Les cellules se connectent à l’ECM et à d’autres cellules via différents complexes d’adhésion, collectivement appelés jonctions d’ancrage.
Dans les cellules épithéliales, les adhérences focales et les hémidesmosomes relient les filaments d’actine et les filaments intermédiaires à une forme spécialisée de MEC appelée lame basale.
Les hémidesmosomes contiennent une plaque dense sur le côté cytosolique de la membrane plasmique formée par la protéine cytolinker plectine.
Les filaments intermédiaires contenant de la kératine sont connectés à cette plaque via les intégrines alpha, six, bêta-quatre, et d’autres protéines telles que BP230 et BP180.
Sur la face ECM, les intégrines se lient à la laminine, une glycoprotéine qui est un composant majeur de la lame basale.
Les hémidesmosomes qui en résultent peuvent transmettre des signaux ECM aidant la cellule à répondre aux forces mécaniques et à changer de forme, de polarité et de direction de migration.

Anchoring Junctions

Les jonctions d'ancrage sont des complexes multiprotéiques qui aident les cellules à se connecter à d'autres cellules et à la matrice extracellulaire. Des jonctions d'ancrage sont présentes sur les surfaces latérales et basales des cellules, fournissant des connexions solides et flexibles. Les adhérences focales se forment souvent en raison d'interactions cellulaires avec les substrats de la MEC, qui déclenchent la transduction du signal via des cascades de kinases et d'autres mécanismes. Ensemble, ils assurent la stabilité et l’intégrité des tissus. Il existe trois types de jonctions d’ancrage : les desmosomes, les hémidesmosomes et les adhérents.

Les desmosomes apparaissent sous forme de plaques denses et symétriques en forme de disque sur la membrane cellulaire. Les protéines d'adhésion appelées cadhérines sont impliquées dans la formation des plaques. L’apparence distinctive est due aux protéines structurelles présentes sur la surface interne de la membrane cellulaire.

Les hémidesmosomes ressemblent à un demi-desmosome et relient les cellules à la lame basale. Bien qu’ils soient d’apparence similaire aux desmosomes, ils comprennent des protéines d’adhésion appelées intégrines plutôt que cadhérines. Ces connexions sont essentielles au maintien des cellules.

Les jonctions adhérentes utilisent soit des cadhérines, soit des intégrines selon qu'elles se lient à d'autres cellules ou à la matrice. Les jonctions sont caractérisées par la présence de la protéine contractile d'actine située à la surface cytoplasmique de la membrane cellulaire. L'actine peut relier des zones isolées ou former une structure en forme de ceinture à l'intérieur de la cellule.

Les trois jonctions d'ancrage influencent la forme et le repliement du tissu épithélial.

Anchoring junctions are multiprotein complexes that help cells connect to other cells and to the extracellular matrix. They sit on the lateral and basal surfaces of cells. These junctions create strong, flexible links that help maintain tissue stability and integrity.

Focal adhesions often form when cells interact with the extracellular matrix, or ECM, substrata. These contacts can start signal transduction through kinase cascades and other pathways. Together, these junctions help cells stay attached and support the structure of tissues.

There are three main types of anchoring junctions: desmosomes, hemidesmosomes, and adherens junctions. Desmosomes appear as dense, symmetrical, disc-shaped plaques on the cell membrane. Adhesion proteins called cadherins help form these plaques, and structural proteins on the inner surface of the membrane give them their distinct look.

Hemidesmosomes look like half a desmosome. They link cells to the basal lamina. Unlike desmosomes, hemidesmosomes use integrins instead of cadherins.

Adherens junctions can use cadherins or integrins, depending on whether they connect cells to other cells or to the matrix. These junctions are marked by actin, a contractile protein on the cytoplasmic side of the cell membrane. Actin may form isolated patches or a belt-like structure inside the cell.

Together, these three anchoring junctions help shape epithelial tissue and influence how it folds. They are essential for holding cells together and preserving tissue form.
