Zellen interagieren mit einem Netzwerk lokal sezernierter Moleküle, das die Plasmamembran umgibt, die sogenannte extrazelluläre Matrix oder EZM.
Diese Wechselwirkungen werden durch große multimolekulare Komplexe, sogenannte Zell-Matrix-Adhäsionskomplexe, vermittelt, die breit verteilt oder in unterschiedlichen Flecken auf der Plasmamembran geclustert sind, die als Zell-Matrix-Verbindungen bezeichnet werden.
Jeder Adhäsionskomplex besteht aus Transmembranrezeptoren und zytosolischen Adapterproteinen, die die Zytoskelettfilamente mit Liganden in der EZM verbinden.
Integrin, der bekannteste Adhäsionsrezeptor, und sein Adapter Talin können jede der drei Hauptklassen von EZM-Molekülen binden: Proteoglykane, die die Zelle polstern, Kollagen, das für mechanische Festigkeit sorgt, und multiadhäsive lösliche Proteine wie Fibronektin, die Adhäsionsrezeptoren mit anderen Matrixkomponenten vernetzen.
Integrine überbrücken die Zytoskelett-Filamente mit einer speziellen Form der EZM an der Basallamina, die die äußere Oberfläche der Zellmembran auskleidet.
Der resultierende Adhäsionskomplex interagiert mit den EZM-Molekülen und sendet mechanische und chemische Signale, um das Zellwachstum, die Migration und die Differenzierung zu regulieren.

Overview of Cell-Matrix Interactions

Die extrazelluläre Matrix oder ECM hält Zellen zu einem Gewebe zusammen und ermöglicht die Kommunikation der Zellen innerhalb des Gewebes. ECM umfasst Proteine wie Fibronektin, Kollagen, Laminin usw. Das am häufigsten vorkommende Protein in diesem Bereich ist Kollagen. Kollagenfasern sind mit kohlenhydrathaltigen Proteinmolekülen, sogenannten Proteoglykanen, verwoben. ECM ermöglicht die Zellmigration und stellt ein strukturelles Gerüst bei der Zelladhäsion bereit, das die Zelle verankert, wenn die extrazellulären Matrixproteine mit Transmembranproteinrezeptoren interagieren.

Ein ECM-Ligand bindet an den Rezeptor und ändert dessen Konformation. Der Rezeptor wiederum verändert die Konformation der Mikrofilamente, die direkt unter der Plasmamembran positioniert sind. Diese Konformationsänderungen induzieren chemische Signale innerhalb der Zelle, die den Zellkern erreichen und die Transkription bestimmter DNA-Abschnitte „an“ oder „aus“ schalten, was sich auf die Produktion assoziierter Proteine auswirkt und so die Zellaktivitäten verändert.

The extracellular matrix, or ECM, is the material that helps hold cells together in a tissue. It also lets cells in that tissue communicate with one another. The ECM contains proteins such as fibronectin, collagen, and laminin. Collagen is the most abundant protein in this space.

Collagen fibers are woven together with proteoglycans, which are protein molecules with carbohydrates attached. This mix gives the ECM its structure. It also supports cell migration and provides a scaffold for cell adhesion. At these adhesion sites, ECM proteins connect with transmembrane protein receptors and help anchor the cell.

When an ECM ligand binds to a receptor, the receptor changes shape. This change also affects the microfilaments just under the plasma membrane. The shape changes trigger chemical signals inside the cell that travel to the nucleus. There, they can turn specific DNA sections on or off and change the proteins the cell makes.

These signals can alter cellular activities. Integrins are the main transmembrane receptors that mediate cell-matrix junctions such as focal adhesions and hemidesmosomes. On the inside of the cell, integrins bind to actin and intermediate filaments. On the ECM side, they bind to fibronectin and collagen.
