Zell-Matrix-Verbindungen, wie z. B. fokale Adhäsionen, sind direkt mit einem Netzwerk kontraktiler Proteine wie Aktin und Myosin verbunden.
Wenn mechanische Kräfte die Verbindungen von einer starren extrazellulären Matrix wegziehen, bildet die Zelle zusätzliche fokale Adhäsionen, um der Spannung standzuhalten.
Eine weichere EZM erzeugt weniger Widerstand, so dass die Zelle entsprechend der Steifigkeit der EZM in verschiedenen Geweben reagieren kann.
Eine solche Mechanosensorik beruht auf Übergangsproteinen wie Talin und Fibronektin, die ihre Konformation ändern, wenn die Verbindung gedehnt wird.
Wenn Aktinfilamente von Myosin gezogen werden, entfaltet sich Talin und legt Vinculin-Bindungsstellen frei. Vinculin rekrutiert zusätzliche Aktinfilamente, um die Verbindung zu stärken.
Auf der EZM-Fläche legt die Spannung, die während des Aufbaus der Fibrillenmatrix über die gesamte Länge des Fibronektins ausgeübt wird, verborgene Bindungsstellen für Kollagen frei.
Die Kollagenbindung stärkt die Verbindung und ermöglicht es den Zellen, auf mechanische Kräfte aus ihrer Umgebung zu reagieren.

Cell-matrix's Response to Mechanical Forces

In tierischen Zellen sorgt die extrazelluläre Matrix dafür, dass Zellen im Gewebe äußeren Belastungen standhalten und Signale von der Außenseite der Zelle nach innen übertragen. Die extrazelluläre Matrix ist umfangreich und ihre Zusammensetzung variiert zwischen verschiedenen Gewebetypen. Beispielsweise bilden die retikulären Fasern und die Grundsubstanz die ECM in lockerem Bindegewebe, während Kollagen und Knochenmineralien die ECM des Knochengewebes bilden.

Verankerungsverbindungen verbinden eine Zelle mechanisch mit der extrazellulären Matrix oder anderen Zellen. Eine Ansammlung von Verankerungsverbindungen bildet makromolekulare Komplexe, die als fokale Adhäsionen oder FAs bezeichnet werden. FAs regulieren die Zelladhäsion und Mechanotransduktion, also die Umwandlung mechanischer Signale in chemische Signale in der Zelle. FAs treten häufig auf den Seiten- und Basalflächen von Zellen auf und sorgen für starke und flexible Verbindungen. Sie wurden erstmals mithilfe eines Antitumor-Serums in mit dem Rous-Sarkom-Virus transformierten Nierenzellen von Ratten erkannt. Drei Proteine, Talin, Vinculin und Tensin, und ihre Isoformen verbinden Integrine mit dem Zytoskelett und stärken so die Verbindung. Spannungsabhängige Konformationsänderungen aktivieren Talin und Vinculin, was darauf hindeutet, dass sie an der mechanischen Wahrnehmung von Signalen aus ihrer Umgebung beteiligt sind.

Focal adhesions help animal cells connect to the extracellular matrix and sense mechanical forces. The extracellular matrix, or ECM, sits outside cells and helps tissues withstand stress. It also sends signals from outside the cell to the inside.

The ECM is large and its makeup changes from one tissue to another. In loose connective tissue, reticular fibers and ground substance form the ECM. In bone tissue, collagen and bone minerals make up the ECM.

Anchoring junctions hold a cell to the extracellular matrix or to other cells. When many of these junctions cluster together, they form focal adhesions, or FAs. These structures support cell adhesion and mechanotransduction, which is the conversion of mechanical signals into chemical signals inside the cell.

Focal adhesions are often found on the lateral and basal surfaces of cells. This location gives cells strong but flexible connections. They were first recognized with an antitumor serum in Rous sarcoma virus-transformed rat kidney cells.

Several proteins connect integrins to the cytoskeleton and strengthen the junction. These proteins include talin, vinculin, and tensin, along with their isoforms. Tension can change the shape of talin and vinculin, which suggests that they help cells sense forces from their environment.
