Genomic DNA in Eukaryotes

Chez les eucaryotes, l'ADN à double brinest organisé spécialement au sein d'un noyau membranairepour s'adapter à l'espace limité de la cellule. 
Au premier niveau de compaction,l'ADN est étroitement enroulé autourde protéines particulières, les histones. 
L'histone et l'ADN qui y est associéforme le nucléosome,l'unité de base du paquet d'ADN. 
Les nucléosomes sont réunis par de l'ADN de liaisonpour former ce qui ressemble à des perles sur un fil. 
Au niveau suivant de l'empaquetage,les nucléosomes et l'ADN de liaison se torsadentpour former des fibres de chromatine. 
Enfin, des protéines fibreuses supplémentaires compactentencore plus la chromatine, permettant ces longs tronçonsd'ADN de former ces unités étroitement condensées,qu'on identifie comme chromosomes,selon la phase de division cellulaire.

Les eucaryotes ont de grands génomes par rapport aux procaryotes. Pour insérer leurs génomes dans une cellule, l’ADN eucaryote est empaqueté de manière extraordinairement compacte à l’intérieur du noyau. Pour ce faire, l’ADN est étroitement enroulé autour de protéines nommées histones, qui sont empaquetées en nucléosomes reliés par l’ADN lieur et s’enroulent en fibres de chromatine. D’autres protéines fibreuses compactent davantage la chromatine, qui est reconnaissable en tant que chromosomes au cours de certaines phases de la division cellulaire.
 
 <h4>Le génome humain mesuré en mètres</h4>
 
 La plupart des cellules du corps humain contiennent environ 6 milliards de paires de bases d’ADN compactées en 23 paires de chromosomes. Il est difficile d’imaginer exactement la quantité d’ADN que ces nombres représentent, et il est donc difficile de comprendre la densité nécessaire pour que l’ADN puisse s’insérer dans une cellule. Nous pouvons avoir un aperçu en exprimant le génome en termes de longueur. Si l’on disposait l’ADN d’une seule cellule diploïde en ligne droite, il ferait environ deux mètres de long ! 
 
 Notez que les humains n’ont pas de génomes anormalement grands. Beaucoup de poissons, d’amphibiens et de plantes à fleurs ont des génomes beaucoup plus grands que les humains. Par exemple, le génome haploïde de la plante à fleurs japonaise Paris japonica contient environ 50 fois plus d’ADN que le génome haploïde humain. Ces chiffres soulignent le travail étonnant que les histones et d’autres protéines de remodelage de la chromatine doivent faire pour empaqueter l’ADN.

Eukaryotic DNA fits inside the nucleus by packing into a highly compact form. Eukaryotes have large genomes compared with prokaryotes, so their DNA must be tightly organized to fit inside one cell.

The process starts when DNA wraps around proteins called histones. These DNA-histone units form nucleosomes, which are connected by linker DNA. The nucleosomes then coil into chromatin fibers, and additional fibrous proteins compact the chromatin even more.

This packed material is called chromatin, and it becomes recognizable as chromosomes during certain stages of cell division. In human cells, there are about 6 billion base pairs of DNA arranged into 23 pairs of chromosomes. If the DNA from one diploid cell were stretched out in a straight line, it would measure about two meters long.

Humans are not unusual in genome size. Many fish, amphibians, and flowering plants have much larger genomes than humans. The Japanese flowering plant Paris japonica has a haploid genome with about 50 times more DNA than the human haploid genome. These examples show how strongly histones and other chromatin remodeling proteins must work to package DNA.
