Biofilms

Ein Biofilm ist ein mikrobielles Konsortium, das in eine selbst hergestellte Polysaccharidmatrix eingebettet ist, die an einer Oberfläche befestigt ist. Die Biofilmbildung erfolgt in vier Stadien – Anheftung, Besiedlung, Entwicklung und Ausbreitung. Während der Anheftung haften planktonische Zellen an einer Oberfläche. Bei der Besiedlung produzieren Zellen extrazelluläre Polysaccharide und bilden eine klebrige Matrix, die sie zu einem Biofilm zusammenbindet. Während der Reifung entwickelt sich der Biofilm zu mehrschichtigen Strukturen, die Mikroorganismen vor Umwelteinflüssen schützen. Gelegentlich können sich einige Zellen ablösen und zerstreuen, um neue Oberflächen zu besiedeln. Während die Dispergierung die Expansion des Biofilms ermöglicht, widersteht die verbleibende Struktur weiterhin externen Einflüssen, einschließlich Antibiotika, indem sie die Wirkstoffpenetration begrenzt und die Stoffwechselaktivität innerhalb der Matrix reduziert. Dies ermöglicht es Biofilmen, Immunreaktionen zu umgehen und bei Krankheiten wie Mukoviszidose zu überleben, wo sie die Atemwege verstopfen. Weitere schädliche Wirkungen sind Zahnbelag, der zu Karies führt, und Biofilme auf Medizinprodukten wie Kathetern, die Infektionen verursachen. Biofilme können auch ökologische Vorteile bieten, indem sie organische Substanz für den Nährstoffkreislauf abbauen und die Abwasserbehandlung durch Schadstoffabbau unterstützen.

Die Bildung von Biofilmen erfolgt in vier entscheidenden Phasen: Anheftung, Kolonisation, Entwicklung und Ausbreitung.
Zu Beginn der Anheftung steht die Adh&#228;sion frei schwimmender planktonischer Zellen an einer Oberfl&#228;che. Dies wird oft durch zellul&#228;re Kommunikationsmechanismen wie Quorum Sensing unterst&#252;tzt. Durch die Sekretion von Signalstoffen erm&#246;glicht Quorum Sensing den Bakterien, ihr Verhalten zu koordinieren, was zu Zellaggregation f&#252;hren kann. Es folgt die Kolonisation, bei der Mikroorganismen extrazellul&#228;re Polysaccharide, die eine klebrige Matrix bilden, produzieren. Diese Matrix bindet die Zellen zusammen und etabliert den Biofilm.
In der Entwicklungsphase baut sich der Biofilm zu einer komplexen, mehrschichtigen Struktur auf, die Wasserkan&#228;le f&#252;r den Transport von N&#228;hrstoffen und Abf&#228;llen enth&#228;lt. Diese strukturellen Anpassungen sch&#252;tzen die Konsortien vor &#228;u&#223;eren Stressfaktoren wie dem Eindringen von Antibiotika oder Immunantworten. W&#228;hrend der Ausbreitungsphase k&#246;nnen Zellen sich gelegentlich aus dem Biofilm l&#246;sen, und so die Kolonisation neuer Oberfl&#228;chen und die Expansion des Biofilms erm&#246;glichen.
Biofilmen kommen sowohl &#246;kologische als auch pathogene Funktionen zu. Indem sie organische Substanzen abbauen, tragen sie zum N&#228;hrstoffkreislauf bei und sind durch Schadstoffabbau f&#252;r die Abwasserbehandlung von Bedeutung. Ihre Pathogenit&#228;t stellt jedoch erhebliche Gesundheitsrisiken dar. Zum Beispiel k&#246;nnen Biofilme die Atemwege bei Patienten mit Mukoviszidose (haupts&#228;chlich verursacht durch Pseudomonas aeruginosa) blockieren oder Zahnbel&#228;ge bilden (haupts&#228;chlich gebildet von Streptococcus mutans), die Karies verursachen. Auf medizinischen Ger&#228;ten wie Kathetern l&#246;sen Biofilme durch ihre Resistenz gegen Antibiotika, persistierende Infektionen aus.
Die Pr&#228;vention der Biofilmbildung bleibt eine Herausforderung. G&#228;ngige Strategien umfassen den Einsatz von Lactoferrin, das die bakterielle Aggregation durch Eisenbindung hemmt, sowie die Entwicklung antimikrobieller Oberfl&#228;chen, um die Adh&#228;sion des Biofilms zu verringern. Um wirksamere Interventionen zu entwickeln, setzt die Forschung ihre Untersuchung der molekularen Mechanismen des Quorum Sensing und seiner Rolle bei der Resistenz von Biofilmen fort. Das Verst&#228;ndnis der Dynamik der Biofilmbildung ist entscheidend f&#252;r den Umgang mit ihren Auswirkungen in industriellen und medizinischen Bereichen.

Biofilms are communities of microorganisms that attach to a surface and build a self-made sticky matrix around themselves. This matrix is made of extracellular polysaccharides, which are sugar-based substances secreted by the cells. Biofilms may contain one species or several species, and their organized layers help the microbes survive better together.

Biofilm formation happens in four main stages: attachment, colonization, development, and dispersal. In the first stage, free-swimming planktonic cells stick to a surface. Cell-to-cell communication called quorum sensing often helps this process by using signaling molecules to coordinate bacterial behavior and promote cell aggregation.

During colonization, the microorganisms release extracellular polysaccharides that create the matrix holding the cells in place. The biofilm then matures into a multilayered structure with water channels. These channels move nutrients in and waste out, while the overall structure helps protect the community from environmental stress, including antibiotics and immune responses.

Biofilms also have a dispersal stage. At this point, some cells leave the biofilm and move to new surfaces, which allows the community to spread. This ability, along with its protective matrix, makes the biofilm a resilient form of microbial growth.

Biofilms can be helpful in nature and in industry. They help break down organic matter and support nutrient cycling. They are also important in wastewater treatment because they can help degrade pollutants. At the same time, biofilms can cause problems in the body. They can block airways in cystic fibrosis patients, often because of Pseudomonas aeruginosa, and dental plaque is a biofilm mainly formed by Streptococcus mutans that leads to tooth decay.

Biofilms are also a major concern on medical devices such as catheters, where they can cause long-lasting infections that are hard to treat with antibiotics. Preventing them is difficult. Strategies include lactoferrin, which blocks bacterial aggregation by binding iron, and antimicrobial surfaces that reduce attachment. Scientists continue to study quorum sensing and biofilm resistance to find better ways to stop them.
