Nucleic Acids

Nukleinsäuren sind Polymere von Nukleotiden – Moleküle, die aus einem Pentosezucker, einer stickstoffhaltigen Base und einer Phosphatgruppe bestehen.
Es gibt zwei Arten von Nukleinsäuren: Desoxyribonukleinsäure, DNA, und Ribonukleinsäure, RNA. 
Ihre chemische Struktur unterscheidet sich je nachdem, welche Pentose, Zucker und stickstoffhaltigen Basen sie enthalten.
Der Pentosezucker in RNA ist Ribose, die eine Hydroxylgruppe hat, die an Kohlenstoff-2 gebunden ist. Der Zucker in der DNA ist Desoxyribose, die nur ein Wasserstoffatom, aber keinen Sauerstoff an Kohlenstoff-2 hat.
Die stickstoffhaltige Base ist an Kohlenstoff-1 und das Phosphat an Kohlenstoff-5 gebunden. Sowohl RNA als auch DNA enthalten die Basen Adenin, Cytosin und Guanin; aber DNA hat Thymin, während RNA Uracil hat. 
In DNA und RNA bilden Guanin und Cytosin komplementäre Basenpaare, die durch drei Wasserstoffbrückenbindungen verbunden sind. Adenin und Thymin bilden Basenpaare in der DNA, während Adenin und Uracil in RNA paaren, die beide durch zwei Wasserstoffbrückenbindungen miteinander verbunden sind.
Verschiedene DNA- oder RNA-Polymerase-Enzyme katalysieren die Polymerisation von Nukleotiden.
Eine Phosphodiesterbindung wird zwischen einer Hydroxylgruppe, die an Kohlenstoff-3 gebunden ist, und der Phosphatgruppe, die an Kohlenstoff-5 des nächsten Nukleotids gebunden ist, gebildet. Diese Reaktion hinterlässt ein ungebundenes 5'-Ende mit einer freien Phosphatgruppe und ein ungebundenes 3'-Ende mit einer freien Hydroxylgruppe.
Wenn sie mit einem komplementären Strang gepaart sind, sind die beiden Moleküle antiparallel, was bedeutet, dass das 5'-Ende eines Strangs mit dem 3'-Ende des anderen paart.
Die Stränge werden durch intermolekulare Kräfte zusammengehalten, darunter hydrophobe Effekte, Van-der-Waals-Wechselwirkungen und die spezifischen Wasserstoffbrückenbindungen, die sich zwischen den stickstoffhaltigen Basen bilden. DNA ist eine Doppelhelix, die aus zwei Polynukleotidketten besteht, die umeinander gewickelt sind. Im Gegensatz dazu kommt RNA oft als einzelsträngiges Molekül vor.
RNA kann jedoch an eine komplementäre RNA oder DNA binden. Es kann auch eine komplementäre Basenpaarung innerhalb des Strangs aufweisen, was zu verschiedenen Arten von RNA-Sekundärstrukturen führt, die unterschiedliche Funktionen innerhalb der Zelle haben.

DNA und RNA Die beiden Haupttypen von Nukleinsäuren sind Desoxyribonukleinsäure (DNA) und Ribonukleinsäure (RNA). DNA ist das genetische Material in allen lebenden Organismen, von einzelligen Bakterien bis hin zu mehrzelligen Säugetieren. Es befindet sich im Kern von Eukaryoten sowie in den Organellen, Chloroplasten und Mitochondrien. Bei Prokaryoten ist die DNA nicht von einer Membranhülle umgeben. Der gesamte genetische Inhalt der Zelle ist ihr Genom, und die Untersuchung von Genomen ist Genomik. In eukaryotischen Zellen, jedoch nicht in Prokaryoten, bildet DNA einen Komplex mit Histonproteinen, um Chromatin zu bilden, die Substanz der eukaryotischen Chromosomen. Ein Chromosom kann Zehntausende Gene enthalten. Viele Gene enthalten die Informationen zur Herstellung von Proteinen. Andere Gene kodieren für RNA-Produkte. Die DNA steuert alle zellulären Aktivitäten, indem sie die Gene „ein“ oder „ausschaltet“. Die andere Art von Nukleinsäure, die RNA, ist hauptsächlich an der Proteinsynthese beteiligt. Die DNA-Moleküle verlassen den Zellkern nie, sondern kommunizieren über einen Vermittler mit dem Rest der Zelle. Dieser Vermittler ist die Messenger-RNA (mRNA). Andere Arten von RNA – wie rRNA, tRNA und microRNA – sind an der Proteinsynthese und ihrer Regulierung beteiligt. DNA und RNA bestehen aus Monomeren, die Nukleotide genannt werden. Jedes Nukleotid besteht aus drei Komponenten: einer stickstoffhaltigen Base, einem Pentosezucker (fünf Kohlenstoffatome) und einer Phosphatgruppe. Jede stickstoffhaltige Base in einem Nukleotid ist an ein Zuckermolekül gebunden, das an eine oder mehrere Phosphatgruppen gebunden ist. Die stickstoffhaltigen Basen, wichtige Bestandteile von Nukleotiden, sind organische Moleküle und werden so