden werden, sind Kalium (K+), Natrium (Na+), Chlorid (Cl-) und Kalzium (Ca2+). Wenn sich ein Neuron im Ruhezustand befindet, sind die Kalium (K+)-Kanäle die Ionenkanäle, die meistens offen sind-wodurch K+ durch die Membran wandern kann. Diese Permeabilität, zusammen mit den großen intrazellulären Mengen, bilden das Ruhemembranpotential des Neurons, das hauptsächlich durch die Bewegung von K+ bestimmt wird.
 
 <h4>Pumpen erzeugen Konzentrationsgradienten</h4>
 
 Die Unterschiede in den Ionenkonzentrationen zwischen dem Zellinneren und dem Zelläußeren der Neuronen sind hauptsächlich auf die Aktivität der Natrium-Kalium (Na+/ K+) Pumpe zurückzuführen. Das ist ein Transmembranprotein, die kontinuierlich drei Na+-Ionen für zwei K+-Ionen aus der Zelle herauspumpen. Dadurch werden Konzentrationsgradienten aufgebaut, wobei die Konzentration der Na+-Ionen außerhalb der Neuronen höher ist, während die Konzentration der K+-Ionen innerhalb der Neuronen höher ist.
 
 Da die Membran im Ruhezustand primär für K+ durchlässig ist, kann K+ aufgrund der offenen K+-Kanäle entlang seines Konzentrationsgradientens zu der Region mit der geringeren Konzentration diffundieren, also aus der Zelle raus. Diese positiven Ladungen, die die Zelle verlassen, kombiniert mit der Tatsache, dass es viele negativ geladene Proteine im Inneren der Zelle gibt, bewirken, dass das Innere der Zelle negativer ist.
 
 Letztlich wird die Diffusion von K+ nach außen durch die elektrostatische Abstoßung von positiven Ladungen, die sich außerhalb der Zelle ansammeln, ausgeglichen. Es wird ein elektrochemisches Gleichgewicht erreicht. Der Nettoeffekt ist das beobachtete negative Ruhepotential. Das Ruhepotential ist im Nervensystem sehr wichtig, da Änderungen des Membranpotentials-wie das Aktionspotential-die Grundlage für die neuronalen Signale sind.
 
 <h4>Achtung vor dem Kugelfisch</h4>
 
 Der Kugelfisch ist außerhalb Japans nicht oft auf Speisekarten für Meeresfrüchte zu finden. Das liegt zum Teil daran, dass er ein starkes Neurotoxin enthält. Das Tetrodotoxin (TTX) ist ein sehr selektiver spannungsgesteuerter Natriumkanalblocker, der in minimalen Dosen tödlich ist. Die mittlere tödliche Dosis (LD50) für Mäuse beträgt 334 μg/kg, verglichen mit 8,5 mg/kg bei Kaliumcyanid. Es hat auch als ein wesentliches Werkzeug in der neurowissenschaftlichen Forschung gedient. Das Toxin blockiert den Fluss von Na+ in die Zelle, wenn sich der Kanal öffnet. Es stört daher Aktionspotentiale, aber nicht das Ruhemembranpotential, und kann zur Stilllegung der neuronalen Aktivität verwendet werden. Sein Wirkungsmechanismus wurde 1964 von Toshio Narahashi und John W. Moore an der Duke University demonstriert, die an den riesigen Axonen von Hummern arbeiteten.

The Resting Membrane Potential

Neurons keep a negative resting charge across their membrane. This electrical difference, called the membrane potential, comes from how ions move and how the membrane lets them pass. In a resting neuron, the inside is typically around -70 millivolts and is more negative than the outside, so the cell is polarized.

Two main factors create this resting state. First, the membrane is selectively permeable, which means most ions and molecules cannot cross the lipid bilayer without help from ion channels. Second, ion concentrations are different inside and outside the cell. In nervous tissue, common ions include potassium (K+), sodium (Na+), chloride (Cl-), and calcium (Ca2+).

At rest, potassium channels are the main channels that are open. This makes potassium the ion that most strongly shapes the resting membrane potential. Potassium can move across the membrane more easily than many other ions, and its movement helps set the neuron’s negative charge.

The sodium-potassium pump creates the ion gradients that support this process. This transmembrane protein pumps three Na+ ions out of the cell for every two K+ ions it pumps in. As a result, Na+ builds up outside the neuron, and K+ builds up inside.

Because the membrane is mostly permeable to K+ at rest, potassium diffuses out of the cell down its concentration gradient. As positive charges leave, the inside becomes more negative. Negatively charged proteins inside the cell also contribute to this effect. Eventually, the outward movement of K+ is balanced by the electrical push from the positive charge outside the cell, and electrochemical equilibrium is reached.

This resting potential is important because changes in membrane potential are the basis of neural signaling. One example is the action potential, which depends on these voltage changes. The pufferfish toxin tetrodotoxin (TTX) blocks voltage-gated sodium channels and stops Na+ from entering when the channel opens. It disrupts action potentials, but it does not change the resting membrane potential, so it can be used to silence neuronal activity.
