<h4>Descripción general</h4> La diferencia relativa de carga eléctrica, o voltaje, entre el interior y el exterior de una membrana celular se denomina potencial de membrana. Se generan por diferencias en la permeabilidad de la membrana a varios iones y las concentraciones de estos iones a través de la membrana. <h4>El interior de una neurona es más negativo</h4> El potencial de membrana de una célula se puede medir insertando un microelectrodo en una célula y comparando la carga con un electrodo de referencia en el líquido extracelular. El potencial de membrana de una neurona en reposo,es decir, una neurona que actualmente no recibe ni envía mensajes, es negativo, normalmente alrededor de -70 milivoltios (mV). Esto se llama potencial de membrana en reposo. El valor negativo indica que el interior de la membrana es relativamente más negativo que el exterior, está polarizado. El potencial de reposo resulta de dos factores principales: la permeabilidad selectiva de la membrana y las diferencias en la concentración de iones dentro de la célula en comparación con el exterior. <h4>Permeabilidad de la membrana</h4> Las membranas celulares son selectivamente permeables porque la mayoría de los iones y moléculas no pueden cruzar la bicapa lipídica sin ayuda, a menudo de proteínas de canales iónicos que atraviesan la membrana. Esto se debe a que los iones cargados no pueden difundirse a través del interior hidrofóbico no cargado de las membranas. Los iones intra y extracelulares más comunes que se encuentran en el tejido nervioso son el potasio (K+), el sodio (Na+), el cloruro (Cl-) y el calcio (Ca2+). Cuando una neurona está en reposo, los canales de potasio (K+) son el principal tipo de canal iónico que está abierto, lo que permite que el K+ migre a través de la membrana. Esta permeabilidad, junto con las grandes concentraciones intracelulares, hacen que el potencial de membrana en reposo de la neurona esté determinado principalmente por el movimiento de K+. <h4>Las bombas crean gradientes de concentración</h4> Las diferencias en la concentración de iones entre el interior y el exterior de las neuronas se deben principalmente a la actividad de la bomba de sodio-potasio (Na+/K+), una proteína,transmembrana que bombea continuamente tres iones Na+ fuera de la célula por cada dos iones K+ que bombea hacia adentro. Esto establece gradientes de concentración, con una mayor concentración de iones Na+ en el exterior de las neuronas y una mayor concentración de iones K+ en el interior. Dado que la membrana es principalmente permeable al K+ en reposo,debido a los canales de K+ abiertos, el K+ puede difundirse a favor de su gradiente de concentración hasta la región de menor concentración, fuera de la célula. Estas cargas positivas que salen de la célula, combinadas con el hecho de que hay muchas proteínas cargadas negativamente dentro de la célula, hacen que el interior sea relativamente más negativo. Con el tiempo, la difusión de K+ hacia el exterior se equilibra mediante la repulsión electrostática de las cargas positivas que se acumulan fuera de la célula y se alcanza el equilibrio electroquímico. El potencial de reposo es muy importante en el sistema nervioso porque los cambios en el potencial de membrana, como el potencial de acción, son la base de la señalización neuronal. <h4>Cuidado con el pez globo</h4> El pez globo no suele encontrarse en muchos menús de mariscos fuera de Japón, en parte porque contiene una potente neurotoxina. La tetrodotoxina (TTX) es un bloqueador de los canales de sodio dependiente de voltaje muy selectivo que es letal en dosis mínimas. La dosis letal media (LD50) para ratones es de 334 μg/kg, en comparación con 8,5 mg/kg para el cianuro de potasio. También ha servido como herramienta esencial en la investigación en neurociencia. The Resting Membrane Potential

Neurons keep a negative resting charge across their membrane. This electrical difference, called the membrane potential, comes from how ions move and how the membrane lets them pass. In a resting neuron, the inside is typically around -70 millivolts and is more negative than the outside, so the cell is polarized.

Two main factors create this resting state. First, the membrane is selectively permeable, which means most ions and molecules cannot cross the lipid bilayer without help from ion channels. Second, ion concentrations are different inside and outside the cell. In nervous tissue, common ions include potassium (K+), sodium (Na+), chloride (Cl-), and calcium (Ca2+).

At rest, potassium channels are the main channels that are open. This makes potassium the ion that most strongly shapes the resting membrane potential. Potassium can move across the membrane more easily than many other ions, and its movement helps set the neuron’s negative charge.

The sodium-potassium pump creates the ion gradients that support this process. This transmembrane protein pumps three Na+ ions out of the cell for every two K+ ions it pumps in. As a result, Na+ builds up outside the neuron, and K+ builds up inside.

Because the membrane is mostly permeable to K+ at rest, potassium diffuses out of the cell down its concentration gradient. As positive charges leave, the inside becomes more negative. Negatively charged proteins inside the cell also contribute to this effect. Eventually, the outward movement of K+ is balanced by the electrical push from the positive charge outside the cell, and electrochemical equilibrium is reached.

This resting potential is important because changes in membrane potential are the basis of neural signaling. One example is the action potential, which depends on these voltage changes. The pufferfish toxin tetrodotoxin (TTX) blocks voltage-gated sodium channels and stops Na+ from entering when the channel opens. It disrupts action potentials, but it does not change the resting membrane potential, so it can be used to silence neuronal activity.
