Stereoisomere von Koordinationsverbindungen sind Moleküle, die die gleiche chemische Formel und die gleiche Konnektivität der Liganden an das Metallatom haben, sich aber in der Anordnung der Ligandenmoleküle um das Zentralatom unterscheiden.
Basierend auf der molekularen Struktur werden Stereoisomere von Koordinationsverbindungen als geometrische Isomere oder optische Isomere klassifiziert.
Geometrische Isomere sind Stereoisomere, die völlig unterschiedliche Molekülgeometrien aufweisen und unterschiedliche physikalische und chemische Eigenschaften aufweisen.
cis-trans-Isomere sind ein Beispiel für geometrische Isomere. Sie treten in quadratisch-planaren Komplexen mit zwei Sätzen von zwei identischen Liganden oder MA2B2 und in oktaedrischen Komplexen mit einem Satz von vier identischen Liganden und einem Paar eines anderen Liganden oder MA4B2 auf. 
Im cis-Isomer des quadratisch-planaren Komplexes Diamminedichlorplatin(II), bekannt als Cisplatin, befinden sich die Chloridliganden beispielsweise nebeneinander und auf derselben Seite des Moleküls. Im trans-Isomer Transplatin befinden sich die Chloridliganden auf den gegenüberliegenden Seiten des Moleküls. 
Oktaederkomplexe mit zwei Sätzen von drei identischen Liganden oder MA3B3 können eine andere Art von geometrischer Isomerie aufweisen, die als Fac-mer-Isomerie bekannt ist. 
Zum Beispiel befinden sich im fac-Isomer von Triamminetrichlorocobalt(III) die drei Aminliganden und drei Chloridliganden auf gegenüberliegenden Seiten des Moleküls und bilden somit entgegengesetzte Flächen des Oktaeders. Im Mer-Isomer bilden drei gleiche Liganden einen Bogen um das zentrale Metallatom.
Die andere Klasse von Stereoisomeren wird als optische Isomere definiert. Wie unsere Hände sind auch optische Isomere spiegelbildlich zueinander und nicht überlagerbar. Diese Isomere, auch Enantiomere genannt, werden als chiral bezeichnet und unterscheiden sich nur sehr wenig in ihren physikalischen und chemischen Eigenschaften.
Optische Isomere zeichnen sich durch ihre Wechselwirkung mit planpolarisiertem Licht aus. Polarisiertes Licht hat elektrische Feldvektoren, die in einer einzigen Ebene oszillieren, die um einen bestimmten Betrag gedreht wird, wenn das polarisierte Licht eine Lösung eines Enantiomers passiert. 
Zum Beispiel dreht ein Enantiomer von Tris(ethylendiamin)cobalt(III) die Ebene nach rechts und wird als rechtsdrehend oder das optische Isomer d bezeichnet. Das andere Enantiomer dreht die Ebene nach links und wird als levorotatorisch oder optisches Isomer l bezeichnet.

