Erinnern Sie sich daran, dass bei einer nukleophilen Substitutionsreaktion ein Nukleophil seine Elektronen an ein Elektrophil abgibt.
Elektrophile sind elektronensuchende Reagenzien – entweder neutral oder positiv geladen –, die ein leeres Atomorbital oder ein niederenergetisches Antibindungsorbital enthalten.
Ein positives Elektrophil wie das Proton – mit einem leeren, niederenergetischen 1s Orbital – ist sehr reaktiv. Folglich greift ein Nukleophil wie das Hydroxidion das Proton an, neutralisiert die Ladung und bildet Wasser.
Ein weiteres positives Elektrophil – das Carbokation – hat ein leeres p-Orbital, wodurch es auf einen nukleophilen Angriff reagiert.
Ein Neutralelektrophil, wie das Lewis-Säure-Bortrifluorid, hat ein leeres p-Orbital, das Elektronen aus dem Nukleophil aufnehmen kann, wodurch eine Bindung gebildet wird und ein stabiler Komplex entsteht.
In einem neutralen Molekül wie Chlorbutan resultiert das elektrophile Zentrum aus der elektronenziehenden induktiven Wirkung des elektronegativeren Substituenten, der an die Molekülkette gebunden ist.
In einem organischen Elektrophil mit einem doppelt gebundenen elektronegativen Atom – wie der Carbonylgruppe – gibt der C=O-Bindungsdipol eine teilweise positive Ladung an das Kohlenstoffatom ab.
In einer Reaktion legt das Nukleophil seine Elektronen in das Antibindungs-Orbital mit niedrigerer Energie π Orbital des Elektrophilen ab. Infolgedessen bricht die C-O-π-Bindung und die Elektronen wandern weiter zum Sauerstoffatom. 
In einem Elektrophil wie HCl, das aus einem einfachgebundenen elektronegativen Atom besteht, zwingt der Dipol der σ Bindung die nukleophilen Elektronen, in das niedrigenergetische HCl-Antibindungsorbital σ zu wandern, und bricht somit die Bindung.
Moleküle wie Halogene, mit σ Bindungen und ohne Dipole, eignen sich ebenfalls gut als Elektrophile. Bei Brom zum Beispiel führen schlechte Überlappungen zwischen den Atomorbitalen von Bromatomen zu einer schwachen Br-Br-Bindung.
Somit greift ein Nukleophil die niedrigere Energie σ Antibindungsorbital an, bricht die Br-Br-Bindung und bildet eine neue Bindung.

In dieser Lektion werden die Definition, Klassifizierung und charakteristischen Merkmale eines Elektrophils erläutert, die Schlüsselmerkmale nukleophiler Substitutionsreaktionen sind. Eine Analyse ihrer Ladung und ihres Orbitalbildes hilft, ihre Reaktivität bei der Suche nach Elektronen zu verstehen. Elektrophile können in positive und neutrale Spezies eingeteilt werden. Andere Klassen umfassen freie Radikale und polare funktionelle Gruppen.

Während ein positives Elektrophil wie ein Proton aufgrund seines leeren, niederenergetischen 1s-Orbitals reagiert, sind die anderen positiven Elektrophile wie Carbokationen aufgrund ihres leeren p-Orbitals reaktiv.

Andererseits besitzen neutrale Elektrophile, analog zu Lewis-Säuren, leere p-Orbitale, die Elektronen vom Nukleophil aufnehmen können, um stabile Komplexe zu erzeugen. Aufgrund des elektronenziehenden induktiven Effekts in Gegenwart eines elektronegativeren Substituenten an der Molekülkette kann in einem neutralen Molekül häufig ein elektrophiles Zentrum gebildet werden. Dies erklärt die teilweise positive Ladung des Kohlenstoffatoms in einer Carbonylgruppe.

Im Rahmen einer chemischen Reaktion ist ein umfassendes Bild des Elektronentransfers in diesem Prozess notwendig. Ein Nukleophil lagert seine Elektronen im antibindenden π-Orbital niedrigerer Energie des Elektrophils ab. Im Gegensatz dazu zwingt der Dipol der σ-Bindung die nukleophilen Elektronen dazu, sich in das antibindende σ-Orbital niedrigerer Energie zu bewegen, was zum Aufbrechen der Bindung führt. Diese Phänomene werden oft am Beispiel von Carbonylgruppen und HCl erklärt. Typischerweise sind die niedrigsten besetzten Molekülorbitale (LUMOs) in organischen Elektrophilen antibindende Orbitale mit niedriger Energie, da sie mit elektronegativen Atomen assoziiert sind. Dies sind entweder π*-Orbitale oder σ*-Orbitale.

Einige Moleküle, wie zum Beispiel Halogene, sind auch gute Elektrophile. Hier schwächt eine schlechte Überlappung zwischen den Atomorbitalen der beiden Halogenide trotz des Fehlens eines Dipols die Bindung und macht sie anfälliger für einen nukleophilen Angriff. Dies führt zu der anderen Klassifizierung von starken und schwachen Elektrophilen, die in späteren Lektionen behandelt wird. Electrophile reactivity in organic reactions depends on how a species attracts electrons. Electrophiles are electron-poor and are key partners in nucleophilic substitution reactions. Their charge and orbital structure help explain why they react the way they do.

Electrophiles can be positive or neutral species. A positive electrophile such as a proton reacts because it has a vacant, low-energy 1s orbital. Other positive electrophiles, such as carbocations, are reactive because they have an empty p orbital.

Neutral electrophiles act like Lewis acids. They have empty p orbitals that can accept electrons from a nucleophile and form stable complexes. In some neutral molecules, an electrophilic center appears because an electronegative substituent pulls electron density away through the inductive effect. This explains the partial positive charge on the carbon atom in a carbonyl group.

During a reaction, electron transfer must be viewed as a whole. A nucleophile donates electrons into the lower-energy antibonding pi orbital of the electrophile. In other cases, the dipole of a sigma bond drives the nucleophilic electrons into a lower-energy antibonding sigma orbital, which leads to bond breaking. Carbonyl groups and HCl are common examples of these ideas.

In organic molecules, the lowest unoccupied molecular orbitals, or LUMOs, are often low-energy antibonding orbitals. These are usually pi-star or sigma-star orbitals, especially when electronegative atoms are involved. Halogens can also act as electrophiles. Even without a dipole, poor overlap between the two halide atomic orbitals weakens the bond and makes it easier for a nucleophile to attack. In many cases, molecules with a single or double bond linked to O, N, Cl, or Br make good electrophiles.
