Sie erklären jedoch nicht genau, wie sich die Moleküle während der Reaktion anordnen. Um den vollständigen Mechanismus einer S N 2-Reaktion abzuleiten, betrachten Sie die folgendenTheorien.
Zunächst enthält das Substrat ein elektronegatives Halogen, das eine polarisierte Halogenkohlenstoff-Bindung erzeugt. Dies führt zu einem elektrophilen Zentrum am Kohlenstoff, das das Nukleophil mit seinem einsamen Elektronenpaar anzieht.
Das Vorhandensein einer hohen Elektronendichte um das Halogenid herum blockiert jedoch effektiv den Frontside-Angriff. Somit nähert sich das Nukleophil dem Elektrophil von der Seite, die der verlassenden Gruppe gegenüberliegt, was zu einem Backside-Angriff führt.
Wenn das Nukleophil sein einzelnes Paar an das Elektrophil abgibt, zieht sich die verlassende Gruppe mit dem an den Kohlenstoff gebundenen Elektronenpaar zurück. Dies führt zu einem Übergangszustand, in dem die Bindungsbildung zwischen dem Nukleophil und dem Substrat und der Bindungsbruch zwischen dem Substrat und der Abgangsgruppe gleichzeitig stattfinden.
Der Übergangszustand ist sehr instabil. Um die Stabilität wiederherzustellen, verlässt die verlassende Gruppe das Elektronenpaar in einer konzertierten Weise, was zu einer Umkehrung der Substratkonfiguration führt.
Die Molekülorbitaltheorie unterstützt den Backside-Angriff zusätzlich. Das einzige Elektronenpaar des Nukleophils nimmt das höchste Molekülorbital (HOMO) ein. Um eine Bindung zu bilden, muss sich der HOMO mit dem kleinsten freien Molekülorbital (LUMO) des Elektrophils überlappen.
Wenn sich ein Nukleophil dem Elektrophil von der gleichen Seite wie die verlassende Gruppe nähert, steht es einem Knoten gegenüber, was dazu führt, dass sich der HOMO mit dem bindenden und antibindenden LUMO überlappt. Es bildet sich jedoch keine Bindung, da die Antibonding-Überlappung die Bonding-Überlappung aufhebt.
Im Gegensatz dazu überlappt der Rückseitenansatz des Nukleophils effizient den HOMO mit dem LUMO des Elektrophils, was zur Bindungsbildung führt.
BeideTheoriensprechen also dafür, dass der S N 2-Reaktionsmechanismus aufeinander abgestimmt ist, bei dem das Nukleophil von der Rückseite angreift, während es gleichzeitig die Abgangsgruppe verdrängt und eine Umkehrung der Konfiguration verursacht.

Die kinetischen Studien von SN2-Reaktionen legen ein wesentliches Merkmal ihres Mechanismus nahe: Es handelt sich um einen einstufigen Prozess ohne Zwischenprodukte. Dabei sind sowohl das Nukleophil als auch das Substrat am geschwindigkeitsbestimmenden Schritt beteiligt.

Das Vorhandensein des elektronegativeren Halogens im Substrat erzeugt eine polarisierte Kohlenstoff-Halogenid-Bindung. Das Halogenid zieht die Elektronenwolke an und erzeugt ein elektrophiles Zentrum am Kohlenstoffatom. Somit trägt das Kohlenstoffatom eine teilweise positive Ladung, während das Halogenid eine teilweise negative Ladung trägt. Der elektrophile Kohlenstoff zieht das Nukleophil mit seinem freien Elektronenpaar an.

Allerdings blockiert die hohe Elektronendichte um das Halogenid effektiv den Angriff des Nukleophils auf der gleichen Seite. Somit nähert sich das Nukleophil dem Elektrophil von der elektronenarmen Seite des Substrats, was zu einem rückseitigen Angriff führt. Daher spendet das Nukleophil sein freies Elektronenpaar an den elektrophilen Kohlenstoff, 180° von der Abgangsgruppe entfernt.

Wenn sich das Halogenid vom elektrophilen Kohlenstoff löst, bewegt es sich zusammen mit dem an den Kohlenstoff gebundenen Elektronenpaar weg. Dies führt zu einem Übergangszustand mit einer teilweise gebildeten Bindung zwischen Nukleophil und Substrat und einer teilweise gebrochenen Bindung zwischen Substrat und Abgangsgruppe.

Mit drei festen und zwei Teilbindungen am Kohlenstoff ist der Übergangszustand äußerst instabil. Somit verlässt die Abgangsgruppe das an den Kohlenstoff gebundene Elektronenpaar, was zu einer Umkehrung der Substratkonfiguration führt. (Abbildung 1)

Darüber hinaus unterstützt die Molekülorbitaltheorie auch den Rückseitenangriff. Wenn sich das Bindungsorbital des Nukleophils, d. h. das höchstbesetzte Molekülorbital (HOMO), dem niedrigsten unbesetzten Molekülorbital (LUMO) des Substrats von der gleichen Seite wie die abgehende Gruppe nähert, steht es einem Knoten gegenüber, der sowohl die Bindungs- als auch die Anti-Bindungsüberlappung aufhebt. Im Gegensatz dazu überlappt ein Rückseitenangriff des Nukleophils effizient mit dem LUMO des Substrats, was zur Bindungsbildung führt.

Somit läuft der SN2-Mechanismus in einem einzigen Schritt ab, bei dem das ankommende Nukleophil mit dem Substrat aus einer Richtung reagiert, die der verdrängten Abgangsgruppe entgegengesetzt ist.

SN2 reactions happen in one step and end with inversion of configuration. The nucleophile and the substrate both take part in the rate-determining step, and no intermediate forms.

The substrate contains a polarized carbon-halide bond. The more electronegative halogen pulls the electron cloud toward itself. That leaves the carbon atom electrophilic, or electron-poor, with a partial positive charge. The halide has a partial negative charge.

The electrophilic carbon attracts the nucleophile, which brings a lone pair of electrons. But the electron-rich halide blocks attack from the same side. For that reason, the nucleophile approaches from the opposite side of the leaving group. This is called a backside attack.

In the transition state, the nucleophile is partly bonded to the carbon while the carbon-halide bond is partly broken. The carbon has three solid bonds and two partial bonds, so this state is very unstable. As the halide leaves with the electron pair, the substrate flips and its configuration inverts.

Molecular orbital theory also supports this path. The nucleophile’s highest occupied molecular orbital, or HOMO, matches poorly with the substrate’s lowest unoccupied molecular orbital, or LUMO, from the same side because of a node. A backside attack gives better overlap and allows bond formation. So the SN2 mechanism is a single-step reaction in which the nucleophile attacks opposite the leaving group and displaces it.
