pH Regulation in Cells

Der pH-Wert ist ein Maß für die H+-Konzentration in einer Lösung und gibt den Säuregehalt oder die Alkalität an.
Die pH-Skala reicht von 0 bis 14, wobei sieben als neutral, ein Wert unter sieben als sauer und ein Wert über sieben als basisch gilt.
Der typische zytoplasmatische pH-Wert liegt in einem leicht alkalischen Bereich von 7,0 bis 7,4. Große Schwankungen dieses intrazellulären pH-Werts können kritische zelluläre Prozesse wie den Stoffwechsel, das Membranpotenzial und die Signalwege beeinflussen.
So verbindet sich beispielsweise CO2 – das Endprodukt der Zellatmung – mit H2O zu H2CO3 .
H2CO3 ist eine schwache Säure, die leicht dissoziiert, das Zytoplasma langsam ansäuert und zu zellulärem Stress führt.
Daher regulieren die Zellen ihren internen pH-Wert mithilfe von Ionenaustauschern, um die Wasserstoffionenkonzentration zu verändern.
Der Na+/H+-Austauscher hilft, den intrazellulären pH-Wert auszugleichen, indem er den Ausfluss von H+ mit dem Einstrom von Na+ koppelt.
In ähnlicher Weise spielen Cl-HCO3−-Austauscher eine wichtige Rolle bei der Aufrechterhaltung des intrazellulären pH-Werts, indem sie HCO-3−-Ionen im Austausch gegen Cl−-Ionen ausstoßen.

Der pH-Wert spielt eine entscheidende Rolle bei der Aufrechterhaltung normaler Zellaktivitäten. Es hilft, die Struktur und Funktion verschiedener Proteine aufrechtzuerhalten, bestimmt die Ladung der Zellmembranen und ist entscheidend für Stoffwechselreaktionen innerhalb der Zelle. Darüber hinaus nutzen Zellen die Energie der Protonenantriebskraft, um ATP zu erzeugen.

Zytosolischer pH-Wert

Unter physiologischen Bedingungen ist der zytosolische pH-Wert etwas saurer als der extrazelluläre pH-Wert. Allerdings müssen Zellen eine weitere Ansäuerung ihres Zytosols verhindern, um ihr Membranpotential aufrechtzuerhalten und eine normale Funktion zu gewährleisten. Daher verwenden sie mehrere spezialisierte Protonentranslokationsmaschinen – Antiporter, Symporter und protonenpumpende ATPasen –, um den pH-Wert im Steady-State genau zu regulieren.

Zellorganellen und pH-Homöostase

Die Kompartimentierung innerhalb der eukaryontischen Zellen trägt dazu bei, unterschiedliche Umgebungsbedingungen bereitzustellen, um bestimmte Funktionen innerhalb verschiedener membranumschlossener Organellen auszuführen. Der interne pH-Wert in diesen verschiedenen Kompartimenten schwankt stark, ist jedoch ein wichtiger Faktor für deren effektive Funktion.

Beispielsweise strömen Cytochrome, die sich auf der inneren Mitochondrienmembran befinden, Protonen aus, indem sie Energie aus dem Elektronenfluss nutzen. Dies trägt dazu bei, einen Protonengradienten über die Mitochondrienmembran aufzubauen, der zur Erzeugung chemischer Energie in Form von ATP verwendet wird. In ähnlicher Weise halten Lysosomen einen internen sauren pH-Wert aufrecht, indem sie Protonen aus dem Zytosol pumpen und dabei Energie aus der ATP-Hydrolyse nutzen. Dies ist für die Funktion der lysosomalen Enzyme unerlässlich.

Allerdings müssen einige Organellen, einschließlich des Zellkerns, des endoplasmatischen Retikulums und der Peroxisomen, ihren internen pH-Wert im Gleichgewicht mit dem Zytoplasma halten. Daher verfügen sie nicht über intrinsische pH-regulierende Systeme und sind für Protonen sehr durchlässig.

Fehlregulation des intrazellulären pH-Werts

Bestimmte metabolische oder genetische Bedingungen können den stabilen pH-Wert des Zytoplasmas oder der Organellen innerhalb der Zelle stören und zu Krankheiten führen. Beispielsweise wurden Mutationen in Chloridträgern wie CLCN6 und CLCN7, die auf späten Endosomen und Lysosomen vorhanden sind, mit Osteoporose und lysosomaler Speicherkrankheit in Verbindung gebracht. In ähnlicher Weise behindern Mutationen im CLCN5-Kanal die luminale Ansäuerung und die Produktion früher Endosomen in Nierenepithelzellen und können schließlich zu Nierenversagen führen.s

Cell pH and organelle balance are essential for normal cell function. pH affects protein shape and activity, the charge on cell membranes, and many metabolic reactions inside the cell. Cells also use the proton motive force, which is the energy stored in a proton gradient, to make ATP, the cell’s main energy molecule.

The cytosol, or fluid inside the cell, is usually slightly more acidic than the fluid outside the cell. Even so, cells must stop the cytosol from becoming too acidic. They use proton-translocating machinery, including antiporters, symporters, and proton-pumping ATPases, to keep steady-state pH under control and preserve membrane potential.

Eukaryotic cells also separate functions into membrane-bound organelles with different internal pH levels. This compartmentalization creates the conditions needed for each organelle to work properly. In mitochondria, cytochromes on the inner membrane move protons using energy from electron flow. This builds the proton gradient that helps produce ATP. Lysosomes keep an acidic interior by pumping protons from the cytosol with energy from ATP hydrolysis, which is needed for lysosomal enzymes to function.

Some organelles need their internal pH to stay close to the cytoplasm. These include the nucleus, endoplasmic reticulum, and peroxisomes. They do not have their own pH-control systems and are highly permeable to protons, so their pH stays in equilibrium with the cytosol.

Problems with intracellular pH can cause disease. Changes in metabolism or gene function can disturb the steady-state pH of the cytoplasm or organelles. Mutations in chloride carriers such as CLCN6 and CLCN7, which are found on late endosomes and lysosomes, have been linked to osteoporosis and lysosomal storage disease. Mutations in the CLCN5 channel can block luminal acidification and early endosome formation in renal epithelial cells, and this can eventually lead to kidney failure.
