Bei einer nukleophilen Substitutionsreaktion wird das Reaktantenmolekül, das die Abgangsgruppe im Substrat verdrängt, als Nukleophil bezeichnet.
Nukleophile sind elektronenreiche Spezies und per Definition Lewis-Basen. Das nukleophile Atom hat eine hohe Elektronendichte und gibt seine Elektronen an ein teilweise positives, elektronenarmes Zentrum ab, wodurch eine neue Bindung entsteht.
Nukleophile Reagenzien sind entweder anionisch oder neutral. Anionische Nukleophile sind negative Ionen, die ein oder mehrere einsame Paare gleicher Energie enthalten, normalerweise auf Heteroatomen. Die nicht bindenden Einzelpaare besetzen hochenergetische Molekülorbitale, wodurch sie weniger stabil und reaktionsschneller sind.
Zum Beispiel hat das anionische Kohlenstoffnukleophil wie das Cyanidion ein einzelnes Paar sowohl auf dem Kohlenstoff als auch auf dem Stickstoff. Da das sp-Orbital des Kohlenstoffs jedoch energiereicher ist als das des Stickstoffs, ist Kohlenstoff das nukleophile Zentrum.
Neutrale Nukleophile haben ein oder mehrere ungeteilte Elektronenpaare auf den höchsten besetzten Molekülorbitalen von meist Heteroatomen.
Darüber hinaus fungiert bei Spezies wie Alkenen, die keine einsamen Paare haben, ein Bereich mit hoher Elektronendichte – die Pi-Bindung – als nukleophile Stelle.
Ein neutrales Nukleophil ist weniger nukleophil als seine anionische Form, da keine negative Ladung vorhanden ist.
Im Allgemeinen können pKa-Werte von Säuren verwendet werden, um die Stärke ihrer konjugierten Basen oder Nukleophile zu bewerten. Bei Molekülen, die das gleiche nukleophile Atom enthalten, ist das Nukleophil umso stärker, je höher der pKas ihrer konjugierten Säuren ist.     
Das Produkt einer nukleophilen Substitutionsreaktion hängt von der Art des verwendeten Nukleophils ab.
Wenn ein anionisches Nukleophil mit dem Substrat reagiert, neutralisiert die gebildete kovalente Bindung die formale Ladung des Nukleophils, was zu einem neutralen Produkt führt.
Im Vergleich dazu erhält ein neutrales Nukleophil, wenn es mit dem Substrat reagiert, eine positive formale Ladung. Ein anschließender Deprotonierungsschritt schließt die Reaktion ab, wodurch ein neutrales Produkt entsteht.

Das Wort „Nukleophil“ hat eine griechische Wurzel und bedeutet kernliebend. Nukleophile sind entweder negativ geladene oder neutrale Spezies mit einem Elektronenpaar in einem hochenergetisch besetzten Molekülorbital (HOMO). Da diese Spezies dazu neigen, Elektronenpaare abzugeben, werden Nukleophile ebenfalls als Lewis-Basen betrachtet. Negativ geladene Spezies wie OH−, Cl− oder HS− mit einem oder mehreren Elektronenpaaren sind typischerweise Nukleophile. Ebenso haben neutrale Spezies wie Ammoniak, Amine, Wasser und Alkohol nichtbindende freie Elektronenpaare und können als Nukleophile wirken. Darüber hinaus können Moleküle ohne ein einzelnes Elektronenpaar immer noch als Nukleophile wirken, wie etwa Alkene und aromatische Ringe mit bindenden π-Orbitalen.

Die relative Stärke eines Nukleophils, eine Abgangsgruppe in einer Substitutionsreaktion zu verdrängen, wird als Nukleophilie bezeichnet. Die negativ geladenen Spezies sind nukleophiler als ihre neutralen Gegenstücke. Als empirische Regel gilt: Je höher der pKa-Wert einer konjugierten Säure, desto besser das Nukleophil. Beispielsweise ist das Hydroxidion – eine konjugierte Base von Wasser (pKa 15,7) – ein besseres Nukleophil als das Acetation – eine konjugierte Base von Essigsäure (pKa ~ 5).

Da Nukleophilie keine inhärente Eigenschaft einer bestimmten Spezies ist, wird sie von vielen Faktoren beeinflusst, einschließlich der Art des Lösungsmittels, in dem die Reaktion durchgeführt wird. In polaren protischen Lösungsmitteln verringert eine hohe Solvatation von Anionen die Verfügbarkeit des Nukleophils zur Teilnahme an Substitutionsreaktionen.

Beim Vergleich von Halogeniden wird Fluorid, das kleinste und elektronegativste Anion, am stärksten solvatisiert, während Iodid, das größte und am wenigsten elektronegative Ion, am wenigsten solvatisiert wird. Daher ist Iodid in polaren protischen Lösungsmitteln das beste Nukleophil. In polaren aprotischen Lösungsmitteln sind Anionen jedoch aufgrund der schlechten Solvatation „nackt“ und können ungehindert an einem nukleophilen Angriff teilnehmen. In polaren aprotischen Lösungsmitteln bestimmt die Basizität des Nukleophils seine Nukleophilie, was Fluorid zum besten Nukleophil macht.

Darüber hinaus beeinflusst die Polarisierbarkeit von Atomen die Nukleophilie. Die Polarisierbarkeit beschreibt, wie leicht Elektronen in der Wolke verzerrt werden können. Ein Nukleophil mit einem großen Atom weist eine größere Polarisierbarkeit auf, was bedeutet, dass es eine höhere Elektronendichte an das Elektrophil abgeben kann als ein kleines Atom, dessen Elektronen fester gehalten werden.

Nucleophiles are electron-pair donors that take part in substitution reactions. The word comes from Greek and means nucleus-loving. Because they donate electrons, nucleophiles are also Lewis bases.

Many nucleophiles are negatively charged species, such as hydroxide, chloride, and hydrosulfide. Neutral molecules can also act as nucleophiles when they have lone pairs of electrons. Examples include ammonia, amines, water, and alcohols.

Some molecules without a lone pair can still behave as nucleophiles. Alkenes and aromatic rings can donate electrons from their bonding pi orbitals. The strength of a nucleophile is called nucleophilicity, and it describes how well the species can displace a leaving group in a substitution reaction.

In general, negatively charged species are more nucleophilic than similar neutral species. A useful rule is that a higher pKa of the conjugate acid often means a better nucleophile. For example, hydroxide, the conjugate base of water with a pKa of 15.7, is a better nucleophile than acetate, the conjugate base of acetic acid with a pKa of about 5.

Nucleophilicity also depends on the solvent. In polar protic solvents, anions are strongly solvated, which lowers their ability to react. Among halides in these solvents, fluoride is solvated the most and iodide the least, so iodide is the better nucleophile. In polar aprotic solvents, anions are less solvated and more free to attack, so basicity becomes the main guide and fluoride is the best nucleophile.

Atom size also matters because of polarizability. Polarizability is how easily an electron cloud can be distorted. Larger atoms are more polarizable, so they can donate electron density to an electrophile more easily than smaller atoms with more tightly held electrons.
