Bacterial Cell Wall 

La paroi cellulaire bactérienne est une structure rigide entourant la membrane plasmique. Il maintient la forme de la cellule et la protège de la pression osmotique. Il est composé de peptidoglycane, un polymère de sucres alternatifs – N-acétylglucosamine et acide N-acétylmuramique – liés par des acides aminés, formant un maillage flexible. Les bactéries à Gram positif ont d’épaisses couches de peptidoglycane. Ils contiennent également des acides teichoïques, qui facilitent l’absorption des ions et protègent les cellules des substances nocives et des molécules de défense de l’hôte. Les bactéries à Gram négatif ont une couche de peptidoglycane plus mince et une membrane externe avec des lipopolysaccharides, qui ont une activité endotoxine qui déclenche la réponse immunitaire de l’hôte et assure l’intégrité structurelle. La coloration de Gram distingue les bactéries par la structure de la paroi cellulaire. Les bactéries à Gram positif apparaissent violettes car elles conservent le colorant violet cristallin, tandis que les bactéries à Gram négatif apparaissent roses en raison d’une contre-tache. La composition de la paroi cellulaire détermine également la sensibilité aux antibiotiques. Par exemple, les bactéries à Gram positif sont plus sensibles à la pénicilline que les bactéries à Gram négatif.

Des acides teicho&#239;ques y sont int&#233;gr&#233;s ; ils interviennent notamment dans le transport des cations et la d&#233;fense contre des substances d&#233;l&#233;t&#232;res, y compris certains antibiotiques et des mol&#233;cules du syst&#232;me immunitaire. Cet agencement multicouche retient le colorant violet de gentiane lors de la coloration de Gram, conf&#233;rant &#224; ces bact&#233;ries leur coloration violette caract&#233;ristique.
&#192; l&#8217;inverse, les bact&#233;ries &#224; Gram n&#233;gatif disposent d&#8217;une couche de peptidoglycane plus mince recouverte d&#8217;une membrane externe riche en lipopolysaccharides (LPS). Les diff&#233;rences structurales des parois bact&#233;riennes conditionnent leur interaction avec les antibiotiques. La p&#233;nicilline et les autres &#946;-lactamines ciblent la synth&#232;se du peptidoglycane, ce qui inhibe efficacement la formation de la paroi chez les bact&#233;ries &#224; Gram positif. Bacterial cell walls give bacteria their shape and help protect them from osmotic stress. This rigid but flexible layer surrounds the plasma membrane and supports cellular integrity. Its main building block is peptidoglycan, a mesh-like polymer that adds strength while still allowing some flexibility.

Peptidoglycan is made of alternating N-acetylglucosamine, or NAG, and N-acetylmuramic acid, or NAM. Short amino acid chains connect these sugar units and help form the strong network. The thickness and makeup of this layer differ in Gram-positive and Gram-negative bacteria, which affects physical properties and antibiotic sensitivity.

Gram-positive bacteria have several layers of peptidoglycan in a thick wall. Teichoic acids are embedded in this structure and help with cation transport and defense against harmful substances, including antibiotics and immune molecules. This thick wall holds the crystal violet dye during Gram staining, so Gram-positive bacteria appear purple.

Gram-negative bacteria have a thinner peptidoglycan layer and an outer membrane rich in lipopolysaccharides, or LPS. This outer membrane acts as a selective barrier and adds structural support. It also acts as an endotoxin during infections. In Gram staining, Gram-negative bacteria appear pink because they keep the counterstain instead of the primary dye.

These wall differences also affect antibiotics. Penicillin and related beta-lactam antibiotics block peptidoglycan synthesis, so they can stop cell wall formation in Gram-positive bacteria. Gram-negative bacteria are naturally resistant to some antibiotics like penicillin because of their outer membrane. However, beta-lactam antibiotics that can pass through this barrier may still work. Learning cell wall structure helps explain targeted antibiotics and bacterial resistance.
