Cluster von Integrinen auf der Zellmembran können extrazelluläre Matrixliganden vorübergehend mit dem Zytoskelett verbinden, um fokale Adhäsionskomplexe zu bilden.
Fokale Adhäsionen können wachsen und schrumpfen, so dass die Zelle während der Migration kriechen kann.
Während die Adhäsion von der Zellmembran ausgeht, werden Signale von Proteinkinasen und G-Proteinen in einer Kaskade durch die Zelle übertragen.
Zum Beispiel aktiviert Thrombin, ein extrazelluläres Signalmolekül in Blutplättchen, den G-Protein-gekoppelten Rezeptor cAR1/cAR3, der wiederum die Rap1-GTPase aktiviert.
Aktives Rap1 interagiert mit einem Rap1-GTP-interagierenden Adaptermolekül oder RIAM, das inaktives Talin an die Zellmembran rekrutiert.
Die N-terminalen Kopf- und C-terminalen Schwanzdomänen von inaktivem Talin sind zusammengefaltet. Die Bindung eines Phosphoinositids in die Zellmembran stört die Kopf-Schwanz-Interaktion und hilft dabei, Talin zu entfalten.
Zusammen mit Kindlin aktiviert Talin Integrin und bindet andere Adapterproteine wie Vinculin, die Integrin an die Aktinfilamente binden.

Integrine fungieren sowohl als extrazelluläre Eingangsempfänger als auch als intrazelluläre Verarbeitungsaktivatoren. Wie der Name schon sagt, sind Integrine vollständig in die Membranstruktur integriert. Ihre hydrophoben membrandurchspannenden Regionen interagieren mit der hydrophoben Region der Phospholipiddoppelschicht. Diese Membranrezeptoren bieten extrazelluläre Bindungsstellen für Effektoren wie Hormone und Wachstumsfaktoren. Sie aktivieren intrazelluläre Reaktionskaskaden, wenn ihre Effektoren gebunden und aktiv sind.

Einige Integrin-vermittelte Stimulationswege induzieren die Zellproliferation und fördern das Zellüberleben. Die Adhäsionen entstehen an der Zellmembran, die Signale werden jedoch durch G-Proteine und Proteinkinasen in einer Kaskade durch die Zelle übertragen. Beispielsweise löst die Bindung von Thrombin an G-Protein-gekoppelte Rezeptoren eine intrazelluläre Signalübertragung zur Aktivierung von Integrinen in Blutplättchen aus. Aktivierte Integrine binden an RGD-Peptidsequenzen auf Blutproteinen wie Fibrinogen. Wenn die Fibrinogene beginnen, Brücken zu bilden, häufen sich die fokalen Adhäsionen und die Blutplättchen aggregieren.

Integrine können auch Signalproteine direkt binden und die Signale in die Zelle weiterleiten. Im Ras/MAP-Kinase-Weg ist die fokale Adhäsionskinase (FAK), eine zytoplasmatische Proteinkinase, der am besten untersuchte Modus der Integrin-Signalisierung. Studien deuten darauf hin, dass fokale Adhäsionen viele Tyrosinphosphorylierungsstellen und somit tyrosinphosphorylierte Proteine wie FAK enthalten. Integrine sammeln sich an Zell-Matrix-Übergängen und die β-Untereinheit rekrutiert FAK über Adapterproteine wie Paxillin oder Talin, die an die α-Untereinheit gebunden sind. Die FAK-Proteine bündeln sich an der Stelle und phosphorylieren eine bestimmte Tyrosinstelle aneinander. Dadurch entsteht eine Phospho-Tyrosin-Andockstelle für Src-Proteine, die zur Familie der zytoplasmatischen Tyrosinkinasen gehören. Der FAK phosphoryliert zusätzlich andere Tyrosinstellen und schafft so Andockstellen für verschiedene Proteine, die an der intrazellulären Signalübertragung beteiligt sind.

Integrins trigger focal adhesion signaling at the cell membrane. These membrane receptors sit in the phospholipid bilayer with hydrophobic regions that match the membrane interior. They provide attachment sites for outside signals such as hormones and growth factors. When these effectors bind and become active, integrins can start response pathways inside the cell.

Some integrin-driven pathways support cell growth and help cells survive. The signal begins at the membrane, but G-proteins and protein kinases carry it through the cell in a cascade. In platelets, thrombin binds to G-protein-coupled receptors and activates integrins. The activated integrins then bind RGD peptide sequences on blood proteins such as fibrinogen. As fibrinogen bridges form, focal adhesions cluster and the platelets aggregate.

Integrins can also pass signals directly into the cell by binding signaling proteins. In the Ras/MAP kinase pathway, focal adhesion kinase, or FAK, is the best-studied example of integrin signaling. Focal adhesions contain many tyrosine phosphorylation sites, so they include tyrosine-phosphorylated proteins such as FAK. Integrins cluster at cell-matrix junctions, and the beta subunit recruits FAK through adaptor proteins such as paxillin or talin bound to the alpha subunit.

FAK proteins cluster at the site and phosphorylate a specific tyrosine on each other. This creates a phospho-tyrosine docking site for Src proteins, which are cytoplasmic tyrosine kinases. FAK also phosphorylates other tyrosine sites. These new docking sites recruit more proteins that help carry out intracellular signaling.
