Las células interactúan con una red de moléculas secretadas localmente que rodean la membrana plasmática, llamada matriz extracelular o MEC.

Cada complejo de adhesión comprende receptores transmembrana y proteínas adaptadoras citosólicas que unen los filamentos del citoesqueleto con ligandos en la MEC.
La integrina, el receptor de adhesión más prominente, y su adaptador, la talina, pueden unirse a cualquiera de las tres clases principales de moléculas de MEC: proteoglicanos, que amortiguan la célula; colágeno, que proporciona resistencia mecánica; y proteínas solubles multiadhesivas como la fibronectina que entrecruzan los receptores de adhesión con otros componentes de la matriz.

Overview of Cell-Matrix Interactions

La matriz extracelular o ECM mantiene unidas las células para formar un tejido y permite que las células dentro del tejido se comuniquen. La MEC está compuesta por proteínas como fibronectina, colágeno, laminina, etc. La proteína más abundante en este espacio es el colágeno. Las fibras de colágeno están entrelazadas con moléculas de proteínas que contienen carbohidratos llamadas proteoglicanos. La ECM permite la migración celular y proporciona un andamio estructural en la adhesión celular que ancla la célula cuando las proteínas de la matriz extracelular interactúan con los receptores de proteínas transmembrana.

Un ligando de ECM se une al receptor y cambia su conformación. El receptor, a su vez, cambia la conformación de los microfilamentos situados justo debajo de la membrana plasmática. Estos cambios conformacionales inducen señales químicas dentro de la célula que llegan al núcleo y “activan” o “desactivan” la transcripción de secciones específicas de ADN, lo que afecta la producción de proteínas asociadas y cambia así las actividades celulares.

Las integrinas son los receptores transmembrana más destacados que median en las uniones entre la célula y la matriz, como las adherencias focales y los hemidesmosomas. The extracellular matrix, or ECM, is the material that helps hold cells together in a tissue. It also lets cells in that tissue communicate with one another. The ECM contains proteins such as fibronectin, collagen, and laminin. Collagen is the most abundant protein in this space.

Collagen fibers are woven together with proteoglycans, which are protein molecules with carbohydrates attached. This mix gives the ECM its structure. It also supports cell migration and provides a scaffold for cell adhesion. At these adhesion sites, ECM proteins connect with transmembrane protein receptors and help anchor the cell.

When an ECM ligand binds to a receptor, the receptor changes shape. This change also affects the microfilaments just under the plasma membrane. The shape changes trigger chemical signals inside the cell that travel to the nucleus. There, they can turn specific DNA sections on or off and change the proteins the cell makes.

These signals can alter cellular activities. Integrins are the main transmembrane receptors that mediate cell-matrix junctions such as focal adhesions and hemidesmosomes. On the inside of the cell, integrins bind to actin and intermediate filaments. On the ECM side, they bind to fibronectin and collagen.
