Les intégrines sont une famille de récepteurs d’adhésion cellulaire qui se lient aux ligands de la matrice extracellulaire ou ECM pour médier les adhésions de la matrice cellulaire.
Ils passent d’un état actif à un état inactif. Sous la forme active, ils servent de liants mécaniques reliant le cytosquelette à la matrice extracellulaire.
L’intégrine comprend des sous-unités de glycoprotéines alpha et bêta associées de manière non covalente, formant un hétérodimère transmembranaire.
La queue C-terminale intracellulaire de la sous-unité bêta interagit avec les filaments d’actine dans le cytosol par l’intermédiaire de protéines adaptatrices telles que la taline et la paxilline. Les grands domaines extracellulaires N-terminaux de l’intégrine se lient à des motifs spécifiques de séquence d’acides aminés dans les protéines ECM, tels que la séquence RGD dans la fibronectine.
Le complexe d’adhésion formé transmet des signaux bidirectionnels à travers la membrane plasmique à l’aide de protéines effectrices, facilitant ainsi la motilité, la croissance et la survie des cellules.

Les cellules animales et protozoaires n’ont pas de parois cellulaires pour aider à conserver leur forme et assurer leur stabilité structurelle. Au lieu de cela, ces cellules eucaryotes sécrètent une masse collante de glucides et de protéines dans les espaces entre les cellules adjacentes. Ce réseau de protéines et de molécules est appelé matrice extracellulaire ou MEC.

Certaines protéines MEC s'assemblent dans une membrane basale à laquelle adhèrent les composants restants. Les protéoglycanes constituent généralement la majeure partie de la MEC, tandis que les protéines fibreuses, comme le collagène, lui confèrent sa force. Les protéoglycanes et le collagène sont attachés aux protéines de la fibronectine, qui, à leur tour, sont attachées à des protéines membranaires intégrales appelées intégrines.

Les intégrines de mammifères sont des récepteurs d'adhésion transmembranaire hétérodimères comprenant des sous-unités glycoprotéiques α et β. Dix-huit gènes différents de la chaîne α et huit gènes différents de la chaîne β se dimérisent dans diverses combinaisons, ce qui donne 24 types d'intégrines spécifiques aux vertébrés. Ils traversent la membrane et interagissent avec les protéines du cytosol et de la MEC. La sous-unité α contient un domaine de tête d'hélice β à sept pales tandis que la sous-unité β a un domaine de tête semblable à un  I ; ensemble, ils lient le ligand. La tête de liaison du ligand est reliée à deux structures en forme de pattes, une de chaque sous-unité. La sous-unité β interagit avec la protéine du cytosquelette et aide à transmettre des signaux entrant et sortant de la cellule.

Integrins help animal and protozoan cells attach to the extracellular matrix, or ECM. These cells do not have cell walls, so the ECM helps maintain shape and provide structural stability. The ECM is a sticky network of carbohydrates and proteins found between adjacent cells.

Some ECM proteins form a basement membrane, and other ECM parts adhere to it. Proteoglycans make up much of the ECM, while fibrous proteins such as collagen give it strength. Fibronectin connects proteoglycans and collagen to integral membrane proteins called integrins.

Integrins are heterodimeric transmembrane adhesion receptors in mammals. That means each receptor has two different glycoprotein subunits, an alpha subunit and a beta subunit. Eighteen alpha-chain genes and eight beta-chain genes can pair in different ways to make 24 vertebrate integrins.

These receptors span the cell membrane and link the ECM to the cytosol. The alpha subunit has a seven-bladed beta-propeller head domain, and the beta subunit has an I-like head domain. Together, these head domains bind ligand, and the two leg-like parts connect the head to the membrane.

The beta subunit also interacts with a cytoskeletal protein. This helps integrins transmit signals into and out of the cell. Their structure and connections make them important for adhesion and communication in cells that rely on the ECM.
