Peptidoglycan Synthesis

Le peptidoglycane, un composant clé de la paroi cellulaire bactérienne, est composé d’une alternance d’unités de N-acétylglucosamine, ou NAG, et d’acide N-acétylmuramique ou NAM reliées par des liaisons glycosidiques β-1,4. La synthèse du peptidoglycane comporte trois phases. Dans le cytoplasme, les molécules d’uridine diphosphate (UDP) sont attachées de manière covalente au NAG. UDP-NAG est converti enzymatiquement en UDP-NAM. Ensuite, une chaîne pentapeptide est ajoutée à UDP-NAM pour former un UDP-NAM-pentapeptide. Dans la phase associée à la membrane, le NAM-pentapeptide est lié au phosphate de bactoprénol, formant le lipide I sur le côté cytoplasmique de la membrane plasmique. L’ajout d’une unité NAG au lipide I forme le lipide II, qui est transloqué à travers la membrane vers le périplasme par une flippase. Dans la phase périplasmique, les glycosyltransférases forment des liaisons glycosidiques pour ajouter le NAG-NAM-pentapeptide à la chaîne de peptidoglycane en croissance attachée au bactoprénol. Dans la dernière étape, l’enzyme transpeptidase forme des réticulations peptidiques entre les chaînes de glycanes, libère la chaîne du bactoprénol et termine la synthèse en liant les peptides souches.

Phase cytoplasmique &#8211; Synth&#232;se des pr&#233;curseurs
La biosynth&#232;se du peptidoglycane d&#233;bute dans le cytoplasme avec la formation de pr&#233;curseurs solubles. &#192; ce stade, l&#8217;uridine diphosphate (UDP), un ester de l&#8217;acide pyrophosphorique, se lie au NAG et au NAM pour former respectivement l&#8217;UDP-NAG et l&#8217;UDP-NAM. L&#8217;UDP-NAG est ensuite converti enzymatiquement en UDP-NAM. Une cha&#238;ne pentapeptidique&#8212;compos&#233;e typiquement de L-alanine, d&#8217;acide D-glutamique, d&#8217;acide m&#233;so-diaminopim&#233;lique (ou de L-lysine), et de deux r&#233;sidus de D-alanine&#8212;est ajout&#233;e de mani&#232;re s&#233;quentielle &#224; l&#8217;UDP-NAM, formant l&#8217;UDP-NAM-pentapeptide. Ce compos&#233; constitue le bloc de construction fondamental pour les &#233;tapes ult&#233;rieures de la synth&#232;se du peptidoglycane.
Phase membranaire &#8211; Transfert par transporteur lipidique
Au cours de la phase membranaire, l&#8217;UDP-NAM-pentapeptide est transf&#233;r&#233; au bactopr&#233;no&#173;lphosphate, un transporteur lipidique ancr&#233; dans la monocouche interne de la membrane plasmique. Ce transfert aboutit &#224; la formation du Lipide I. Ensuite, une unit&#233; NAG est ajout&#233;e au Lipide I, pour g&#233;n&#233;rer le Lipide II, qui contient d&#233;sormais la structure compl&#232;te du disaccharide-pentapeptide. Une enzyme flippase transloque ensuite le Lipide II &#224; travers la membrane, livrant le pr&#233;curseur du c&#244;t&#233; p&#233;riplasmique pour son incorporation dans la paroi en croissance.
Phase p&#233;riplasmique &#8211; Polym&#233;risation et r&#233;ticulation
Dans la phase finale, localis&#233;e dans le p&#233;riplasme, les glycosyltransf&#233;rases incorporent les unit&#233;s disaccharide-pentapeptide dans le treillis de peptidoglycane existant en catalysant la formation de liaisons glycosidiques. La derni&#232;re &#233;tape, appel&#233;e &#8220;transpeptidation&#8221;, est r&#233;alis&#233;e par des enzymes transpeptidases. Cette r&#233;action lib&#232;re le sous-unit&#233; du bactopr&#233;nol et forme des liaisons peptidiques entre les peptides de tige adjacents, renfor&#231;ant l&#8217;int&#233;grit&#233; structurale de la couche de peptidoglycane et compl&#233;tant ainsi le processus de biosynth&#232;se.

Peptidoglycan builds the bacterial cell wall and gives the cell its shape and strength. It is a mesh-like polymer around the bacterial plasma membrane. The polymer is made of repeating sugar units, N-acetylglucosamine (NAG) and N-acetylmuramic acid (NAM), linked by β-1,4 glycosidic bonds. Short peptide chains cross-link the sugar chains and make the wall more rigid.

Peptidoglycan biosynthesis starts in the cytoplasm with soluble precursors. Uridine diphosphate, or UDP, binds to NAG and NAM to form UDP-NAG and UDP-NAM. UDP-NAG is then converted into UDP-NAM by enzymes. A pentapeptide chain is added step by step to UDP-NAM. This chain usually includes L-alanine, D-glutamic acid, meso-diaminopimelic acid or L-lysine, and two D-alanine residues. The product, UDP-NAM-pentapeptide, becomes the core building block for the later stages.

Next, the pathway moves to the membrane-associated phase. UDP-NAM-pentapeptide is transferred to bactoprenol phosphate, a lipid carrier in the inner leaflet of the plasma membrane, and this forms Lipid I. A NAG unit is then added to make Lipid II. Lipid II contains the complete disaccharide-pentapeptide structure. A flippase enzyme then moves Lipid II across the membrane to the periplasmic side, where the cell wall grows.

The final stage happens in the periplasm. Glycosyltransferases add the disaccharide-pentapeptide units to the growing peptidoglycan mesh by forming glycosidic bonds. The last step is transpeptidation, which is carried out by transpeptidase enzymes. This reaction releases the subunit from bactoprenol and creates peptide cross-links between neighboring stem peptides. These links strengthen the peptidoglycan layer and complete the synthesis process.
