Die Potentiometrie misst die Potentialdifferenz zwischen zwei Elektroden, ohne einen signifikanten Strom zu ziehen, der die Zusammensetzung der Lösung durch die elektrochemische Zelle verändern würde.

Bei dieser Methode werden eine Messelektrode, die Elektronen mit der Analytlösung austauscht, und eine Bezugselektrode mit konstantem Potential verwendet. Jede Elektrode wird in eine Lösung eingetaucht, die aus zwei Halbzellen besteht. In einem herkömmlichen Aufbau dient die Bezugselektrode als Anode (linke Elektrode), der Ort der Oxidation, an dem Elektronen verloren gehen, während die Messelektrode als Kathode (rechte Elektrode) fungiert, an der eine Reduktion stattfindet und Elektronen gewonnen werden. Potentiometry measures the potential difference between two electrodes in an electrochemical cell. It does this without drawing significant current, so the solution’s composition is not altered. The method uses an indicator electrode, which exchanges electrons with the analyte solution, and a reference electrode, which keeps a constant potential.

Each electrode is placed in a solution that makes up a half-cell. In a common setup, the reference electrode is the anode, or left-hand electrode, where oxidation occurs and electrons are lost. The indicator electrode is the cathode, or right-hand electrode, where reduction occurs and electrons are gained.

The two half-cells are linked by a salt bridge. This bridge usually contains an inert electrolyte. It helps prevent short-circuiting and keeps the two solutions from mixing.

The cell potential is found from the difference between the reduction potentials at the cathode and the anode. Reduction potential describes a substance’s tendency to gain electrons. In cells that use salt bridges, liquid junction potentials at the boundaries between different electrolyte solutions must also be considered.

These junction effects can be reduced by balancing ion mobilities and using high salt concentrations. The Nernst equation can estimate analyte concentration from the measured potential. For more precise results, temperature dependency and matrix effects must also be taken into account. Potentiometry is therefore useful for analyzing solutions and identifying their chemical composition.
