Les cellules interagissent avec un réseau de molécules sécrétées localement autour de la membrane plasmique, appelé matrice extracellulaire ou ECM.
Ces interactions sont médiées par de grands complexes multimoléculaires appelés complexes d’adhésion cellule-matrice qui sont largement distribués ou regroupés en taches distinctes sur la membrane plasmique appelées jonctions cellule-matrice.
Chaque complexe d’adhésion comprend des récepteurs transmembranaires et des protéines adaptatrices cytosoliques qui lient les filaments du cytosquelette aux ligands de l’ECM.
L’intégrine, le récepteur d’adhésion le plus important, et son adaptateur, la taline, peuvent se lier à l’une des trois grandes classes de molécules ECM : les protéoglycanes, qui amortissent la cellule, le collagène, qui fournit une résistance mécanique, et les protéines solubles multiadhésives telles que la fibronectine qui réticulent les récepteurs d’adhésion avec d’autres composants de la matrice.
Les intégrines relient les filaments du cytosquelette avec une forme spécialisée de l'ECM au niveau de la lame basale qui tapisse la surface externe de la membrane cellulaire.
Le complexe d’adhésion qui en résulte interagit avec les molécules ECM et transmet des signaux mécaniques et chimiques pour réguler la croissance, la migration et la différenciation cellulaires.

Overview of Cell-Matrix Interactions

La matrice extracellulaire ou MEC maintient les cellules ensemble pour former un tissu et permet aux cellules du tissu de communiquer. La MEC comprend des protéines telles que la fibronectine, le collagène, la laminine, etc. La protéine la plus abondante dans cet espace est le collagène. Les fibres de collagène sont entrelacées avec des molécules protéiques contenant des glucides appelées protéoglycanes. La MEC permet la migration cellulaire et fournit un échafaudage structurel au niveau de l'adhésion cellulaire qui ancre la cellule lorsque les protéines de la matrice extracellulaire interagissent avec les récepteurs protéiques transmembranaires.

Un ligand de la MEC  se lie au récepteur et modifie sa conformation. Le récepteur modifie à son tour la conformation des microfilaments situés juste sous la membrane plasmique. Ces changements conformationnels induisent des signaux chimiques dans la cellule qui atteignent le noyau et activent ou désactivent la transcription de sections d'ADN spécifiques, affectant la production de protéines associées, modifiant ainsi les activités cellulaires.

Les intégrines sont les récepteurs transmembranaires les plus importants qui assurent la médiation des jonctions cellule-matrice comme les adhérences focales et les hémidesmosomes. Du côté cytosolique, les intégrines se lient à l'actine et aux filaments intermédiaires tandis qu'elles se lient à la fibronectine et au collagène dans la face de la MEC.

The extracellular matrix, or ECM, is the material that helps hold cells together in a tissue. It also lets cells in that tissue communicate with one another. The ECM contains proteins such as fibronectin, collagen, and laminin. Collagen is the most abundant protein in this space.

Collagen fibers are woven together with proteoglycans, which are protein molecules with carbohydrates attached. This mix gives the ECM its structure. It also supports cell migration and provides a scaffold for cell adhesion. At these adhesion sites, ECM proteins connect with transmembrane protein receptors and help anchor the cell.

When an ECM ligand binds to a receptor, the receptor changes shape. This change also affects the microfilaments just under the plasma membrane. The shape changes trigger chemical signals inside the cell that travel to the nucleus. There, they can turn specific DNA sections on or off and change the proteins the cell makes.

These signals can alter cellular activities. Integrins are the main transmembrane receptors that mediate cell-matrix junctions such as focal adhesions and hemidesmosomes. On the inside of the cell, integrins bind to actin and intermediate filaments. On the ECM side, they bind to fibronectin and collagen.
