Lorsque les neurones présynaptiques déclenchentet stimulent à plusieurs reprises la cellule postsynaptique,cette action induit des modifications du type et du nombrede canaux ioniques de la membrane postsynaptique,tels qu'une classe de récepteurs du glutamateappelée N-méthyl-D-aspartate ou NMDA. 
Les récepteurs NMDA sont inactivés par les ions magnésium,cependant, avec une forte dépolarisationrésultant d'une stimulation répétée, les ions magnésiumsont déplacés, ce qui permet aux ions calcium d'entrer. 
Cet afflux de calcium initie une cascade de signalisationqui aboutit à une seconde classe de récepteurs du glutamate. 
L'acide alpha-amino-3-hydroxy-méthyl- 4-isoxazolyl-propionique,AMPA en abrégé, s'insérant alors dans la membrane. 
Dans ce cas, plus d'ions positifs pénètrent dans le neurone,ce qui renforce la réponse postsynaptiqueà la même stimulation présynaptique. 
La PLT est essentielle à l'apprentissage et constitueun moyen d'expliquer l'adage "la pratique rend parfait" carla réponse renforcée dure des minutes, des semaines,voire plus, si la stimulation présynaptique persiste.

potentialisation à long terme

La potentialisation à long terme, ou PLT, est l’un des moyens par lesquels la plasticité synaptique — les changements dans la force des synapses chimiques — peut se produire dans le cerveau. La PLT est le processus de renforcement synaptique qui se produit au fil du temps entre les connexions neuronales pré- et postsynaptiques. Le renforcement synaptique de la PLT fonctionne en opposition à l’affaiblissement synaptique de la dépression à long terme (DLT) et sont ensemble les principaux mécanismes qui sous-tendent l’apprentissage et la mémoire. 
 
 <h4>La PLT de Hebb </h4>
 
 La PLT peut se produire lorsque les neurones présynaptiques déclenchent et stimulent à plusieurs reprises le neurone postsynaptique. C’est ce qu’on appelle la PLT hebbienne puisqu’elle découle du postulat de Donald Hebb en 1949 selon lequel “ les neurones qui se déclenchent ensemble se raccordent ensemble ”. La stimulation répétée des neurones présynaptiques induit des changements dans le type et le nombre de canaux ioniques dans la membrane postsynaptique.
 
 Deux types de récepteurs postsynaptiques du neurotransmetteur excitateur glutamate sont impliqués dans la PLT : 1) les récepteurs N-méthyl-D-aspartate ou NMDA et 2) les récepteurs à l’acide α-amino-3-hydroxy-5-méthyl-4-isoxazolepropionique ou récepteurs AMPA. Bien que les récepteurs NMDA s’ouvrent lors de la liaison du glutamate, leur pore est habituellement bloqué par des ions magnésium qui empêchent d’autres ions chargés positivement d’entrer dans le neurone. Cependant, le glutamate libéré par les neurones présynaptiques peut se lier aux récepteurs AMPA postsynaptiques, provoquant un influx d’ions sodium qui entraîne la dépolarisation de la membrane. Lorsque la membrane postsynaptique est dépolarisée par de multiples entrées présynaptiques fréquentes, l’ion magnésium bloquant le pore du récepteur NMDA est déplacé, ce qui permet aux ions sodium et calcium d’affluer dans le neurone.
 
 L’influx accru d’ions calcium déclenche alors une cascade de signalisation qui aboutit à l'insertion d'un plus grand nombre de récepteurs AMPA dans la membrane plasmique. Alternativement, la cascade de signalisation peut phosphoryler les récepteurs de glutamate, leur permettant de rester ouvert pendant une plus longue durée et d'augmenter la conductivité des ions chargés positivement dans la cellule. En conséquence, la même stimulation présynaptique évoquera maintenant une réponse postsynaptique plus forte, étant donné que plus de récepteurs de glutamate seront activés et plus d’ions chargés positivement entreront dans le neurone postsynaptique. L’amplification qui se produit est connue sous le nom de renforcement synaptique ou potentialisation.
 
 L’adage “ c’est en forgeant qu’on devient forgeron ” peut être expliqué en partie par la PLT. Lorsqu’une nouvelle tâche est apprise, de nouveaux circuits neuronaux sont renforcés à l’aide de la PLT. Après chaque itération de l’exercice, la force synaptique dans les circuits neuronaux devient plus grande, et bientôt la tâche peut être exécutée correctement et efficacement. Les connexions nouvellement renforcées peuvent durer de quelques minutes à des semaines ou plus si la stimulation présynaptique persiste, ce qui signifie qu’à chaque fois ultérieure que la tâche est exécutée, la PLT est maintenue. 
 
 <h4>La PLT et la maladie</h4>
 
 Lorsque la PLT fonctionne normalement, nous pouvons apprendre et former des souvenirs avec facilité. Cependant, des anomalies dans la PLT ont été impliquées dans beaucoup de troubles neurologiques et cognitifs tels que la maladie d’Alzheimer, l’autisme, la dépendance, la schizophrénie et la sclérose en plaques. Une meilleure compréhension des mécanismes soutenant la PLT pourrait éventuellement mener à des thérapies.

Long-term Potentiation

Long-term potentiation, or LTP, is a form of synaptic strengthening in the brain. It changes the strength of chemical synapses over time between pre- and postsynaptic neurons. LTP works against long-term depression, or LTD, which weakens synapses. Together, LTP and LTD are major mechanisms behind learning and memory.

LTP can happen when a presynaptic neuron fires again and again and keeps stimulating a postsynaptic neuron. This is called Hebbian LTP, based on Donald Hebb’s idea that neurons that fire together wire together. Repeated stimulation changes the types and number of ion channels in the postsynaptic membrane. Two glutamate receptors are involved: NMDA receptors and AMPA receptors.

NMDA receptors open when glutamate binds, but magnesium ions usually block the pore. At the same time, glutamate can activate AMPA receptors on the postsynaptic cell. This causes sodium ions to enter and depolarize the membrane. When many presynaptic inputs arrive in quick succession, the magnesium block is removed from the NMDA receptor. Sodium and calcium ions then enter the neuron.

The calcium influx starts a signaling cascade inside the cell. This cascade leads to more AMPA receptors being inserted into the plasma membrane. It can also phosphorylate glutamate receptors, which helps them stay open longer and lets more positively charged ions enter the cell. The same presynaptic signal then creates a stronger postsynaptic response. This increase is called synaptic strengthening, or potentiation.

LTP also helps explain why practice makes perfect. When a new task is learned, new neural circuits are reinforced by LTP. With each round of practice, the synaptic strength in those circuits becomes stronger. Over time, the task can be performed more correctly and efficiently. These strengthened connections can last from minutes to weeks or longer if presynaptic stimulation continues, so later practice can maintain the LTP.

When LTP works normally, learning and memory are easier. Problems with LTP have been linked to Alzheimer’s disease, autism, addiction, schizophrenia, and multiple sclerosis. Understanding how LTP works may help guide future therapies for these neurological and cognitive disorders.