<h3>Isomerie in Komplexen</h3> Isomere sind verschiedene chemische Spezies, die die gleiche chemische Formel haben. Übergangsmetallkomplexe liegen häufig als geometrische Isomere vor, in denen dieselben Atome über dieselben Bindungstypen verbunden sind, jedoch Unterschiede in ihrer Orientierung im Raum aufweisen. Koordinationskomplexe mit zwei verschiedenen Liganden in der cis- und trans-Position eines interessierenden Liganden bilden Isomere. Beispielsweise weist das oktaedrische [Co(NH3)4Cl2]+-Ion zwei Isomere auf (Figure 1). In der cis-Konfiguration liegen die beiden Chloridliganden nebeneinander. Beim anderen Isomer, der trans-Konfiguration, liegen sich die beiden Chloridliganden direkt gegenüber. <img alt="Image1" src="/files/ftp_upload/11459/11459_Image1.svg" style="text-align: center; height: 200px;"> Figure 1. Verschiedene geometrische Isomere eines Stoffes sind unterschiedliche chemische Verbindungen. Sie weisen unterschiedliche Eigenschaften auf, obwohl sie die gleiche Formel haben. Beispielsweise unterscheiden sich die beiden Isomere von [Co(NH3)4Cl2]NO3 in der Farbe; die cis-Form ist violett und die trans-Form ist grün. Darüber hinaus weisen diese Isomere unterschiedliche Dipolmomente, Löslichkeiten und Reaktivitäten auf. Als Beispiel dafür, wie die Anordnung im Raum die molekularen Eigenschaften beeinflussen kann, betrachten wir die Polarität der beiden [Co(NH3)4Cl2]NO3-Isomere. Denken Sie daran, dass die Polarität eines Moleküls oder Ions durch die Bindungsdipole (die auf den Unterschied in der Elektronegativität der Bindungsatome zurückzuführen sind) und ihre Anordnung im Raum bestimmt wird. In einem Isomer verursachen cis-Chloridliganden eine höhere Elektronendichte auf einer Seite des Moleküls als auf der anderen, wodurch es polar wird. Beim trans-Isomer steht jedem Liganden ein identischer Ligand direkt gegenüber, sodass sich die Bindungsdipole aufheben und das Molekül unpolar ist. Eine weitere wichtige Art von Isomeren sind optische Isomere oder Enantiomere, bei denen zwei Objekte exakte Spiegelbilder voneinander sind, sich aber nicht so anordnen lassen, dass alle Teile übereinstimmen. Das bedeutet, dass optische Isomere nicht überlagerbare Spiegelbilder sind. Ein klassisches Beispiel dafür ist ein Händepaar, bei dem die rechte und die linke Hand spiegelbildlich zueinander sind, aber nicht übereinander gelegt werden können. Optische Isomere sind in der organischen Chemie und Biochemie sehr wichtig, da lebende Systeme häufig ein bestimmtes optisches Isomer enthalten und das andere nicht. Im Gegensatz zu geometrischen Isomeren haben Paare optischer Isomere nahezu identische Eigenschaften (Siedepunkt, Polarität, Löslichkeit usw.). Optische Isomere unterscheiden sich nur in der Art und Weise, wie sie polarisiertes Licht beeinflussen und wie sie mit anderen optischen Isomeren reagieren. Bei Koordinationskomplexen bilden viele Koordinationsverbindungen wie [M(en)3]n+ [in dem Mn+ ein zentrales Metallion wie Eisen(III) oder Kobalt(II) ist] Enantiomere, wie in Figure 2 dargestellt. Diese beiden Isomere reagiert anders mit anderen optischen Isomeren. Beispielsweise sind DNA-Helices optische Isomere, und die in der Natur vorkommende Form (rechtsdrehende DNA) bindet nur an ein Isomer von [M(en)3]n+ und nicht an das andere. <img alt="Image2" src="/files/ftp_upload/11459/11459_Image_2.svg" style="text-align: center; height: 200px;"> Figure 2. Isomer shapes in coordination complexes can change a compound’s properties even when the chemical formula stays the same. Isomers are different chemical species with the same formula. In transition metal complexes, the atoms stay connected through the same types of bonds, but their arrangement in space can differ.

One common form is geometric isomerism. In these complexes, ligands (the ions or molecules attached to the metal) can sit in cis or trans positions. The octahedral [Co(NH3)4Cl2]+ ion has two geometric isomers. In the cis form, the two chloride ligands are next to each other. In the trans form, the chlorides are directly across from one another.

These geometric isomers can behave very differently. The two isomers of [Co(NH3)4Cl2]NO3 have different colors. The cis form is violet, and the trans form is green. They also differ in dipole moment, solubility, and reactivity. Polarity depends on bond dipoles and how they are arranged in space. In the cis isomer, the chloride ligands create more electron density on one side of the ion, so it is polar. In the trans isomer, matching ligands are opposite each other, so the dipoles cancel and the ion is nonpolar.

Another important category is optical isomerism, also called enantiomerism. Enantiomers are mirror images that cannot be superposed, like right and left hands. These optical isomers are important in organic chemistry and biochemistry because living systems often use one optical isomer but not the other. Unlike geometric isomers, enantiomers usually have nearly identical boiling points, polarity, and solubility. They differ in how they affect polarized light and how they react with other optical isomers.

Many coordination compounds also form enantiomers. A complex such as [M(en)3]n+, where M is a metal ion like iron(III) or cobalt(II) and en is ethylenediamine, can exist as a pair of non-superposable mirror images. These forms react differently with other optical isomers. For example, DNA helices are optical isomers, and natural right-handed DNA binds to only one isomer of [M(en)3]n+.

Some complexes show both geometric and optical isomerism. The [Co(en)2Cl2]+ ion has cis and trans forms. Its trans isomer has the chlorines 180° apart and has very different properties from the cis form. The cis isomer also exists as a pair of optical isomers. These mirror-image forms behave the same in most ways, except when they react with other enantiomers.
