Le spectromètre de masse ionise les molécules et produit un spectre de masse à partir de la distribution des fragments.
Il remplit trois fonctions : l’ionisation de molécules ou d’atomes, la séparation des fragments d’ions et la détection des ions.
Dans l’ionisation électronique, une méthode d’ionisation courante, un échantillon vaporisé dans la chambre d’ionisation bombardé d’un faisceau d’électrons de haute énergie produit des cations radicaux. Le cation radical se fragmente rapidement en molécules neutres, radicaux et cations.
Les cations sont ensuite accélérés dans une chambre d’analyse par une série de plaques accélératrices chargées négativement.
De nombreux analyseurs courants appliquent un champ magnétique ou électrique à la chambre d’analyse, ce qui courbe les trajectoires des ions en fonction de leur masse et de leur charge.
Seuls les ions de valeurs m/z particulières ont la trajectoire exacte pour passer à travers la fente étroite placée devant le détecteur d’ions pour un champ magnétique ou électrique donné.
En modifiant le champ, l’analyseur de masse balaie une gamme de masses ioniques, leurs abondances étant enregistrées dans le spectre de masse.

Cette leçon détaille l'instrumentation d'un spectromètre de masse, un instrument physique permettant d'effectuer une spectrométrie de masse sur des molécules d'analytes et d'enregistrer les spectres de masse caractéristiques. Ceci est réalisé via trois fonctions principales:

</ul>

Dans la chambre d'ionisation du spectromètre de masse, l'analyte vaporisé sous vide est frappé par des électrons de haute énergie. L'énergie de l'électron est d'environ 70 eV, suffisante pour retirer un électron de l'analyte. L’ion moléculaire résultant se fragmente en espèces chargées et en molécules neutres. Le spectromètre de masse enregistre uniquement la masse des espèces chargées, car la charge permet le contrôle des molécules par un champ électrique ou magnétique.

L'ion moléculaire et ses fragments chargés sont accélérés par une série de plaques accélératrices chargées négativement positionnées de manière appropriée dans le détecteur de la chambre d'analyse. Un champ magnétique est appliqué sur le trajet entre la plaque accélératrice et le détecteur, ce qui provoque une courbe sur le trajet des espèces chargées. À champ magnétique constant, le rayon de courbure dépend de la masse moléculaire des espèces chargées. A mass spectrometer sorts charged ions by their mass-to-charge ratio and records their relative abundance. It turns vaporized analyte molecules into ions, separates them, and then measures the signal from each type of ion.

The process begins in the ionization chamber. The analyte is in the gas phase inside a vacuum, and it is struck by high-energy electrons. The electrons have about 70 eV of energy, which is enough to remove an electron from the analyte and form a molecular ion.

After ionization, the molecular ion can break into charged fragments and neutral molecules. The instrument records only the charged species. Their charge allows them to be controlled by electric or magnetic fields.

Next, the molecular ion and its charged fragments are accelerated by a set of negatively charged accelerator plates. These plates move the ions toward the detector in the analyzing chamber. A magnetic field bends the path of the ions as they travel between the accelerator plate and the detector.

At a constant magnetic field, the radius of the curved path depends on the molecular mass of the charged species. A slit placed in front of the detector helps admit only ions with a particular molecular weight. By changing the magnetic field, ions of different molecular weights can reach the detector one at a time.

As the instrument scans through the magnetic fields, it records the relative abundance of the charged species. The result is a mass spectrum. This spectrum shows the ions detected across the range of molecular weights.
