Die Erforschung von
individuellen Genen hat
zu außerordentlichen Einblicken
in nahezu alle Aspekte
der Biologie geführt, von
Krankheiten und Vererbung,
bis hin zur Landwirtschaft und Evolution.
Allerdings können viele Fragen darüber,
wie Gene funktionieren und sich entwickeln,
nur durch die Erforschung der DNA -
dem Genom eines Organismus beantwortet werden,
zusammen als ein Ganzes.
Die Erforschung vollständiger
Genome ist bekannt als Genomik.
Sie bezieht die Sequenzierung
ganzer Genome mit ein,
um ihre Organisation,
Funktion, Regulierung
und Evolution zu untersuchen.
Genomik befindet sich
am Schnittpunkt zwischen
Genetik, Molekularbiologie
und rechnergestützten Wissenschaften.
Nachdem die Struktur der DNA
im Jahr 1953 identifiziert wurde,
begannen Wissenschaftler Techniken
zu entwickeln, um individuelle
Gene zu sequenzieren.
Ein wichtiger Durchbruch
bei der DNA-Sequenzierung
kam im Jahr 1977, als
Frederick Sanger
eine Methode entwickelte, die es
ermöglichte, ganze Genome zu sequenzieren.
Das führte zur Sequenzierung der
Genome von Backhefe
und der Fruchtfliege unter Verwendung der
Shotgun-Sequenzierungsmethode.
Das Human Genome-Projekt,
das im Jahr 1990 begann,
war ein 13-jähriges internationales
Unterfangen von 2,7 Mrd US-Dollars,
mit dem das erste menschliche Genom
im Jahr 2003 erfolgreich sequenziert wurde.
Der Erfolg dieses Projekts führte
zu Innovationen bei Sequenzierungstechnologien
und menschliche Genome
können nun an einem Tag für
weniger als tausend Dollar sequenziert werden.
Die großen Mengen an
generierten Daten führten
zur Entstehung des
Fachbereichs der Bioinformatik,
die mathematische Wissenschaften
und Techniken der Informatik
kombiniert, um die Unmengen von
genomischen Daten zu organisieren,
zu analysieren und
miteinander zu vergleichen.
Genomik ist nun eine zentrale
Komponente personalisierter
Medizin, zum Beispiel
bei der Behandlung von Krebs,
und sie spielt weiterhin
eine Schlüsselrolle
in Spezialgebieten wie Anthropologie,
Evolution, Forensik
und Landwirtschaft,
unter anderen.

Genomics

Die Genomforschung ist die Wissenschaft der Gene: Es ist die Untersuchung des gesamten genetischen Materials eines Organismus. Beim Menschen besteht das Genom aus Informationen, die in 23 Chromosomenpaaren im Zellkern sowie aus mitochondrialer DNA gespeichert werden. In der Genomforschung wird sowohl kodierende als auch nicht-kodierende DNA sequenziert und analysiert. Die Genomforschung ermöglicht ein besseres Verständnis aller Lebewesen, ihrer Evolution und ihrer Vielfalt. Es hat eine Vielzahl von Anwendungen: zum Beispiel, um phylogenetische Bäume zu erstellen, die Produktivität und Nachhaltigkeit von Kulturen zu verbessern, strafrechtliche Untersuchungen zu unterstützen, krankheitsgebende Gene zu identifizieren, oder für eine optimale Behandlung der Krebspatienten.
 
 In den letzten 30 Jahren fanden bedeutende technologische Entwicklungen in der DNA-Sequenzierung statt, insbesondere dank der internationalen Bemühungen des menschlichem Genomprojekts. Es gibt jetzt Technologien, die 60 Milliarden DNA-Basen sequenzieren können, was 20-mal so groß ist wie das menschliche Genom. Mit dieser Fülle an Informationen und Big Data arbeiten Informatiker und Bioinformatiker Hand in Hand mit Biologen, um die DNA-Sequenzierungsergebnisse zu sammeln, zu speichern und zu analysieren, während die Daten sicher abgerufen werden können.
 
 Der Erfolg der Genomforschung hat den Weg für die Erforschung biologischer Systeme in ihrer Gesamtheit durch andere groß angelegte Methoden geebnet. Genom-Studien können nun mit anderen "Omics"-Studien vereint werden, um Daten über lebende Gruppen aller Ebenen zu sammeln: Arten, Populationen, Individuen, Zellen, Proteine, RNA, DNA. Die Transkriptomforschung ist die Untersuchung wie die Gesamtexpression von Genen unter verschiedenen experimentellen oder pathologischen Bedingungen variiert. Proteomforschung umfasst die Untersuchung aller Proteine, die einen Organismus oder ein System bilden. Metabolomics ist die Untersuchung aller Metabolite (Zucker, Fette, andere Moleküle), die in einem gegebenen biologischen System enthalten sind, und ihrer Wechselwirkungen. Untrennbar von der Bioinformatik ermöglichen die "Omics"-Studien einen globalen Blick auf komplexe Lebensvorgänge in ihrer Umgebung und ebnen damit den Weg für eine personalisierte medizinische Behandlung.

Genomics studies the full set of genetic material in an organism. In humans, that genome includes the DNA carried in 23 pairs of chromosomes in the nucleus, along with mitochondrial DNA. Genomics looks at both coding DNA and non-coding DNA, which helps scientists study how living things are built and how they differ.

This field is useful in many areas of biology and medicine. Scientists use genomics to build phylogenetic trees, improve crop productivity and sustainability, help with criminal investigations, find genes linked to disease, and support better cancer treatment choices. It also gives researchers a clearer view of evolution and biological diversity.

DNA sequencing has changed genomics over the past 30 years. Major progress came from the international Human Genome Project, which helped drive faster and more powerful sequencing methods. Today, technology can sequence about 60 billion DNA bases, or around 20 times the size of a human genome.

That amount of data requires bioinformatics, the use of computers to manage and study biological information. Computer scientists and bioinformaticians work with biologists to collect, store, analyze, and secure DNA sequencing results. Their teamwork makes it possible to handle large-scale genomic data.

Genomics also led to other large-scale &quot;omics&quot; studies. Transcriptomics examines how gene expression changes under different experimental or disease conditions. Proteomics studies all the proteins in an organism or system, and metabolomics studies the metabolites, such as sugars, fats, and other molecules, and how they interact.

Together, these omics fields give a broad view of living systems at the level of species, populations, individuals, cells, proteins, RNA, and DNA. Because they rely on bioinformatics, they help scientists understand complex biology in context. They also support the growth of personalized medicine.
