Zellen verbinden sich mit der EZM und anderen Zellen über verschiedene Adhäsionskomplexe, die zusammen als Verankerungsverbindungen bezeichnet werden.
In Epithelzellen verbinden fokale Adhäsionen und Hemidesmosomen Aktinfilamente und intermediäre Filamente mit einer spezialisierten Form der EZM, der Basallamina.
Hemidesmosomen enthalten eine dichte Plaque auf der zytosolischen Seite der Plasmamembran, die durch das Zytolinkerprotein Plektin gebildet wird.
Keratinhaltige Zwischenfilamente sind über Alpha-Sechs-Beta-Vier-Integrine und andere Proteine wie BP230 und BP180 mit dieser Plaque verbunden.
Auf der EZM-Fläche binden die Integrine Laminin, ein Glykoprotein, das ein Hauptbestandteil der Basallamina ist.
Die resultierenden Hemidesmosomen können EZM-Signale übertragen, die der Zelle helfen, auf mechanische Kräfte zu reagieren und Form, Polarität und Richtung der Migration zu ändern.

Anchoring Junctions

Ankerverbindungen sind Multiproteinkomplexe, die Zellen dabei helfen, sich mit anderen Zellen und der extrazellulären Matrix zu verbinden. Auf den Seiten- und Basalflächen der Zellen befinden sich Verankerungsverbindungen, die für starke und flexible Verbindungen sorgen. Fokale Adhäsionen entstehen häufig aufgrund von Zellinteraktionen mit den ECM-Substraten, die die Signaltransduktion über Kinasekaskaden und andere Mechanismen initiieren. Zusammen sorgen sie für Stabilität und Gewebeintegrität. Es gibt drei Arten von Verankerungsverbindungen: Desmosomen, Hemidesmosomen und Adhärenten.

Desmosomen erscheinen als dichte, symmetrische, scheibenförmige Plaques auf der Zellmembran. Adhäsionsproteine, sogenannte Cadherine, sind an der Plaquebildung beteiligt. Das charakteristische Erscheinungsbild ist auf Strukturproteine auf der inneren Oberfläche der Zellmembran zurückzuführen.

Hemidesmosomen sehen aus wie ein halbes Desmosom und verbinden Zellen mit der Basallamina. Obwohl sie den Desmosomen ähneln, enthalten sie anstelle von Cadherinen Adhäsionsproteine, die Integrine genannt werden. Diese Verbindungen sind für den Zusammenhalt der Zellen unerlässlich.

Adherens-Verbindungen verwenden entweder Cadherine oder Integrine, je nachdem, ob sie sich mit anderen Zellen oder der Matrix verbinden. Die Verbindungen sind durch das Vorhandensein des kontraktilen Proteins Aktin gekennzeichnet, das sich auf der zytoplasmatischen Oberfläche der Zellmembran befindet. Das Aktin kann einzelne Flecken verbinden oder eine gürtelartige Struktur innerhalb der Zelle bilden.

Die drei Verankerungsstellen beeinflussen die Form und Faltung des Epithelgewebes.

Anchoring junctions are multiprotein complexes that help cells connect to other cells and to the extracellular matrix. They sit on the lateral and basal surfaces of cells. These junctions create strong, flexible links that help maintain tissue stability and integrity.

Focal adhesions often form when cells interact with the extracellular matrix, or ECM, substrata. These contacts can start signal transduction through kinase cascades and other pathways. Together, these junctions help cells stay attached and support the structure of tissues.

There are three main types of anchoring junctions: desmosomes, hemidesmosomes, and adherens junctions. Desmosomes appear as dense, symmetrical, disc-shaped plaques on the cell membrane. Adhesion proteins called cadherins help form these plaques, and structural proteins on the inner surface of the membrane give them their distinct look.

Hemidesmosomes look like half a desmosome. They link cells to the basal lamina. Unlike desmosomes, hemidesmosomes use integrins instead of cadherins.

Adherens junctions can use cadherins or integrins, depending on whether they connect cells to other cells or to the matrix. These junctions are marked by actin, a contractile protein on the cytoplasmic side of the cell membrane. Actin may form isolated patches or a belt-like structure inside the cell.

Together, these three anchoring junctions help shape epithelial tissue and influence how it folds. They are essential for holding cells together and preserving tissue form.