genannt, weil sie Kohlenstoff und Stickstoff enthalten. Sie sind Basen, weil sie eine Aminogruppe enthalten, die das Potenzial hat, zusätzlichen Wasserstoff zu binden und so die Wasserstoffionenkonzentration in ihrer Umgebung zu verringern, wodurch sie basischer wird. Jedes Nukleotid in der DNA enthält eine von vier möglichen stickstoffhaltigen Basen: Adenin (A), Guanin (G), Cytosin (C) und Thymin (T). Adenin und Guanin werden zu den Purinen gezählt. Die Primärstruktur des Purins besteht aus zwei Kohlenstoff-Stickstoff-Ringen. Cytosin, Thymin und Uracil werden als Pyrimidine klassifiziert, deren Primärstruktur einen einzelnen Kohlenstoff-Stickstoff-Ring aufweist. An jeden dieser grundlegenden Kohlenstoff-Stickstoff-Ringe sind unterschiedliche funktionelle Gruppen gebunden. In der Kurzform der Molekularbiologie kennen wir die stickstoffhaltigen Basen an ihren Symbolen A, T, G, C und U. DNA enthält A, T, G und C; RNA hingegen A, U, G und C. Der Pentosezucker in der DNA ist Desoxyribose und in der RNA ist der Zucker Ribose. Der Unterschied zwischen den Zuckern besteht im Vorhandensein der Hydroxylgruppe am zweiten Kohlenstoffatom der Ribose und des Wasserstoffs am zweiten Kohlenstoffatom der Desoxyribose. Der Phosphatrest bindet an die Hydroxylgruppe des 5‘-Kohlenstoffs eines Zuckers und an die Hydroxylgruppe des 3‘-Kohlenstoffs des Zuckers des nächsten Nukleotids, wodurch eine 5‘-3‘-Phosphodiesterbindung entsteht. DNA-Doppelhelix-Struktur DNA hat eine Doppelhelix-Struktur. Zucker und Phosphat liegen an der Außenseite der Helix und bilden das Rückgrat der DNA. Die stickstoffhaltigen Sockel sind im Inneren wie zwei Treppenstufen gestapelt. Wasserstoffbrückenbindungen binden die Paare aneinander. Jedes Basenpaar in der Doppelhelix ist vom nächsten Basenpaar 0,34 nm entfernt. Die beiden Stränge der Helix verlaufen in entgegengesetzte Richtungen, was bedeutet, dass das 5‘-Kohlenstoffende eines Strangs dem 3‘-Kohlenstoffende des entsprechenden Strangs gegenüberliegt. Es sind nur bestimmte Arten der Basenpaarung zulässig: A kann mit T gepaart werden und G kann mit C gepaart werden. Dies ist die komplementäre Basenregel. Mit anderen Worten: Die DNA-Stränge sind zueinander komplementär. RNA Ribonukleinsäure oder RNA ist hauptsächlich am Prozess der Proteinsynthese unter der Leitung der DNA beteiligt. RNA ist normalerweise einzelsträngig und besteht aus Ribonukleotiden, die durch Phosphodiesterbindungen verbunden sind. Es gibt vier Haupttypen von RNA: Messenger-RNA (mRNA), ribosomale RNA (rRNA), Transfer-RNA (tRNA) und microRNA (miRNA). Die erste, mRNA, trägt die Botschaft der DNA, die alle zellulären Aktivitäten in einer Zelle steuert. Die RNA-Basensequenz ist komplementär zur kodierenden Sequenz der DNA, von der sie kopiert wurde. Im Zytoplasma interagiert die mRNA mit Ribosomen und anderen zellulären Maschinen. Die mRNA wird in Sätzen von drei Basen gelesen, die als Codons bekannt sind. Jedes Codon kodiert für eine einzelne Aminosäure. Auf diese Weise wird die mRNA abgelesen und das Proteinprodukt hergestellt. Ribosomale RNA (rRNA) ist ein Hauptbestandteil von Ribosomen, an die die mRNA bindet. Die rRNA sorgt für die richtige Ausrichtung der mRNA und der Ribosomen. Die rRNA des Ribosoms hat auch eine enzymatische Aktivität (Peptidyltransferase) und katalysiert die Bildung von Peptidbindungen zwischen zwei ausgerichteten Aminosäuren. Transfer-RNA (tRNA) ist eine der kleinsten der vier Arten von RNA, normalerweise 70–90 Nukleotide lang. Es transportiert die richtige Aminosäure zur Proteinsynthesestelle. Es ist die Basenpaarung zwischen tRNA und mRNA, die es der richtigen Aminosäure ermöglicht, sich in die Polypeptidkette einzufügen. MicroRNAs sind die kleinsten RNA-Moleküle und ihre Rolle besteht darin, die Genexpression zu regulieren, indem sie die Expression bestimmter mRNA-Nachrichten stört. Obwohl die RNA einzelsträngig ist, weisen die meisten RNA-Typen eine ausgedehnte intramolekulare Basenpaarung zwischen komplementären Sequenzen auf, wodurch eine vorhersehbare dreidimensionale Struktur entsteht, die für ihre Funktion unerlässlich ist.
