Eine willkürliche Kontraktion der Skelettmuskulatur beginnt im Gehirn als bewusste Anstrengung vom Frontallappen zum primären motorischen Kortex, bevor ein Alpha-Motoneuron im Vorderhorn des Rückenmarks aktiviert wird.
 Das Signal leitet einen Nerv an die spezifische Muskelfaser wie den Bizeps weiter, wo die Aktionspotentiale an der neuromuskulären Endplatte enden, wo das Motoneuron einen synaptischen Kontakt mit einer Muskelfaser herstellt und die Freisetzung des Neurotransmitters Acetylcholin auslöst, die über den synaptischen Spalt diffundiert und an Rezeptoren bindet.
 Infolgedessen wird das Sarkolemma für Natriumionen durchlässiger, was zu mehr Aktionspotentialen führt, die sich entlang seiner äußeren Oberfläche und in das Innere der Muskelfaser durch Transfers oder T-Tubuli, die die Freisetzung von Calciumionen aus dem sarkoplasmatischen Retikulum in die Myofibrillen auslösen.
 Diese Freisetzung von Calcium initiiert die Actin-Myosin-Überbrückungsaktivität und die Beobachtung der Verkürzung und Kontraktion der Muskeln.

In Skelettmuskeln wird Acetylcholin durch Nervenenden an den motorischen Endplatten freigesetzt. Das ist der Ort der synaptischen Kommunikation zwischen Motoneuronen und Muskelfasern. Die Bindung des Acetylcholins an seine Rezeptoren in dem Sarkolemm ermöglicht den Einfluß von Natrium-Ionen in die Zelle und löst ein Aktionspotential in der Muskelzelle aus. Dadurch werden elektrische Signale von dem Gehirn an den Muskel übertragen. Anschließend baut das Acetylcholinesterase-Enzym Acetylcholin ab, um eine übermäßige Muskelstimulation zu verhindern.
 
  
 
 Personen die an der Krankheit Myasthenia gravis leiden, entwickeln Antikörper gegen die Acetylcholinrezeptoren. Dadurch wird die Übertragung elektrischer Signale zwischen Motoneuron und Muskelfaser verhindert und somit die Skelettmuskelkontraktion beeinträchtigt. Myasthenia gravis wird mit Medikamenten behandelt, die die Acetylcholinesterase hemmen. Dadurch hat der Neurotransmitter mehr Möglichkeiten, die verbleibenden Rezeptoren zu stimulieren. Außerdem kann auch das Immunsystem unterdrückt werden, wodurch die Bildung von Antikörpern verhindert wird.
 
 <h4>Kontraktion der glatte Muskulatur</h4>
 
 Im Gegensatz zu den Skelettmuskeln werden die glatten Muskelzellen, die sich in den Wänden der inneren Organe befinden, durch das autonome Nervensystem innerviert und unterliegen unwillkürlichen Kontraktionen. Die Kontraktion wird durch die Interaktion zwischen den zwei Filamentproteinen Aktin und Myosin ermöglicht. Die Interaktion von Aktin und Myosin ist stark mit der intrazellulären Kalziumkonzentration verknüpft. Als Antwort auf Neurotransmitter- bzw. Hormonsignale oder der Dehnung des Muskels tritt extrazelluläres Kalzium durch die Kalziumkanäle im Sarkolemm in die Zelle ein oder wird intrazellulär von dem sarkoplasmatischem Retikulum freigesetzt. Im Inneren der Zelle bindet sich Kalzium an das Regulationsprotein Calmodulin. Der Kalzium-Calmodulin-Komplex aktiviert dann die Myosin-Light-Chain-Kinase, welche das Myosin phosphoryliert und die Wechselwirkung mit Aktin ermöglicht, wodurch der Muskel sich zusammen zieht.

Nerve signals trigger muscle contraction in both skeletal and smooth muscle, but the two muscle types use different control systems. In skeletal muscle, the signal begins at the motor end plate, which is the synapse between a motor neuron and a muscle fiber. In smooth muscle, contraction is controlled by the autonomic nervous system and happens without conscious control.

In skeletal muscle, motor nerve terminals release acetylcholine. This neurotransmitter binds to receptors on the sarcolemma, the muscle cell membrane, and lets sodium ions enter the cell. That change starts an action potential in the muscle cell and carries the electrical signal from the brain to the muscle. The enzyme acetylcholinesterase then breaks down acetylcholine so the muscle is not overstimulated.

Myasthenia gravis shows what happens when this signaling step is disrupted. In this disorder, antibodies form against the acetylcholine receptor. As a result, the signal between the motor neuron and the muscle fiber is blocked, and skeletal muscle contraction is weakened. Treatment may include drugs that inhibit acetylcholinesterase or medicines that suppress the immune system.

Smooth muscle in the walls of internal organs contracts in a different way. Its contraction depends on the interaction between actin and myosin, two filament proteins. This process is closely linked to calcium levels inside the cell, which can rise after neurotransmitter or hormone signals or when the muscle is stretched.

Calcium can enter smooth muscle from outside the cell through calcium channels on the sarcolemma, or it can be released from the sarcoplasmic reticulum inside the cell. Once inside, calcium binds to calmodulin, a regulatory protein. The calcium-calmodulin complex activates myosin light chain kinase, which phosphorylates myosin and lets it interact with actin, causing contraction.
