Integrine sind eine Familie von Zelladhäsionsrezeptoren, die Liganden in der extrazellulären Matrix oder EZM binden, um Zell-Matrix-Adhäsionen zu vermitteln.

Integrin besteht aus nichtkovalent assoziierten Alpha- und Beta-Glykoprotein-Untereinheiten, die ein Transmembran-Heterodimer bilden.
Der intrazelluläre C-terminale Schwanz der beta-Untereinheit interagiert mit Aktinfilamenten im Zytosol über Adapterproteine wie Talin und Paxillin. Die großen N-terminalen extrazellulären Domänen von Integrin binden an spezifische Aminosäuresequenzmotive in EZM-Proteinen, wie z. B. die RGD-Sequenz in Fibronektin.
Der gebildete Adhäsionskomplex leitet Signale mit Hilfe von Effektorproteinen bidirektional über die Plasmamembran weiter und erleichtert so die Beweglichkeit, das Wachstum und das Überleben der Zelle.

Tier- und Protozoenzellen haben keine Zellwände, die zur Formerhaltung und strukturellen Stabilität beitragen. Stattdessen sezernieren diese eukaryotischen Zellen eine klebrige Masse aus Kohlenhydraten und Proteinen in die Räume zwischen benachbarten Zellen. Dieses Netzwerk aus Proteinen und Molekülen wird als extrazelluläre Matrix oder ECM bezeichnet.

Einige ECM-Proteine ​​lagern sich zu einer Basalmembran zusammen, an der die übrigen Komponenten haften. Proteoglykane bilden typischerweise den Großteil der ECM, während faserige Proteine wie Kollagen für Festigkeit sorgen. Sowohl Proteoglykane als auch Kollagen sind an Fibronektinproteine gebunden, die wiederum an integrale Membranproteine, sogenannte Integrine, gebunden sind.

Integrine von Säugetieren sind heterodimere Transmembran-Adhäsionsrezeptoren, die α- und β-Glykoprotein-Untereinheiten umfassen. Achtzehn verschiedene Gene der α-Kette und acht verschiedene Gene der β-Kette dimerisieren in verschiedenen Kombinationen, was zu 24 Arten von Integrinen führt, die für Wirbeltiere spezifisch sind. Sie durchspannen die Membran und interagieren mit Proteinen im Zytosol und der ECM. Die α-Untereinheit enthält eine siebenflügelige β-Propeller-Kopfdomäne, während die β-Untereinheit eine I-ähnliche Kopfdomäne aufweist; Zusammen binden sie Liganden. Der Ligandenbindungskopf ist mit zwei beinartigen Strukturen verbunden, eine von jeder Untereinheit. Integrins help animal and protozoan cells attach to the extracellular matrix, or ECM. These cells do not have cell walls, so the ECM helps maintain shape and provide structural stability. The ECM is a sticky network of carbohydrates and proteins found between adjacent cells.

Some ECM proteins form a basement membrane, and other ECM parts adhere to it. Proteoglycans make up much of the ECM, while fibrous proteins such as collagen give it strength. Fibronectin connects proteoglycans and collagen to integral membrane proteins called integrins.

Integrins are heterodimeric transmembrane adhesion receptors in mammals. That means each receptor has two different glycoprotein subunits, an alpha subunit and a beta subunit. Eighteen alpha-chain genes and eight beta-chain genes can pair in different ways to make 24 vertebrate integrins.

These receptors span the cell membrane and link the ECM to the cytosol. The alpha subunit has a seven-bladed beta-propeller head domain, and the beta subunit has an I-like head domain. Together, these head domains bind ligand, and the two leg-like parts connect the head to the membrane.

The beta subunit also interacts with a cytoskeletal protein. This helps integrins transmit signals into and out of the cell. Their structure and connections make them important for adhesion and communication in cells that rely on the ECM.
