Pour qu'une cellule migre à travers l'ECM, l'attachement de la cellule à l'ECM doit être fait et brisé rapidement.
Ceci est facilité par les intégrines liées à la membrane qui passent rapidement d’un état actif, qui forme des attachements, à un état inactif qui ne le fait pas.
Dans la conformation inactive, les queues du dimère de l’intégrine sont verrouillées de manière compacte, empêchant l’intégrine de se lier aux protéines ECM ou aux filaments cytosquelettiques.
Les intégrines peuvent être activées de l’extérieur de la cellule dans le mécanisme « de l’extérieur vers l’intérieur » ou de l’intérieur dans le mécanisme « de l’intérieur vers l’extérieur ».
Dans le mécanisme « de l'extérieur vers l'intérieur », les protéines ECM comme la fibronectine se lient au domaine extracellulaire de l'intégrine. Les queues se décrochent et l’intégrine passe d’un état inactif à un état actif, exposant les sites de liaison des protéines adaptatrices cytosoliques.
À l'inverse, dans le mécanisme « de l'intérieur vers l'extérieur », les protéines adaptatrices comme la taline se lient à la chaîne bêta cytosolique de l'intégrine, brisant ainsi le lien alpha-bêta et forçant les jambes de l'intégrine. L’intégrine active peut maintenant se lier aux protéines ECM.

Activation of Integrins

Les intégrines se lient aux ligands et transmettent des informations de l'extérieur de la cellule vers l'intérieur ou vice versa via une  “signalisation de l'extérieur vers l'intérieur” ou une  “signalisation de l'intérieur vers l'extérieur.”

Dans “la signalisation extérieur-intérieur,” des facteurs externes dans l’espace extracellulaire se lient aux sites de liaison du ligand exposés sur les intégrines. Cela amène la protéine inactive à subir un changement de conformation pour devenir active. Les intégrines sont souvent regroupées sur la membrane cellulaire. Des événements de liaison de ligand répétitifs et régulièrement espacés fournissent un stimulus efficace. Lorsque la cellule rencontre divers signaux extracellulaires, les intégrines activées affectent les propriétés cellulaires telles que la polarité cellulaire, la structure du cytosquelette, l'adhésion, la migration, l'invasion, l'expression des gènes, la survie cellulaire et la prolifération.

“La signalisation de l'intérieur vers l'extérieur” dépend d'activateurs intracellulaires tels que des protéines adaptatrices du cytosquelette comme le talin ou le kindlins. Les talines contiennent des sites de liaison pour les domaines cytosoliques de l'intégrine β, un site de liaison à l'actine C-terminal hautement conservé et un site de liaison à la vinculine. Kindlin comprend des sites de liaison cytosoliques de queue d'intégrine β dans leurs domaines F3, des domaines de liaison membranaire et une extrémité C-terminale qui interagit avec les intégrines, les protéines adaptatrices d'actine ou la kinase liée à l'intégrine (ILK). Kindlin interagit avec le domaine cytoplasmique de l'intégrine β et stabilise l'état activé du complexe de l'intégrine. “La signalisation de l'intérieur vers l'extérieur” peut renforcer l'adhésion médiée par les intégrines avec des ligands extracellulaires, qui transfèrent la force nécessaire à la migration cellulaire, à l'invasion, à l'assemblage de la MEC et au remodelage.

Integrins help cells connect with their environment and send signals across the cell membrane. They can carry information from outside the cell to the inside, or from inside the cell to the outside. These two paths are called outside-in signaling and inside-out signaling.

In outside-in signaling, molecules in the extracellular space bind to exposed ligand-binding sites on integrins. A ligand is a molecule that binds to a protein. This binding makes the inactive integrin change shape and become active. Integrins are often grouped in clusters on the cell membrane, and repeated ligand binding can create a strong signal.

Once integrins are activated, they can affect many cell features. These include cell polarity, cytoskeletal structure, adhesion, migration, invasion, gene expression, cell survival, and proliferation. This is why integrin signaling matters for how cells respond to their surroundings.

Inside-out signaling depends on internal activators such as the cytoskeletal adaptor proteins talin and kindlin. Talin has binding sites for the beta integrin cytosolic domains, a conserved C-terminal actin-binding site, and a vinculin-binding site. Kindlin has beta integrin cytosolic tail binding sites in its F3 domains, membrane-binding domains, and a C-terminus that can interact with integrins, actin adaptor proteins, or integrin-linked kinase (ILK).

Kindlin also binds the beta integrin cytoplasmic domain and helps keep the integrin complex in its activated state. Inside-out signaling can strengthen integrin-mediated adhesion to extracellular ligands. That stronger attachment helps provide the force needed for cell migration, invasion, ECM assembly, and remodeling.
