Des grappes d’intégrines sur la membrane cellulaire peuvent temporairement relier les ligands de la matrice extracellulaire au cytosquelette pour former des complexes d’adhésion focale.
Les adhérences focales peuvent croître et rétrécir, ce qui permet à la cellule de ramper pendant la migration.
Alors que l’adhésion provient de la membrane cellulaire, les signaux sont transmis par les protéines kinases et les protéines G en cascade dans toute la cellule.
Par exemple, la thrombine, une molécule de signalisation extracellulaire dans les plaquettes, active le récepteur couplé aux protéines G, cAR1/cAR3, qui à son tour active Rap1 GTPase.
Active Rap1 interagit avec une molécule adaptatrice interagissant avec Rap1-GTP ou RIAM qui recrute de la taline inactive dans la membrane cellulaire.
Les domaines de la tête N-terminale et de la queue C-terminale du talin inactif sont repliés ensemble. La liaison d’un phosphoinositide dans la membrane cellulaire perturbe l’interaction tête-queue, aidant ainsi la taline à se déployer.
Avec la kindlin, la taline active l’intégrine et se lie à d’autres protéines adaptatrices telles que la vinculine, qui lient l’intégrine aux filaments d’actine.

Intracellular Signaling Affects Focal Adhesions

Les intégrines agissent à la fois comme récepteurs d'entrées extracellulaires et comme activateurs de traitement intracellulaire. Comme leur nom l’indique, les intégrines sont entièrement intégrées à la structure membranaire. Leurs régions hydrophobes couvrant la membrane interagissent avec la région hydrophobe de la bicouche phospholipidique. Ces récepteurs membranaires fournissent des sites de fixation extracellulaires pour des effecteurs comme les hormones et les facteurs de croissance. Ils activent des cascades de réponses intracellulaires lorsque leurs effecteurs sont liés et actifs.

Certaines voies de stimulation médiées par les intégrines induisent la prolifération cellulaire et favorisent la survie cellulaire. Les adhérences proviennent de la membrane cellulaire, mais les signaux sont transmis en cascade dans toute la cellule par les protéines G et les protéines kinases. Par exemple, la liaison de la thrombine aux récepteurs couplés aux protéines G déclenche une signalisation intracellulaire pour activer les intégrines dans les plaquettes. Les intégrines activées se lient aux séquences peptidiques RGD des protéines sanguines, telles que le fibrinogène. À mesure que les fibrinogènes commencent à former des ponts, les adhérences focales se regroupent et les plaquettes s’agrègent.

Les intégrines peuvent également se lier directement aux protéines de signalisation et relayer les signaux dans la cellule. Dans la voie Ras/MAP kinase, la kinase d'adhésion focale (FAK), une protéine kinase cytoplasmique, est le mode de signalisation des intégrines le mieux étudié. Des études suggèrent que les adhérences focales contiennent de nombreux sites de phosphorylation de la tyrosine et contiennent donc des protéines phosphorylées par la tyrosine telles que FAK. Les intégrines se regroupent aux jonctions cellule-matrice et la sous-unité β recrute FAK via des protéines adaptatrices telles que la paxilline ou la taline liées à la sous-unité α. Les protéines FAK se regroupent sur le site et phosphorylent un site tyrosine spécifique les unes sur les autres. Integrins trigger focal adhesion signaling at the cell membrane. These membrane receptors sit in the phospholipid bilayer with hydrophobic regions that match the membrane interior. They provide attachment sites for outside signals such as hormones and growth factors. When these effectors bind and become active, integrins can start response pathways inside the cell.

Some integrin-driven pathways support cell growth and help cells survive. The signal begins at the membrane, but G-proteins and protein kinases carry it through the cell in a cascade. In platelets, thrombin binds to G-protein-coupled receptors and activates integrins. The activated integrins then bind RGD peptide sequences on blood proteins such as fibrinogen. As fibrinogen bridges form, focal adhesions cluster and the platelets aggregate.

Integrins can also pass signals directly into the cell by binding signaling proteins. In the Ras/MAP kinase pathway, focal adhesion kinase, or FAK, is the best-studied example of integrin signaling. Focal adhesions contain many tyrosine phosphorylation sites, so they include tyrosine-phosphorylated proteins such as FAK. Integrins cluster at cell-matrix junctions, and the beta subunit recruits FAK through adaptor proteins such as paxillin or talin bound to the alpha subunit.

FAK proteins cluster at the site and phosphorylate a specific tyrosine on each other. This creates a phospho-tyrosine docking site for Src proteins, which are cytoplasmic tyrosine kinases. FAK also phosphorylates other tyrosine sites. These new docking sites recruit more proteins that help carry out intracellular signaling.
