Die Stereochemie der radikalischen Halogenierung hängt davon ab, ob das reagierende Molekül chiral oder achiral ist.
Zum Beispiel ergibt achirales n-Butan bei radikaler Chlorierung 1- und 2-Chlorbutan. Durch die Reaktion wird ein neues chirales Zentrum eingeführt, da eines der Produkte eine racemische Mischung ist.
Die Bildung eines racemischen Gemisches wird durch die Erzeugung eines achiralen trigonalen planaren Radikalzwischenprodukts angetrieben, auf das Chlor von beiden Seiten angreifen kann, wodurch gleiche Mengen an R- und S-Enantiomeren entstehen.
Dies macht Kohlenstoff-2-Wasserstoffatome in der Natur zu enantiotopischen Wasserstoffatomen.
In ähnlicher Weise bildet auch die radikalische Halogenierung an einem bestehenden chiralen Zentrum ein racemisches Gemisch.
Hier verliert das radikalische Zwischenprodukt die Konfiguration des Reaktanten und wird achiral, wodurch der Halogenangriff von beiden Seiten ermöglicht wird und gleiche Mengen an Enantiomeren erzeugt werden.
Im Gegensatz dazu ergibt eine chirale Verbindung mit einem chiralen Kohlenstoff, der nicht an der radikalischen Halogenierung beteiligt ist, diastereomere Produkte, wodurch ein zweites chirales Zentrum eingeführt wird.
Das vorhandene chirale Zentrum macht das trigonale planare Radikal intermediär chiral. Folglich greift Chlor an einer Seite stärker an als an der anderen und erzeugt chirale, diastereomere Produkte in ungleichen Mengen.

Halogenierung zur Bildung eines neuen chiralen Zentrums:

So entstehen beispielsweise bei der radikalischen Halogenierung von Butan die Produkte 2-Chlorbutan und 1-Chlorbutan, wobei ersteres ein chirales Zentrum aufweist. 2-Chlorbutan wird jedoch aufgrund der trigonal-planaren Struktur des gebildeten radikalischen Zwischenprodukts als racemisches Gemisch erhalten.

Erfolgt die radikalische Halogenierung an einem vorhandenen chiralen Kohlenstoff, entsteht ein 1:1-Verhältnis der Enantiomere. Dies liegt auch an der trigonal-planaren Struktur des radikalischen Zwischenprodukts, bei dem die Chiralität des Ausgangsmaterials verloren gegangen ist.

Erfolgt die radikalische Halogenierung eines Enantiomers an einer anderen Position als dem chiralen Kohlenstoff, entstehen Diastereomere. In diesem Fall wird auch das trigonal-planare radikalische Zwischenprodukt gebildet, ohne dass das vorhandene chirale Zentrum beeinträchtigt wird. Dieses chirale Zentrum unterscheidet die beiden Flächen des radikalischen Zwischenprodukts, so dass das Halogen ungleichmäßig an zwei Flächen angreift, was zur Bildung ungleicher Mengen an Diastereomeren führt.

Radical halogenation can change the chirality of a molecule, which is the three-dimensional arrangement of its atoms. The products depend on whether the reaction creates a new chiral center, reacts at an existing chiral center, or adds a second chiral center.

When radical halogenation forms a new chiral center, the product can be a racemic mixture. For example, halogenation of butane gives 2-chlorobutane and 1-chlorobutane. The 2-chlorobutane product has a chiral center, but it is formed as a racemic mixture because the radical intermediate is trigonal planar, or flat in shape.

If halogenation happens at an existing chiral carbon, the result is a 1:1 ratio of enantiomers. Enantiomers are mirror-image forms of the same molecule. Here, the trigonal planar radical intermediate removes the original chirality of the starting material.

Radical halogenation can also generate a second chiral center. In that case, diastereomers form. The radical intermediate is still trigonal planar, but the existing chiral center is not changed. That chiral center makes the two faces of the radical intermediate different, so halogen attack is unequal and the diastereomers form in unequal amounts.
