La chromatographie en phase gazeuse et la spectrométrie de masse combinent la chromatographie en phase gazeuse et la spectrométrie de masse dans un seul instrument pour analyser un mélange de composés.
Le chromatographe en phase gazeuse est utilisé pour séparer les composés, et le spectromètre de masse est utilisé pour analyser la masse de chaque composé.
La configuration du chromatographe se compose d’une colonne capillaire longue et étroite logée dans un four. La phase stationnaire est un matériau à base de silicone à point d’ébullition élevé avec une faible polarité et une viscosité élevée revêtu sur la surface intérieure de la colonne.
L'analyte est injecté à l'entrée de la colonne du chromatographe, où un gaz porteur inerte – la phase mobile – le transporte vers le spectromètre de masse.
Les composés de l’analyte se déplacent à travers la colonne à des vitesses différentes en raison de la différence de leur point d’ébullition et de leur affinité envers la phase stationnaire.
Ainsi, les composés atteignent l’autre extrémité de la colonne à des intervalles de temps différents, où un spectromètre de masse attaché analyse les signaux de masse de chaque composé séparément.

Gas chromatography-mass spectrometry (GC-MS) separates compounds in a mixture and then identifies them. A gas chromatograph first splits the mixture into its parts. A mass spectrometer then analyzes each part on its own to determine molecular masses and molecular structures.

The gas chromatograph uses a long, narrow capillary column. The inner wall of the column has a polysiloxane coating. This coating serves as the stationary phase. It has a high boiling point, low viscosity, and low polarity.

The analyte is injected into one end of the column. An inert gas carries it through the column as the mobile phase. Each compound moves at a different speed because of its boiling point and its interaction with the stationary phase. As a result, the compounds reach the end of the column at different times.

When each compound leaves the column, it enters the mass spectrometer. The mass spectrometer then analyzes that compound separately. This step reveals the molecular mass and gives information about molecular structure.
