Lorsqu’un halogénure d’alkyle réagit avec un nucléophile, le nucléophile peut déplacer l’halogène pour donner le produit de substitution ou il peut extraire un hydrogène voisin pour former un alcène par une réaction d’élimination.
Dans les réactions d’élimination, les nucléophiles fonctionnent comme des bases de Lewis en donnant une paire d’électrons à un proton. Certaines des bases courantes utilisées pour favoriser les réactions d’élimination comprennent les hydroxydes tels que l’hydroxyde de sodium, les alcoxydes comme le tert-butoxyde de potassium ou les alcools comme l’éthanol.
Les réactions d’élimination impliquent généralement la perte de petits fragments moléculaires d’un substrat pour former au moins une liaison π. Dans les halogénures d’alkyle, la réaction d’élimination se déroule par la perte d’un atome d’hydrogène et d’un atome d’halogène, d’où le nom de déshydrohalogénation.
Étant donné que le carbone lié au groupe partant est un carbone α et que l’hydrogène sur le carbone adjacent est un β hydrogène, ces réactions sont souvent appelées réactions de β-élimination ou de 1,2-élimination.
La plupart des réactions d’élimination se produisent via un mécanisme E2 ou E1.
Pour les réactions E2, des bases fortes comme l’éthoxyde de sodium sont utilisées. Le mécanisme concerté est initié par la déprotonation du carbone β suivie du départ du groupe de départ des halogénures conduisant à la formation d’une liaison π entre les positions α et β.
En revanche, la réaction E1 se déroule en deux étapes. La première implique le départ du groupe partant pour former un intermédiaire de carbocation, suivi d’une déprotonation du carbocation par la base pour former une liaison π.
Les halogénures d’alkyle contenant deux carbones β différents, les réactions d’élimination peuvent produire plus d’un alcène. Ici, l’alcène le plus substitué est le plus stable et s’appelle le produit de Zaitsev, tandis que l’alcène le moins substitué est appelé le produit de Hofmann. Ainsi, les réactions d’élimination sont dites régiosélectives.
De plus, les réactions d’élimination favorisent la formation de trans-alcènes par rapport aux isomères cis, ce qui les rend stéréosélectifs.

Un nucléophile peut réagir avec un halogénure d'alkyle pour donner le produit de substitution en déplaçant l'halogène. Ou bien il peut fonctionner comme une base pour donner le produit d'élimination par déprotonation du carbone voisin pour former un alcène. Dans une réaction d'élimination, le substrat perd deux groupes des carbones adjacents formant au moins une liaison π. Le carbone attaché à l’halogène est appelé carbone α, tandis que le carbone adjacent est appelé carbone β ; par conséquent, ces réactions sont appelées réactions d’élimination β ou 1,2-élimination.

Le nucléophile agit comme une base de Lewis en cédant une paire d'électrons à un proton. Les bases courantes utilisées pour favoriser les réactions d'élimination comprennent les hydroxydes (OH−), les alcoxydes (OR−) et les amides (NH2−). En présence d'une base forte, l'halogénure d'alkyle perd un proton du carbone β et l'halogène du carbone α, permettant la formation d'une liaison π entre les deux atomes de carbone.

<h4>Mécanisme des réactions d'élimination</h4>

Les réactions d'élimination se produisent généralement via les mécanismes E2 ou E1. Le mécanisme E2 se déroule en une seule étape concertée : l’abstraction de l’hydrogène β par la base s’accompagne du clivage de la liaison carbone α-halogène. Ainsi, la réaction E2 se déroule via un état de transition.

La réaction E1 se déroule en deux étapes. Tout d’abord, l’halogénure d’alkyle subit une ionisation formant un intermédiaire carbocation et un ion halogénure. Ensuite, la déprotonation du carbocation par la base aboutit à une liaison π. Ainsi, dans les réactions E1, l’intermédiaire carbocation est formé via un état de transition et un deuxième état de transition existe pour l’étape de déprotonation.

<h4>Régio- et stéréosélectivité</h4>

Lorsque l'halogénure d'alkyle a deux carbones β différents, la réaction d'élimination peut produire plus d'un alcène. Dans de tels cas, l'alcène le plus substitué (et le plus stable) est généralement observé, connu sous le nom de produit de Zaitsev. Elimination reactions of alkyl halides form alkenes by removing groups from adjacent carbons. A base can act on the substrate instead of causing substitution. It deprotonates the neighboring carbon, and the reaction creates at least one pi bond. The carbon bonded to the halogen is the alpha carbon, and the adjacent carbon is the beta carbon. These are also called beta elimination or 1,2-elimination reactions.

Strong bases help drive this process. Common examples include hydroxides, alkoxides, and amides. The base donates an electron pair to a proton, so it behaves as a Lewis base. In the presence of a strong base, the alkyl halide loses a proton from the beta carbon and the halogen from the alpha carbon. This lets a pi bond form between the two carbon atoms.

Elimination reactions often follow either the E2 or E1 mechanism. The E2 pathway is a single concerted step. The base removes the beta hydrogen at the same time that the alpha carbon-halogen bond breaks. This means E2 has one transition state.

The E1 pathway happens in two steps. First, the alkyl halide ionizes to form a carbocation intermediate and a halide ion. Then the base removes a proton from the carbocation, which forms the pi bond. In E1 reactions, the first step has one transition state, and the deprotonation step has a second transition state.

When an alkyl halide has two different beta carbons, more than one alkene can form. The more substituted and usually more stable alkene is called the Zaitsev product. In some cases, the less substituted Hofmann product forms instead, and the base helps decide which product is favored. Elimination reactions also tend to give trans-alkenes over cis isomers, so they are stereoselective.
